N6-methyladenosine(m6A) is a critical regulator in the fate of RNA, but whether and how m6A executes its functions in different tissues remains largely obscure. Here we report downregulation of a crucial m6A reader, YTHDF2, leading to tissue-specific programmed cell deaths(PCDs) upon fluorene-9-bisphenol(BHPF) exposure. Currently, Bisphenol A(BPA) substitutes are widely used in plastic *** eight common BPA substitutes, we detected BHPF in 14% serum samples of pregnant participants. In a zebrafish model, BHPF caused tissue-specific PCDs triggering cardiac and vascular ***, BHPF-mediated downregulation of YTHDF2 reduced YTHDF2-facilitated translation of m6A-gch1 for cardiomyocyte ferroptosis, and decreased YTHDF2-mediated m6A-sting1 decay for caudal vein plexus(CVP) apoptosis. The two distinct YTHDF2-mediated m6A regulations and context-dependent co-expression patterns of gch1/ythdf2 and tnfrsf1a/ythdf2 contributed to YTHDF2-mediated tissue-specific PCDs, uncovering a new layer of PCD regulation. Since BHPF/YTHDF2-medaited PCD defects were also observed in mammals, BHPF exposure represents a potential health threat.

At least six major genotypes of Hepatitis C virus （HCV） cause liver diseases worldwide. The efficacy rates with current standard of care are about 50% against genotype 1, the most prevalent strain in the United States, Europe and Japan. Therefore more effective pan-genotypic therapies are needed. HCV RNA replication provides a number of validated targets for virus-specific and potentially pan-genotypic inhibitors. In vitro assays capturing the different steps of RNA synthesis are needed not only to identify new inhibitors, but also to examine their mechanisms of action. This review attempts to provide a comprehensive summary of the biochemical, cell-based and animal model systems to assess HCV polymerase activity and HCV RNA replication that should be useful for both basic research and applied studies.

为提高云制造服务的聚类质量,提出一种融合多维属性相似度的云制造服务谱聚类算法。将云制造服务的文本型属性值向量化和数值型属性值归一化,分别构建文本型属性相似度矩阵和数值型属性相似度矩阵。设计一种属性相似度融合函数,实现多维度云制造服务属性相似度的融合,建立云制造服务相似度矩阵。引入相似矩阵的本征间隙确定聚类个数,实现高效、精准的云制造服务谱聚类。实验结果表明,该方法较其它聚类方法显著提高了云制造服务的聚类质量。

1840年以降,英国大众报业和公民教育逐步增强了自然知识的社会属性。此后,英国德文郡及近乡居民对达特莫尔荒原的热爱之情和保护行动通过报刊撰稿人和教育者的生动言说,引发了外乡民众对达特莫尔荒原的无限遐思。至1951年国家公园建立前,达特莫尔荒原已成为英国民众心中“风景如画之不列颠”的重要组成部分。历史经验表明,地方民众在充分的人地互动中得出的环保经验,能够在大众媒体和大众教育的传播下成为全民共享的环保理念。此种汇集民众智慧的环保选择,既能为我国国家公园的选建及管理节约大量的社会成本,亦能促使我国国家公园建设事业更好地满足民众的自然审美及户外休闲需求。

Serum Metabonomics of Articular Cartilage Destruction Induced by T-2 Toxin in Wistar Rats

The molecular pathogenesis of T-2 toxin-induced cartilage destruction has not been fully unraveled yet. The aim of this study was to detect changes in serum metabolites in a rat anomaly model with articular cartilage destruction. Thirty healthy male Wistar rats were fed a diet containing T-2 toxin （300 ng/kg chow） for 3 months. Histopathological changes in femorotibial cartilage were characterized in terms of chondrocyte degeneration/necrosis and superficial cartilage defect, and the endogenous metabolite profile of serum was determined by UPLC/Q-TOF MS. Treated rats showed extensive areas of chondrocyte necrosis and superficial cartilage defect in the articular cartilage. In addition, 8 metabolites were found to change significantly in these rats compared to the control group, including lyso PE （18：0/0：0）, lyso PC（14：0）, lyso PC[18：4 （6Z,9Z,12Z,15Z）], lyso PC[（16：1（9Z）], lyso PC（16：0）, L-valine, hippuric acid, and asparaginyl-glycine. These 8 metabolites associated with cartilage injury are mainly involved in phospholipid and amino acid metabolic pathways.

Located at the collision boundary between the Philippine Sea Plate and the Eurasian Continental Plate,the island of Taiwan is generally recognized as an important example in the MARGINS Program Science Plan and "source-to-sink" research because of its high tectonic activity,heavy rainfall and unique *** suspended sediment loads are transported to the adjacent ocean by Taiwan Residents rivers every year,making Taiwan an important source of sediments into the adjacent seas and a natural laboratory for studying the systemic movement of fluvial sediments from source to sink.A detailed study on the clay mineral composition of surface sediments collected from the drainage basins of 12 Taiwan Residents rivers using X-ray diffraction methods was *** results indicated that the clay mineral assemblages consisted dominantly of illite(approximately 73%) and chlorite(approximately 24%),with lesser abundances of kaolinite(approximately 3%) and even lower levels of smectite from the Danshuei River sediments in northwestern *** Jhuoshuei River sediments from western Taiwan contained clay mineral assemblages that consisted of illite(approximately 75%) and chlorite(approximately 25%),but they lacked kaolinite and *** southwestern Taiwan,the clay mineral assemblages were dominated by illite(approximately 75%) and chlorite(approximately 23%),but had a low abundance of kaolinite(generally kaolinite and smectite being scarce,and these trends are different from those in China's mainland *** clay miner

Objective:To investigate the efficacy of frankincense and myrrha in the treatment of acute interstitial cystitis/painful bladder syndrome(IC/PBS).Methods:The effects of frankincense and myrrha on the proliferation and migration of primary human urothelial cells(HUCs)were assessed in *** the animal study,48 virgin female rats were randomized into 4 groups(12 in each group):(1)control group(saline-injected control);(2)cyclophosphamide(CYP)group(intraperitoneal injected 150 mg/kg CYP);(3)CYP+pentosan polysulfate sodium group(orally received 50 mg/kg pentosan polysulfate sodium);and(4)CYP+frankincense and myrrha group[orally received frankincense(200 mg/kg)and myrrha(200 mg/kg)].Rats orally received pentosan polysulfate sodium or frankincense and myrrha on day 1,2,and *** experiments were performed on day *** and cystometry assessment behavior test were *** interval values were assessed in rats under ***,immunohistochemistry and Western blot were used to confirm the location and level,respectively,of cell junction-associated protein zonula occludens-2(ZO-2)***:Low dose frankincense and myrrha increased cell proliferation and migration in HUCs compared with control(P<0.05).Rats with acute IC/PBS rats exhibited lower voiding interval values,pain tolerance,and ZO-2 expression(P<0.05).Voiding interval values and pain tolerance were higher in the frankincense and myrrha group than CYP group(P<0.05).ZO-2 expression in the bladder was increased in the CYP+pentosan polysulfate and frankincense+myrrha groups compared with the CYP-induced acute IC/PBS group(P<0.05).Conclusion:frankincense and myrrha modulate urothelial wound healing,which ameliorates typical features of acute IC/PBS in rats.
