Nitric oxide(NO)is an essential reactive oxygen species and a signal molecule in *** several studies have proposed the occurrence of oxidative NO production,only reductive routes for NO production,such as the nitrate(NO_(3)^(-))-upper-reductase pathway,have been evidenced to date in land ***,plants grown axenically with ammonium as the sole source of nitrogen exhibit contents of nitrite and NO3−,evidencing the existence of a metabolic pathway for oxidative production of *** hypothesized that oximes,such as indole-3-acetaldoxime(IAOx),a precursor to indole-3-acetic acid,are intermediate oxidation products in NO *** detected the production of NO from IAOx and other oximes catalyzed by peroxidase(POD)enzyme using both 4-amino-5-methylamino-2′,7′-difluorescein fluorescence and *** stimulated the reaction,while superoxide dismutase inhibited ***,mouse NO synthase can also use IAOx to produce NO at a lower rate than *** provided a full mechanism for POD-dependent NO production from IAOx consistent with the experimental data and supported by density functional theory *** showed that the addition of IAOx to extracts from medicago truncatula increased the in vitro production of NO,while in vivo supplementation of IAOx and other oximes increased the number of lateral roots,as shown for NO donors,and a more than 10-fold increase in IAOx dehydratase ***,we found that in vivo supplementation of IAOx increased NO production in Arabidopsis thaliana wild-type plants,while prx33-34 mutant plants,defective in POD33-34,had reduced *** data show that the release of NO by IAOx,as well as its auxinic effect,explain the superroot ***,our study reveals that plants produce NO utilizing diverse molecules such as oximes,POD,and flavins,which are widely distributed in the plant kingdom,thus introducing a long-awaited oxidative pathway to NO production in plants.T

Biotransformation of 6:2 fluorotelomer sulfonate (6:2 FTS) by two species of white-rot fungi, Pleurotus ostreatus (P. ostreatus) and Trametopsis cervina (T. cervina), was investigated in a sulfur-rich medium designed to stimulate production of lignin-degrading enzymes. Degradation of 6:2 FTS was observed by T. cervina over the study period of 30 d, but not by P. ostreatus. Biotransformation rates were comparable to those found in other studies investigating mixed culture degradation in non-sulfur limiting media, with approximately 50 mol% of applied 6:2 FTS removed after 30 d. Stable transformation products were short-chain perfluorocarboxylic acids (PFCAs), including PFHxA (2.27 mol%), PFPeA (0.24 mol%), and PFBA (0.28 mol%). The main intermediate products include 5:2 sFTOH (16.3 mol%) and 5:3 FTCA (2.99 mol%), while 6:2 FTCA, 6:2 FTuCA, and 5:2 ketone were also identified at low levels. Approximately 60 mol% of detected products were assigned to the major pathway to 5:2 ketone, and 40 mol% were assigned to the minor pathway to 5:3 FTCA. The overall molar balance was found to decrease to 75 mol% by Day 30, however, was closed to near 95 mol% with a theoretical estimation for the volatile intermediates in the headspace, 5:2 ketone and 5:2 sFTOH. The different capabilities of the two white-rot fungal species for 6:2 FTS biotransformation in sulfur-rich media suggest that the enzyme processes of T. cervina to de-sulfonate 6:2 FTS may be unrelated to sulfur metabolism. Background The mental health of current medical students is predictive of their mental health as future *** prevalence of anxiety,depression and burnout is high among medical students,but less is known about the occurrence of other mental ill-health symptoms,such as eating or personality disorders,and factors contributing to mental ***(1)To explore the prevalence of various mental ill-health symptoms in medical students and(2)to investigate what medical school factors and students’attitudes contribute to these mental ill-health *** Between November 2020 and may 2021,medical students from nine geographically spread medical schools in the UK participated by completing online questionnaires at two points in time,approximately 3 months *** Of the 792 participants who filled in the questionnaire at baseline,over half experienced medium to high somatic symptoms(50.8%;402)and drank alcohol at hazardous levels(62.4%;494).Adjusted longitudinal data analysis of 407 students who completed the follow-up questionnaire demonstrated that less supportive educational climates that were more competitive and less centralised around the students,lower feelings of belongingness,greater stigma towards mental ill-health and lower intentions to seek help for mental ill-health,all contributed to students’mental ill-health *** medical students experience a high prevalence of various mental ill-health *** study suggests that medical school factors and students’attitudes towards mental ill-health are significantly associated with students’mental health.

AIm:To investigate in vitro treatment with NVPAEW541,a small molecule inhibitor of insulin-like growth factor-1 receptor(IGF-1R),in biliary tract cancer (BTC),since this disease is associated with a poor prognosis due to wide resistance to chemotherapeutic agents and ***:Cell growth inhibition by NVP-AEW541 was studied in vitro in 7 human BTC cell lines by automated cell *** addition,the anti-tumoral mechanism of NVP-AEW541 was studied by Western blotting,cell cycle analysis and reverse transcription-polymerase chain reaction(RT-PCR).Anti-tumoral drug effect in combination with gemcitabine,5-fluorouracil(5-FU)and Polo-like kinase 1 inhibitor BI2536 was also studied. RESULTS:In vitro treatment with NVP-AEW541 suppressed growth in all human BTC cell lines,however response was lower in gallbladder *** withNVP-AEW541 was associated with dephosphorylation of IGF-1R and *** contrast,phosphorylation of p42/p44 and Stat3 and expression of Bcl-xL were inconsistently *** addition,treated cells showed cell cycle arrest at the G1/S-checkpoint and an increase in sub-G1 ***,IGF-1R and its ligands IGF-1 and IGF-2 were co-expressed in RT-PCR,suggesting an autocrine loop of tumor cell *** with gemcitabine,NVP-AEW541 exerted synergistic effects, particularly at low concentrations,while effects of combination with 5-FU or BI 2536 were only additive. CONCLUSION:Our findings suggest that NVP-AEW541 is active against BTC in vitro and potentiates the efficacy of gemcitabine.

Rock samples' TOC content is the best indicator of the organic matter in source rocks. The origin rock samples' analysis is used to calculate it manually by specialists. This method requires time and resources because it relies on samples from many well intervals in source rocks. Therefore, research has been done to aid this effort. machine learning algorithms can estimate total organic carbon instead of well logs and stratigraphic studies. In light of these efforts, the current work present a study on automating the total organic carbon estimation using machine learning approaches improved by an evolutionary methodology to give the model flexibility and precision. Genetic algorithms, differential evolution, particle swarm optimization, grey wolf optimization, artificial bee colony, and evolution strategies were used to improve machine learning models to predict TOC. The six metaheuristics were integrated into four machine learning methods: extreme learning machine, elastic net linear model, linear support vector regression, and multivariate adaptive regression splines. Core samples from the YuDong-Nan shale gas field, located in the Sichuan basin, were used to evaluate the hybrid strategy. The findings show that combining machine learning models with an evolutionary algorithms in a hybrid fashion produce flexible models that accurately predict TOC. The results show that, independent of the metaheuristic used to guide the model selection, optimized extreme learning machines attained the best performance scores according to six metrics. Such hybrid models can be used in exploratory geological research,particularly for unconventional oil and gas resources.
