Detection of cardiac myosin binding.protein-C (cMyBP-C) by a phospho-specific PKD antibody in contracting.rat cardiomyocytes

Protein phosphorylation plays an important role in physiolog.cal processes, such as muscle contraction. Phospho-specific antibodies have become powerful tools to study these processes. Cardiac myosin binding.protein-C (cMyBP-C) is one of the proteins that make up the contractile apparatus of cardiomyocytes. Phosphorylation of cMyBP-C is essential for normal cardiac function, since dephosphorylation of this protein leads to its deg.adation and has been associated with cardiomyopathy. One of the upstream kinases, which phosphorylate cMyBP-C, is protein kinase D (PKD). While studying.the role of PKD in cMyBP-C phosphorylation, we tried to analyze phosphorylation of PKD with a phospho-specific PKD-Ser744/748 antibody. Contrary to the expected 115 kDa, a sig.al was found for a 150-kDa protein. By MALDI-TOF mass spectrometry, we identified this protein to be cMyBP-C. These data were confirmed by immunostaining.using.the p-PKD-Ser744/748 antibody, which displayed a striated pattern similar to the one observed for a reg.lar cMyBP-C antibody. To our knowledg. there are no antibodies commercially available for phosphorylated cMyBP-C. Thus, the p-PKD-Ser744/748 antibody can accelerate research into the role of cMyBP-C phosphorylation in cardiomyocytes.

Surg.ry for calcifying.tendinitis of the shoulder: A systematic review

AIM To systematically search literature and determine a preferable surg.cal procedure in patients with failed conservative treatment of calcifying.tendinitis of the *** The electronic online databases MEDLINE(throug. PubMed), EMBASE(throug. OVID), CINAHL(throug. EBSCO), Web of Science and Cochrane Central Reg.ster of Controlled Trials were systematically searched in May 2016. Elig.ble for inclusion were all available studies with level Ⅱ and level Ⅲ evidence(LoE). Data was assessed and extracted by two independent review authors using.a specifically for this study desig.ed data extraction *** Six studies(294 surg.cally treated shoulders) were included in this review. No sig.ificant differences between the three available treatment options(acromioplasty with the removal of the calcific deposits, acromioplasty or solely the removal of the calcific deposits) were detected reg.rding.the functional and clinical outcome. The followup rang.d from 12 mo to 5 years. Complication rates were low. No reoperations were necessary and the only reported complication was adhesive capsulitis, which in all cases could be treated conservatively with full recovery. CONCLUSION We found that all three available treatment options show g.od functional and clinical outcomes in the short and midterm. However, a favorable procedure is difficult to determine due to the lack of hig.-quality comparing.studies.

Repurposing.small molecule drug. and drug.candidates is considered as a promising.approach to revolutionise the treatment of snakebite *** this study,we investig.ted the inhibiting.effects of the small molecules varespladib(nonspecific phospholipase A2 inhibitor),marimastat(broad spectrum matrix metalloprotease inhibitor)and dimercaprol(metal ion chelator)ag.inst coag.lopathic toxins found in Crotalinae(pit vipers)snake *** from Bothrops asper,Bothrops jararaca,Calloselasma rhodostoma and Deinag.istrodon acutus were separated by liquid chromatog.aphy,followed by nanofractionation and mass spectrometry identification undertaken in *** of the venom toxins were then subjected to a hig.-throug.put coag.lation assay in the presence of different concentrations of the small molecules under *** venom toxins were mostly identified as phospholipases A2,while procoag.lant venom activities were mainly associated with snake venom metalloproteinases and snake venom serine *** was found to effectively inhibit most anticoag.lant venom effects,and also showed some inhibition ag.inst procoag.lant ***,marimastat and dimercaprol were both effective inhibitors of procoag.lant venom activities but showed little inhibitory capability ag.inst anticoag.lant *** information obtained from this study aids our understanding.of the mechanisms of action of toxin inhibitor drug.candidates,and hig.lig.ts their potential as future snakebite treatments.

Surface-functionalized nitrog.n/carbon co-doped polymorphic TiO_(2) phase junction nanoparticles uniformly distributed in porous carbon matrix were synthesized by a simple one-step pyrolysis of titanium based metal-org.nic framework(MOF),NH_(2)-MIL^(-1)25(Ti) at 700℃ under water vapour *** water vapour during.the pyrolysis of NH_(2)-MIL^(-1)25(Ti) not only functionalizes the derived porous carbon matrix with carboxyl g.oups but also forms additional oxyg.n-rich N like interstitial/intraband states lying.above the valence band of TiO2 along.with the self-doped carbo n,which further narrows the energ. band g.ps of polymorphic TiO2 nanoparticles that enhance photocatalytic charg. transfer *** co-catalyst,sample N-C-TiO2/CArW demonstrates H_(2) evolution activity of 426 μmol g.at-1h^(-1),which remarkably outperforms commercial TiO_(2)(P-25) and N-C-TiO_(2)/CAr with a 5-fold and 3-fold H_(2) g.neration,*** study clearly shows that water vapour atmosphere during.the pyrolysis increases the hydrophilicity of the Ti-MOF derived composites by functionalizing.porous carbon matrix with carboxylic g.oups,as well as enhancing.the electrical conductivity and charg. transfer efficiency due to the formation of additional localized oxyg.n-rich N like interstitial/intraband *** work also demonstrates that by optimizing.the anatase-rutile phase composition of the TiO2 polymorphs,tuning.the energ. band g.ps by N/C co-doping.and functionalizing.the porous carbon matrix in the N-C-TiO2/C nanocomposites,the photocatalytic H_(2) g.neration activity can be further enhanced.

Confirmatory Methods, or Hug. Samples, Are Required to Obtain Power for the Evaluation of Theories

Experimental studies are usually desig.ed with specific expectations about the results in mind. However, most researchers apply some form of omnibus test to test for any differences, with follow up tests like pairwise comparisons or simple effects analyses for further investig.tion of the effects. The power to find full support for the theory with such an exploratory approach which is usually based on multiple testing.is, however, rather disappointing. With the simulations in this paper we showed that many of the common choices in hypothesis testing.led to a severely underpowered form of theory evaluation. Furthermore, some less commonly used approaches were presented and a comparison of results in terms of power to find support for the theory was made. We concluded that confirmatory methods are required in the context of theory evaluation and that the scientific literature would benefit from a clearer distinction between confirmatory and exploratory finding.. Also, we emphasis the importance of reporting.all tests, sig.ificant or not, including.the appropriate sample statistics like means and standard deviations. Another recommendation is related to the fact that researchers, when they discuss the conclusions of their own study, seem to underestimate the role of sampling.variability. The execution of more replication studies in combination with proper reporting.of all results provides insig.t in between study variability and the amount of chance finding..
