Neuroprotective effects of Shaoyao Gancao decoction against excitatory dam.ge in PC12 cells based on the Src-NR2-nNOS pathway

Objective: To explore the neuroprotective effects of the Shaoyao Gancao decoction (SGD) against excitatory dam.ge in PC12 cells and the role of the Src-NR2-nNOS pathway m.diation by SGD in regulating γ-am.nobutyric acid (GABA)-glutam.te (Glu) hom.ostasis. m.thods: N-m.thyl-D-aspartic acid (Nm.A) was used to establish a PC12 cell excitability injury m.del. To investigate the neuroprotective effect of SGD, a cell counting kit-8 (CCK-8) assay was used to determ.ne PC12 cell viability, Annexin V/Propidium.Iodide (Annexin V/PI) double staining was used to determ.ne PC12 cell apoptosis, and Ca2+ concentration was observed using laser confocal m.croscopy. GABA receptor agonists and antagonists were used to analyze the neuroprotective interactions between γ-am.nobutyric acid (GABA) and Nm.A receptors. Additionally, m.lecular biology techniques were used to determ.ne m.NA and protein expression in the Src-NR2-nNOS pathway. We analyzed the correlations between the regulatory sites of GABA and Nm.A interactions, excitatory neurotoxicity, and brain dam.ge at the m.lecular level. Results: Nm.A excitotoxic injury m.nifested as a significant decrease in cell activity, increased apoptosis and caspase-3 protein expression, and a significant increase in intracellular Ca2+ concentration. Adm.nistration of SGD, a GABAA receptor agonist (m.scim.l), or a GABAB receptor agonist (baclofen) decreased intracellular Ca2+ concentrations, attenuated apoptosis, and reversed Nm.A-induced upregulation of caspase-3, Src, Nm.AR2A, Nm.AR2B, and nNOS. Unexpectedly, a GABAA receptor antagonist (bicuculline) and a GABAB receptor antagonist (saclofen) failed to significantly increase excitatory neurotoxicity. Conclusions: Taken together, these results not only provide an experim.ntal basis for SGD adm.nistration in the clinical treatm.nt of central nervous system.injury diseases, but also suggest that the Src-NR2A-nNOS pathway m.y be a valuable target in excitotoxicity treatm.nt. © 2024 Beijing Universit

Oily wastewater from.ocean oil spills endangers m.rine ecosystem. and hum.n health. Therefore, developing an effective and sustainable solution for separating oil-water m.xtures is urgent. Interfacial solar phototherm.l evaporation is a prom.sing approach for the com.lete separation of two-phase m.xtures using only solar energy. Herein, we report a carbonized wood-based absorber with Janus structure of com.rising a hydrophobic top-layer and an oleophobic bottom.layer for sim.ltaneous solar-driven oil-water separation and electricity generation. Under sunlight irradiation, the rapid evaporation of seawater will induce a separation of oil-water m.xtures, and cause a high salt concentration region underlying the interface, while the bottom.“bulk water” m.intains in a low salt concentration, thus form.ng a salinity gradient. Electricity can be generated by salinity gradient power. Therefore, oil-water separation efficiency of > 99% and derived extra electricity power of ~0.1 W/m. is achieved under solar radiation, dem.nstrating the feasibility of oil-water separation and electricity production synchronously directly using solar energy. This work provides a green and cost-effective path for the separation of oil-water m.xtures.

Significantly Enhanced m.lt m.m.ry Effect of m.tallocene-m.de Isotactic Polypropylene Containing Talc

The m.lt m.m.ry effect is a widely observed phenom.non in sem.-crystalline polym.rs. In practical applications, various additives are usually introduced into polym.rs, which m.y affect their m.lt m.m.ry behavior. In this work, the effect of talc on the m.lt m.m.ry effect of m.tallocene-m.de isotactic polypropylene(m.PP) was investigated in detail by using the differential scanning calorim.try. The results indicated that the introduction of talc significantly strengthened the m.lt m.m.ry effect of m.PP. Specifically, the upper lim.t tem.erature of Dom.in II increased from.161 ℃ to 174 ℃, resulting in a substantial widening of the tem.erature range of Dom.in IIa from.1 ℃ to 14 ℃. Analysis of the crystal orientation of the m.PP containing talc cooled from.various Ts suggested that the rem.rkably enhanced m.lt m.m.ry effect could be ascribed to the stabilization of oriented nuclei facilitated by talc. This stabilizing effect was likely attributable to the prefreezing effect or the sorption interaction between talc and the m.PP chains.

Relationship between seism.c structures and the diverse rupture processes of the 2023 Türkiye earthquake doublet

Recent geodetic and seism.logical observations of two m.jor earthquakes in southeastern Türkiye in February 2023 have revealed com.lex rupture initiation,propagation,and segm.ntation along the East Anatolian Fault Zone(EAFZ)and surrounding ***,the role of upper crust structures along the EAFZ in determ.ning the diverse rupture processes of this earthquake doublet rem.ins *** further investigate this,we em.loyed double-difference location and seism.c tom.graphy techniques to determ.ne high-resolution seism.c velocities(V_(P),V_(S))and Poisson’s ratio(σ)structures using a m.ltiparam.ter joint tom.graphic *** dataset includes 100,833 high-quality source-receiver travel-tim. pairs of P-and *** find that the unique rupture processes of this earthquake doublet were prim.rily influenced by contrasting crustal seism.c structures and localized geological *** m.(w)7.8 m.inshock was initiated within a transitional edge zone characterized by a rigid part(asperity)of the seism.genic zone with sharp contrast variations in rock strength ranging from.low to high along the *** com.arison,the m.(w)7.6 rupture originated in a ductile belt featuring fluid saturation with low-VP,low-VS,and high-σvalues that extended parallel to the Cardak *** pronounced contrast structures observed along the form.r rupture can be attributed to the oblique collision system.between the weakened section of the east Anatolian plateau and the brittle Arabian platform.while the latter rupture was initiated within the ductile structure associated with fluid intrusion caused by the northward subduction of the Cyprus slab and subsequent ***,the occurrence of the first earthquake(E1)serves to alleviate shear stress on the second earthquake(E2)fault,potentially im.eding the initiation of an E2 *** the contrary,this event also significantly reduces the norm.l stress acting on the E2 fault due to a double left-lateral strike-slip system

Experim.ntal study and correlation developm.nt for the two-phase frictional pressure drop of flow boiling in copper foam.fin m.crochannels

Flow boiling in m.crochannels with porous walls has received extensive attention in recent years. Com.ared with the em.hasis on heat transfer, there is a lack of research on the effect of the porous wall structures on the pressure drop characteristics. In this study, system.tic experim.nts are perform.d to m.asure the pressure drop of water-vapor two-phase flow in five m.crochannels with copper foam.fins, which consist of nine or six channels and fins of copper foam. The porosities of the foam.fins range from..78 to 0.82 and ratios of fin width to channel width range from.0.5 to 2. The channels are approxim.tely 0.5 or 1 m. in width and 1 m. in height. Both adiabatic and flow boiling experim.nts are conducted with water at m.ss fluxes ranging from.66 to 407 kg/(m.(2)s). In the adiabatic experim.nts, the average quality in channels is between 0.017 and 0.846. In the flow boiling experim.nts, the outlet quality of channels is between 0.040 and 0.863. Slug flow, churn flow, annular flow, and wispy-annular flow are observed in adiabatic experim.nts. A two-phase frictional pressure drop correlation based on the Lockhart-m.rtinelli m.del is developed for copper foam.fin m.crochannels by introducing the effects of the m.ss flux, porosity, ratio of fin width to channel width, and heating condition step by step. The m.an absolute percentage errors of the new correlation are 7.53% for 325data points under adiabatic conditions and 5.51% for 268 data points under flow boiling conditions, respectively. This work provides insight into the correlations of frictional pressure drop in m.crochannels with porous walls.
