Abnormal subchondral bone remodeling leading to sclerosis is a main feature of osteoarthritis(OA), and osteomodulin(OMD), a proteoglycan involved in extracellular matrix mineralization, is associated with the sclerotic phenotype. However, the functions of OMD remain poorly understood, specifically in vivo. We used Omd knockout and overexpressing male mice and mutant zebrafish to study its roles in bone and cartilage metabolism and in the development of OA. The expression of Omd is deeply correlated with bone and cartilage microarchitectures affecting the bone volume and the onset of subchondral bone sclerosis and spontaneous cartilage lesions. Mechanistically, OMD binds to RANKL and inhibits osteoclastogenesis, thus controlling the balance of bone remodeling. In conclusion, OMD is a key factor in subchondral bone sclerosis associated with OA. It participates in bone and cartilage homeostasis by acting on the regulation of osteoclastogenesis. Targeting OMD may be a promising new and personalized approach for OA.

Marine invasions enter the genomic era： three lessons from the past, and the way forward

The expanding scale and increasing rate of marine biological invasions have been documented since the early 20th century. Besides their global ecological and economic impacts, non-indigenous species (NIS) also have attracted much attention as opportunities to explore important eco-evolutionary processes such as rapid adaptation, long-distance dispersal and range expansion, and secondary contacts between divergent evolutionary lineages. In this context, genetic tools have been extensively used in the past 20 years. Three important issues appear to have emerged from such studies. First, the study of NIS has revealed unexpected cryptic diversity in what had previously been assumed homogeneous entities. Second, there has been surprisingly little evidence of strong founder events accompanying marine introductions, a pattern possibly driven by large propagule loads. Third, the evolutionary processes leading to successful invasion have been difficult to ascertain due to faint genetic signals. Here we explore the potential of novel tools associated with high-throughput sequencing (HTS) to address these still pressing issues. Dramatic increase in the number of loci accessible via HTS has the potential to radically increase the power of analyses aimed at species delineation, exploring the population genomic consequences of range expansions, and examining evolutionary processes such as admixture, introgression, and adaptation. Nevertheless, the value of this new wealth of genomic data will ultimately depend on the ability to couple it with expanded "traditional" efforts, including exhaustive sampling of marine populations over large geographic scales, integrated taxonomic analyses, and population level exploration of quantitative trait differentiation through common-garden and other laboratory experiments.

A Major Role of the MEKK1-MKK1/2-MPK4 Pathway in ROS Signalling

Over the last few years, it has become evident that reactive oxygen species （ROS） signalling plays an important role in various physiological responses, including pathogen defense and stomatal opening/closure. On the other hand, ROS overproduction is detrimental for proper plant growth and development, indicating that the regulation of an appropriate redox balance is essential for plants. ROS homeostasis in plants involves the mitogen-activated protein kinase （MAPK） pathway consisting of the MAPK kinase kinase MEKK1 and the MAPK MPK4. Phenotypic and molecular analysis revealed that the MAPK kinases MKK1 and MKK2 are part of a cascade, regulating ROS and salicylic acid （SA） accumulation. Gene expression analysis shows that of 32 transcription factors reported to be highly responsive to multiple ROS-inducing conditions, 20 are regulated by the MEKK1, predominantly via the MEKK1-MKK1/2-MPK4 pathway. However, MEKK1 also functions on other as yet unknown pathways and part of the MEKK1-dependent MPK4 responses are regulated independently of MKK1 and MKK2. Overall, this analysis emphasizes the central role of this MAPK cascade in oxidative stress signalling, but also indicates the high level of complexity revealed by this signalling network.

Can Automatic Classification Help to Increase Accuracy in Data Collection?

Purpose： The authors aim at testing the performance of a set of machine learning algorithms that could improve the process of data cleaning when building datasets. Design/methodology/approach： The paper is centered on cleaning datasets gathered from publishers and online resources by the use of specific keywords. In this case, we analyzed data from the Web of Science. The accuracy of various forms of automatic classification was tested here in comparison with manual coding in order to determine their usefulness for data collection and cleaning. We assessed the performance of seven supervised classification algorithms （Support Vector Machine （SVM）, Scaled Linear Discriminant Analysis, Lasso and elastic-net regularized generalized linear models, Maximum Entropy, Regression Tree, Boosting, and Random Forest） and analyzed two properties： accuracy and recall. We assessed not only each algorithm individually, but also their combinations through a voting scheme. We also tested the performance of these algorithms with different sizes of training data. When assessing the performance of different combinations, we used an indicator of coverage to account for the agreement and disagreement on classification between algorithms. Findings： We found that the performance of the algorithms used vary with the size of the sample for training. However, for the classification exercise in this paper the best performing algorithms were SVM and Boosting. The combination of these two algorithms achieved a high agreement on coverage and was highly accurate. This combination performs well with a small training dataset （10%）, which may reduce the manual work needed for classification tasks. Research limitations： The dataset gathered has significantly more records related to the topic of interest compared to unrelated topics. This may affect the performance of some algorithms, especially in their identification of unrelated papers. Practical implications： Although the classification achieved by

Endophytic flora plays a vital role in the colonization and survival of host plants, especially in harsh environments, such as arid regions. This flora may, however, contain pathogenic species responsible for various troublesome host diseases. The present study is aimed at investigating the diversity of both cultivable and non-cultivable endophytic fungal floras in the internal tissues(roots and leaves) of Tunisian date palm trees(Phoenix dactylifera). Accordingly, 13 isolates from both root and leaf samples, exhibiting distinct colony morphology, were selected from potato dextrose agar(PDA) medium and identified by a sequence match search wherein their 18S–28S internal transcribed spacer(ITS) sequences were compared to those available in public databases. These findings revealed that the cultivable root and leaf isolates fell into two groups, namely Nectriaceae and Pleosporaceae. Additionally, total DNA from palm roots and leaves was further extracted and ITS fragments were amplified. Restriction fragment length polymorphism(RFLP) analysis of the ITS from 200 fungal clones(leaves: 100; roots: 100) using HaeIII restriction enzyme revealed 13 distinct patterns that were further sequenced and led to the identification of Alternaria, Cladosporium, Davidiella(Cladosporium teleomorph), Pythium, Curvularia, and uncharacterized fungal endophytes. Both approaches confirmed that while the roots were predominantly colonized by Fusaria(members of the Nectriaceae family), the leaves were essentially colonized by Alternaria(members of the Pleosporaceae family). Overall, the findings of the present study constitute, to the authors' knowledge, the first extensive report on the diversity of endophytic fungal flora associated with date palm trees(P. dactylifera).

Introduction of laparoscopic gastrectomy for gastric cancer in a Western tertiary referral centre:A prospective cost analysis during the learning curve

AIM To evaluate the costs of the introduction of a laparoscopic surgery program for gastric cancer in a Western community training hospital and tertiary referral centre for gastric cancer surgery. METHODS All patients who underwent surgery for gastric cancer with curative intent in 2013 and 2014 were prospectively included. Primary outcomes were costs regarding surgery and hospital stay. RESULTS Laparoscopic gastrectomy was used in 52 patients [mean age 68 years(± 9, range 50 to 87) years] and open gastrectomy was used in 25 patients [mean age 70 years(± 10, range 46 to 85)]. Mean costs(in euro's) of surgical instrumentation were significantly higher for laparo-scopic surgery: 2270 ± 670 vs 1181 ± 680 in the open approach(P < 0.001). Costs of theatre use were higher in the laparoscopic group: mean 3818 ± 865 vs 2545± 1268 in the open surgery(P < 0.001). Total costs of hospitalization(i.e., costs of surgery and admission)were not different between laparoscopic and open surgery, 8187 ± 4864 and 6152 ± 2680 respectively(P= 0.729). Mean length of hospital stay was 9 ± 12 d in the laparoscopic group vs 14 ± 14 d in the open group(P= 0.044). CONCLUSION The introduction of laparoscopic gastrectomy for gastric cancer coincided with higher costs for theatre use and surgical instrumentation compared to the open technique. Total costs were not significantly different due to shorter length of stay and less intensive care unit(ICU) admissions and shorter ICU stay in the laparoscopic group.
