Zinc finger factor ZNF121 is a MYC-interacting protein functionally affecting MYC and cell proliferation in epithelial cells

MYC is a potent oncoprotein that modulates multiple cellular processes including proliferation,apoptosis, differentiation, stemness, senescence, and migration. Functioning primarily as a transcriptional factor, MYC interacts with a large number of proteins, and identification and characterization of MYC-interacting proteins are important for understanding how MYC functions. In this study, we used different systems to demonstrate that a novel zinc finger transcription factor, ZNF121, physically interacted with MYC, and the interaction involved their N-terminal regions. Overexpression of ZNF121 increased, while its knockdown decreased, the expression of MYC in multiple epithelial cell lines, and MYC had similar effects on ZNF121 expression. An expression correlation was also detectable in a panel of epithelial cell lines and a cohort of human breast cancer tissues. Functionally, knockdown of ZNF121 in several breast epithelial cell lines attenuated the expressions of MYC and its target genes（e.g., EGR1,CDC2, and nucleolin） and slowed cell proliferation, accompanied by cell cycle arrest in the G1 phase and expression alteration of cell cycle regulators（cyclin D1, p14 and p21）. Analysis of publically available databases showed that ZNF121 expression is up-regulated in human breast cancer, and the up-regulation significantly associates with worse patient survival in the luminal A subtype of breast cancer. These findings establish ZNF121 as a MYC-interacting protein with functional effects on MYC and cell proliferation.

Spray forming and mechanical properties of a new type powder metallurgy superalloy

The deposited billet of a new type powder metallurgy （PM） superalloy FGH4095M for use in turbine disk manufac- turing has been fabricated using spray forming technology. The metallurgical quality of the deposited billet was analyzed in terms of density, texture, and grain size. Comparative research was done on the microstructure and mechanical properties between the flat disk preform prepared with hot isostatic pressing （HIP） and the same alloy forgings prepared with HIP followed by isothermal forging （IF）. The results show that the density of the spray-formed and nitrogen-atomized deposit billet is above 99% of the theoretical density, indicating a compact structure. The grains are uniform and fine. The billet has weak texture with a random distribution in the spray deposition direction and perpendicular to the direction of deposition. A part of atomizing nitrogen exists in the preform in the form of carbonitride. Nitrogen-induced microporosity causes the density reduction of the preform. Compared with the process of HIP＋IF, the superalloy FGH4095M after HIP has better mechanical properties at both room temperature and high temperature. The sizes of the 7~ phase are finer in microstructure of the preform after HIP in comparison with the forgings after HIP＋IE This work shows that SF＋HIP is a viable processing route for FGH4095M as a turbine-disk material.

Nanoceramic composites with duplex microstructure break the strength-toughness tradeoff

There is a strength and fracture toughness tradeoff in nanoceramic composite. The strength varies reciprocally with the grain size whereas the toughness contributed by compressive residual stress increases with the dimension of the second phase. In this work, a novel duplex microstructure with reinforced clusters composing of nanosized grains was proposed and validated using a model system of B_(4)C-TiB_(2) ceramics densified by carbide boronizing. As-obtained ceramics exhibit excellent combined mechanical properties at room temperature, including Vickers hardness, Young's modulus, flexural strength and fracture toughness(by surface crack in flexure method) of 32.1 ± 2.7 GPa, 506.9 ± 2.0 GPa,1175 ± 71 MPa and 5.1 ± 0.4 MPa m^(0.5), respectively. Both strength and toughness are at least ~30 % higher than the counterparts with similar composition but homogenously distributed TiB_(2) grains. Graphite onion was confirmed to be an intermediate product during reactive sintering, it facilitated the grain pullout during fracture and retained the nanometric TiB_(2) grain in the cluster, both of which also contribute the toughening and strengthening mechanisms in the B_(4)C-TiB_(2) ceramics.

Introduction The endothelial cells(ECs)lining every blood vessel wall constantly expose to the mechanical forces generated by the blood *** EC responses to these hemodynamic forces play a critical role in the homeostasis of the circulatory *** addition to forming a transport barrier between the blood and vessel wall,vascular ECs play important roles in regulating circulation *** biochemical stimuli,blood flow induced(hemodynamic)mechanical stimuli,such as shear stress,pressure and circumferential stretch,modulate EC morphology and functions by activating mechanosensors,signaling pathways,and gene and protein *** EC responses to the hemodynamic forces(mechano-sensing and transduction)are critical to maintaining normal vascular *** in the mechano-sensing and transduction leads to serious vascular diseases including hypertension,atherosclerosis,aneurysms and thrombosis,to name a few[1].On the luminal surface of our blood vessels,there is a thin layer called endothelial surface glycocalyx(ESG)which consists of proteoglycans,glycosaminoglycans(GAGs)and *** GAGs in the ESG are heparan sulfate(HS),hyaluronic acid(HA),chondroitin sulfate(CS),and sialic acid(SA)[2].In order to play important roles in vascular functions,such as being a mechanosensor and transducer for the endothelial cells(ECs)to sense the blood flow,a molecular sieve to maintain normal microvessel permeability and a barrier between the circulating cells and endothelial cells forming the vessel wall,the ESG should have an organized structure at the molecular *** to the limitations of optical and electron microscopy,the ultra-structure and organization of ESG has not been revealed until recent development of a super high resolution fluorescence optical microscope,STORM(Stochastic Optical Reconstruction Microscopy).The diffraction of a single fluorescence molecule can be described as the point spread function(PSF).When the light of wavelengt

Effect of sulfate-reducing bacteria on the corrosion behaviors of API-X80 pipeline steel has been studied in crevices under simulated disbonded coatings in red soil *** results show that there are amounts of SRB in the crevice under the simulated disbonded coating in the red soil solution during the whole period of *** electrochemical impedance spectroscopy shows that SRB can enhance the corrosion of the steel in the crevice under the simulated disbonded coating,and the thermodynamic analyses indicate that SRB can accelerate the corrosion of steel using not only sulfate but also ferric hydroxide as electron acceptors.

Acute Hip Pain—A Forgotten Cause

This is a case of acute left hip pain in a young male patient presenting to the Emergency Department (ED). The hip joint is painful with limited range of motion, and the patient is admitted to rule out septic arthritis of the hip. The subsequent inpatient serum uric acid level performed is elevated and the Magnetic Resonance Imaging (MRI) hip reveals a moderate hip effusion. A Computer Tomography (CT) guided hip joint aspiration confirms the diagnosis of gout with uric acid crystals seen in the hip synovial fluid analysis. The patient is treated as for gout with marked improvement in his symptoms.

Clustering behavior of solid particles in two-dimensional liquid-solid fluidized-beds

In this paper, the clustering behavior of solid particles in a two-dimensional (2D) liquid-solid fluidized-bed was studied by using the charge coupled devices (CCD) imaging measuring and processing technique and was characterized by fractal analysis. CCD images show that the distribution of solid particles in the 2D liquid-solid fluidised-bed is not uniform and self-organization behavior of solid particles was observed under the present experimental conditions. The solid particles move up in the 2D fluidized-bed in groups or clusters whose configurations are often in the form of horizontal strands. The box fractal dimension of the cluster images in the 2D liquid-solid fluidized-bed increases with the rising of solid holdup and reduces with the increment of solid particle diameter and superficial liquid velocity. At given solid holdup and solid particle size, the lighter particles show smaller fractal dimensions.

Electro-hydraulic actuator （EHA） system inherently suffers from uncertainties, nonlinearities and time-varying in its model parameters which makes the modeling and controller designs are more complicated. The main objective of this paper is to perform a robust control design using discrete-time sliding mode control （DSMC） with two-degree-of-freedom control strategy. The proposed controller consists of feedback and feedforward combination which capable to reduce phase lag during the trajectory tracking of EHA system. The feedforward controller is developed by implementing the zero phase error tracking control （ZPETC） technique which the main difficulty arises from the nonminimum phase system with no stable inverse. Finally, a chaotic trajectory tracking is performed in the experimental works to show the robustness of DSMC controller and performance comparison is made with Linear-Quadratic-Regulator （LQR） and Proportional-Integral-Derivative （PID） controllers. The findings show that the proposed controller with ZPETC outperforms the LQR and PID controllers in terms of tracking accuracy.
