During vascular development, procambial and cambial cells give rise to xylem and phloem cells. Because the vascular tissue is deeply embedded, it has been difficult to analyze the processes of vascular development in detail. Here, we establish a novel in vitro experimental system in which vascular development is induced in Arabidopsis thaliana leaf-disk cultures using bikinin, an inhibitor of glycogen synthase kinase 3 proteins. Transcriptome analysis reveals that mesophyll cells in leaf disks synchronously turn into procambial cells and then differentiate into tracheary elements. Leaf-disk cultures from plants expressing the procambial cell markers TDRpro：GUs and TDRpro：YFP can be used for spatiotemporal visualization of procambial cell formation. Further analysis with the tdr mutant and TDIF （tracheary element differentiation inhibitory factor） indicates that the key signaling TDIF-TDR-GsK3s regulates xylem differentiation in leaf-disk cultures. This new culture system can be combined with analysis using the rich material resources for Arabidopsis including cell-marker lines and mutants, thus offering a powerful tool for analyzing xylem cell differentiation.

We report on the design and characterization of the plasma mirror system installed on the J-KAREN-P laser at the Kansai Photon science Institute,National Institutes for Quantum science and Technology.The reflectivity of the single plasma mirror system exceeded 80%.In addition,the temporal contrast was improved by two orders of magnitude at 1 ps before the main pulse.Furthermore,the laser near-field spatial distribution after the plasma mirror was kept constant at plasma mirror fluence of less than 100 kJ/cm^(2).We also present the results of investigating the difference and the fluctuation in energy,pulse width and pointing stability with and without the plasma mirror system.

Engineered extracellular matrices(ECMs)that replicate complex in-vivo features have shown great potential in tissue engineering.Biocompatible hydrogel microstructures have been widely used to replace these native ECMs for physiologically relevant research.However,accurate reproduction of the 3D hierarchical and nonuniform mechanical stffness inside one integrated microstructure to mimic the complex mechanical properties of native ECMs presents a major challenge.Here,by using digital holographic microscopy(DHM)-based stffness imaging feedback,we propose a novel closed-loop control algorithm to achieve high-accuracy control of mechanical properties for hydrogel microstructures that recapitulate the physiological properties of native ECMs with high fidelity.During photoprinting,the photocuring area of the hydrogel is divided into microscale grid areas to locally control the photocuring process.With the assistance of a motorized microfluidic channel,the curing thickness is controlled with layer-by-layer stacking.The DHM-based stiffness imaging feedback allows accurate adjustment of the photocuring degree in every grid area to change the crosslinking network density of the hydrogel,thus enabling large-span and high-resolution modulation of mechanical properties.Finally,the gelatin methacrylate was used as a typical biomaterial to construct the highfidelity biomimetic ECMs.The Young's modulus could be flexibly modulated in the 10 kPa to 50 kPa range.Additionally,the modulus gradient was accurately controlled to within 2.9 kPa.By engineering ECM with locally different mechanical properties,cell spreading along the stff areas was observed successfully.We believe that this method can regenerate complex biomimetic ECMs that closely recapitulate in-vivo mechanical properties for further applications in tissue engineering and biomedical research.

In the spatial design fields such as architectural design and urban design, a consensus-building process among a variety of stakeholders like project executors, architects, residents, users, and general citizens is required. New technological developments such as cloud computing and Virtual Design studios （VDs） enable the creation of virtual meeting systems. This paper proposes an approach towards a synchronous distributed design meeting system. In this paper, in addition to sharing a 3D virtual space for a synchronous distributed type design meeting, we developed a prototype system that enables participants to sketch or make annotations and have discussions as well as add viewpoints to them. We applied these functions to evaluate an architectural design and urban landscape examination. In conclusion, the proposed method was evaluated as being effective and feasible. Yet, it shows a few shortcomings including the fact that simultaneous operation is limited to one client, and more arbitrary shapes should be supported in future versions of the application.

A 2998 MHz, 60 MW klystron has been developed for future linear accelerators of synchrotron radiation sources.In order to save development and construction time, the 2998 MHz, 60 MW klystron is a modified version of an existing 2856 MHz klystron used in linear accelerator of Beijing Electron-Positron Collider Ⅱ.Based on the simulations, it was shown that the scaled version klystron was able to produce more than 60 MW at a 350 kV beam voltage.A prototype was built and tested.It successfully produced 50 MW output power with an RF pulse width of 4.0μs, which exceeded the operating requirement of China’s High Energy Photon source.It also produced 62 MW with an RF pulse width of 1.6μs, which proved the validity of new RF circuit design.This paper describes the details of the progresses of this klystron and advantages of this tube development.

BACKGROUND Gastric gastrinoma and spontaneous tumor regression are both very rarely encountered.We report the first case of spontaneous regression of gastric gastrinoma.CAsE sUMMARY A 37-year-old man with a 9-year history of chronic abdominal pain was referred for evaluation of an 8 cm mass in the lesser omentum discovered incidentally on abdominal computed tomography.The tumor was diagnosed as grade 2 neuroendocrine neoplasm(NEN)on endoscopic ultrasound-guided fine-needle aspiration.screening esophagogastroduodenoscopy revealed a 7 mm red polypoid lesion with central depression in the gastric antrum,also confirmed to be a grade 2 NEN.Laparoscopic removal of the abdominal mass confirmed it to be a metastatic gastrinoma lesion.The gastric lesion was subsequently diagnosed as primary gastric gastrinoma.Three months later,the gastric lesion had disappeared without treatment.The patient remains symptom-free with normal fasting serum gastrin and no recurrence of gastrinoma during 36 mo of follow-up.CONCLUsION Gastric gastrinoma may arise as a polypoid lesion in the gastric antrum.spontaneous regression can rarely occur after biopsy.

Great interest has been aroused on deeply-situated Osaka clay since Kobe Earthquake in 1994. In this paper is presented the analysis on the results of a series of lab tests on Osaka clay situated from 100 m to 1500 m under the ground. The wave velocity method, bender element method, LDT and the formula derived by the authors are used, focus is put on the pre-failure mechanical behavior of the clay. The analysis shows that, (i) pore-pressure coefficient B is less than 1.0, (ii) the relationship between shear modulus and Poisson's ratio is not in agreement with that reported before, (iii) the modulus measured with LDT is still less than that measured with bender element method, and (iv) there are two threshold strains, within which the clay can be considered as elastic, and both of them are larger than that reported before.

Effects of Earth's curvature and radial heterogeneity in dislocation studies:Case studies of the 2008 Wenchuan earthquake and the 2004 sumatra earthquake

Recently, effects of Earth＇s curvature and radial heterogeneity on coseismic deformations are often investi- gated based on the 2004 sumatra earthquake. However, such effects are strongly related to earthquake types. As a low dip angle event, the 2004 sumatra earthquake is not a good seismic case for such a topic since the effects for moderate dip angle events are much bigger. In this study, the half-space and spherical dislocation theories are used, respectively, to calculate co- seismic displacements caused by the 2008 Wenchuan earthquake and the 2004 sumatra earthquake. Effects of Earth＇s curva- ture and stratification are investigated through the discrepancies of results calculated using the two dislocation theories. Re- sults show that the effects of Earth＇s curvature and stratification for the 2008 Wenchuan earthquake are much larger than those for the 2004 sumatra earthquake. Ignoring the effects will cause errors up to 100%-200% in far field displacements for a moderate dip angle event like the 2008 Wenchuan earthquake. such great effects are much bigger than those conclusions of previous studies. Besides, comparison with observations verifies that spherical dislocation theories yield better results than half-space ones in far fields.
