PCR-SSCP-DNA sequencing method in detecting PTEN gene mutation and its signifi cance in human gastric cancer

AIM: To discuss the possible effect of PTEN gene mutations on occurrence and development of gastric cancer. METHODS: Fifty-three gastric cancer specimens were selected to probe PTEN gene mutations in genome of gastric cancer and paracancerous tissues using PCR-SSCP-DNA sequencing method based on microdissection and to observe the protein expression by immunohistochemistry technique. RESULTS: PCR-SSCP-DNA sequencing indicated that 4 kinds of mutation sites were found in 5 of 53 gastric cancer specimens. One kind of mutation was found in exons. AA-TCC mutation was located at 40bp upstream of 3’ lateral exon 7 (115946 AA-TCC). Such mutations led to terminator formation in the 297th codon of the PTEN gene. The other 3 kinds of mutation were found in introns,including a G-C point mutation at 91 bp upstream of 5’ lateral exon 5(90896 G-C),a T-G point mutation at 24 bp upstream of 5’ lateral exon 5 (90963 T-G),and a single base A mutation at 7 bp upstream of 5’ lateral exon 5 (90980 A del). The PTEN protein expression in gastric cancer and paracancerous tissues detected using immunohistochemistry technique indicated that the total positive rate of PTEN protein expression was 66% in gastric cancer tissue,which was significantly lower than that (100%) in paracancerous tissues (P < 0.005). CONCLUSION: PTEN gene mutation and expression may play an important role in the occurrence and development of gastric cancer.

High levels of cell-free DNA(cfDNA)induce ***,the treatment of psoriasis has the disadvantages of penetration difficulty,suppression of normal immunity,and skin *** this study,biguanide chitosan microneedles(BGC-MNs)were prepared to treat psoriasis by removing cfDNA from the dermis through the skin *** effects of chitosan with different bisguanidine contents on DNA-binding capacity,biocompatibility,and inflammation inhibition were compared,revealing that chitosan containing 20%bisguanidine(BGC2)was found to have the best overall *** vitro,BGC2 effectively cleared cfDNA and inhibited the production of inflammatory ***-MN made from BGC2 had good mechanical and solubility *** vivo,BGC-MNs cleared cfDNA,reduced the level of inflammatory factors in the dermis,and exerted a good therapeutic effect on mice with *** results suggested that BGC-MNs provided a new approach to treating psoriasis in terms of scavenging cfDNA and exerting anti-inflammatory effects.

Nonadiabatic holonomic quantum computers serve as the physical platform for nonadiabatic holonomic quantum *** quantum computation has entered the noisy intermediate-scale era, building accurate intermediate-scale nonadiabatic holonomic quantum computers is clearly necessary. Given that measurements are the sole means of extracting information, they play an indispensable role in nonadiabatic holonomic quantum computers. Accordingly, developing methods to reduce measurement errors in nonadiabatic holonomic quantum computers is of great importance. However, while much attention has been given to the research on nonadiabatic holonomic gates, the research on reducing measurement errors in nonadiabatic holonomic quantum computers is severely lacking. In this study, we propose a measurement error reduction method tailored for intermediate-scale nonadiabatic holonomic quantum computers. The reason we say this is because our method can not only reduce the measurement errors in the computer but also be useful in mitigating errors originating from nonadiabatic holonomic gates. Given these features,our method significantly advances the construction of accurate intermediate-scale nonadiabatic holonomic quantum computers.

The infections in open fracture induce high morbidity ***,developing efficient anti-infective orthopedic devices is of great *** this work,we designed a kind of infection-responsive long-term antibacterial bone *** a facile and flexible volatilization method,a multi-aldehyde polysaccharide derivative,oxidized sodium alginate,was crosslinked with multi-amino compounds,gentamycin and gelatin,to fabricate a uniform coating on Ti bone plates via Schiff base reaction,which was followed by a secondary crosslinking process by *** double-crosslinked coating was stable under normal condition,and could responsively release gentamycin by the triggering of the acidic microenvironment caused by bacterial metabolism,owning to the pH-responsiveness of imine *** thickness of the coating was ranging from 22.0μm to 63.6μ*** coated bone plates(Ti-GOGs)showed infection-triggered antibacterial properties(>99%)and high *** being soaked for five months,it still possessed efficient antibacterial ability,showing its sustainable antibacterial *** in vivo anti-infection ability was demonstrated by an animal model of infection after fracture fixation(IAFF).At the early stage of IAFF,Ti-GOGs could inhibit the bacterial infection(>99%).Subsequently,Ti-GOGs could promote recovery of fracture of *** work provides a convenient and universal strategy for fabrication of various antibacterial orthopedic devices,which is promising to prevent and treat IAFF.

It is of great importance to treat a bacterial-infected wound by a smart dressing capable of delivering antibiotics in a smart manner without causing drug *** construction of smart release nanocontainers responsive to near-infrared(NIR)laser irradiation in an on-demand and stepwise way is a promising strategy for avoiding the emergence of multidrug-resistant ***,we develop a hydrogel composite made of alginate and nanotubes with an efficient NIR-triggered release of rifampicin and outstanding antibacterial *** composite hydrogel is prepared through co-encapsulating antibacterial drug(rifampicin),NIR-absorbing dye(indocyanine green),and phase-change materials(a eutectic mixture of fatty acids)into halloysite nanotubes,followed by incorporation into alginate hydrogels,allowing the in-situ gelation at room temperature and maintaining the integrity of drug-loaded *** them,the eutectic mixture with a melting point of 39℃ serves as the biocompatible phase-change material to facilitate the NIR-triggered drug *** resultant phase-change material gated-nanotubes exhibit a prominent photothermal efficiency with multistep drug release under laser *** an in vitro assay,composite hydrogel provides good antibacterial potency against Staphylococcus aureus,one of the most prevalent microorganisms of dangerous gas gangrene.A bacterial-infected rat full-thickness wound model demonstrates that the NIR-responsive composite hydrogel inhibits the bacteria colonization and suppresses the inflammatory response caused by bacteria,promoting angiogenesis and collagen deposition to accelerate wound *** NIR-responsive composite hydrogel has a great po-tential as an antibacterial wound dressing functionalized with controlled multistep treatment of the infected sites.
