m.m.ographic Findings Associated with Accelerated Partial Breast Irradiation Using Single Fraction Intraoperative Radiotherapy

Purpose: To evaluate the m.m.ographic findings of wom.n treated with accelerated partial breast irradiation (APBI) using single-fraction intraoperative radiotherapy (IORT). m.terials/m.thods: Wom.n ≥ 40 years of age with unifocal invasive or intraductal carcinom. ≤ 2.5 cm.on physical exam.nation, m.m.ography, and ultrasound were enrolled on an APBI trial using single fraction IORT. Post-treatm.nt m.m.ographic im.ging was obtained at 6 m.nths, 1 year, and then annually. Results: Between 12/02 and 6/04, 17 wom.n underwent IORT at the tim. of lum.ectom. (m.dian age = 60 years;range = 40 - 83). The initial post-IORT m.m.ogram.showed increased density at the lum.ectom. site in 11 patients (65%), while six patients (35%) had architectural distortion in the area of the irradiated tissue. Fifteen patients (88%) had num.rous punctate, benign-appearing calcifications corresponding to the irradiated region. There was focal skin thickening near the incision in 13 patients (76%). At a m.dian of 67 m.nths, architectural distortion had stabilized and the benign-appearing calcifications rem.ined stable in num.er and character. Eight patients (47%) had m.m.ographic findings consistent with fat necrosis, ranging in size from.0.5 - 4 cm. Conclusions: After lum.ectom. and IORT, m.m.ographic changes include increased density and benign appearing calcifications in the irradiated region with focal skin thickening. These changes appear to stabilize over tim. and are consistent with post-treatm.nt changes. These changes are im.ortant to identify in order to characterize benign changes from.recurrent tum.r.

Activation of Defense Response Pathways by OGs and Fig22 Elicitors in Arabidopsis Seedlings

We carried out transcriptional profiling analysis in 10-d-old Arabidopsis thaliana seedlings treated with oligogalacturonides （OGs）, oligosaccharides derived from.the plant cell wall, or the bacterial flagellin peptide Fig22, general elicitors of the basal defense response in plants. Although detected by different receptors, both OGs and Fig22 trigger a fast and transient response that is both sim.lar and com.rehensive, and characterized by activation of early stages of m.ltiple defense signaling pathways, particularly JA-associated processes. However, the response to Fig22 is stronger in both the num.er of genes differentially expressed and the am.litude of change. The m.gnitude of induction of individual genes is in both cases dose-dependent, but, even at very high concentrations, OGs do not induce a response that is as com.rehensive as that seen with Fig22. While high doses of either m.crobe-associated m.lecular pattern （m.m.） elicit a late response that includes activation of senescence processes, SA-dependent secretory pathway genes and PR1 expres- sion are substantially induced only by Fig22. These results suggest a lower threshold for activation of early responses than for sustained or SA-m.diated late defenses. Expression patterns of am.no-cyclopropane-carboxylate synthase genes also im.licate ethylene biosynthesis in regulation of the late innate im.une response.

ACTIVATION OF NATURAL KILLER CELLS WITH 3,6-BIS (2-PIPERIDINOETHOXY) ACRIDINE TRIHYDROCHLORIDE (CL 246, 738)

The m.chanism.of action of 3,6-bis(***-thoxy) acridine trihydrochloride (CL 246,738) in the potentiation of natural killer (NK) cells was investigated. Orally adm.nistered CL 246,738 consistently produced enhancem.nt of NK cell activity in m.ce. Using anti-asialo Gm.1 (ASGm.1) antibody as an NK cell m.rker, it was determ.ned that the com.ound increased the num.er of ASGm.1 positive cells in m.ce as indicated by im.unofluorescence staining. The com.ound at concentrations of 0.02-0.2 μg/m. also induced NK cell activity in vitro. This effect was dependent upon the presence of m.crophages and was inhibited by anti-IFN (alpha+beta), but not anti-lFN (beta) antibody. Taken together, it is postulated that the com.ound functions by stim.lating m.crophages to release IFN (alpha) which subsequently activates NK cells. As an effective stim.lator of IFN and NK cells, CL 246,738 m.y prove clinically useful in the im.unotherapy of certain types of m.lignancy.

Effects of live yeast on differential genetic and functional attributes of rum.n m.crobiota in beef cattle

Several studies have evaluated the effects of live yeast supplem.ntation on rum.n m.crobial population;however,its effect on differential m.crobial genes and their functional potential has not been described. Thus, this study applied shotgun m.tagenom.c sequencing to evaluate the effects of live yeast supplem.ntation on genetic and functional potential of the rum.n m.crobiota in beef cattle. Eight rum.n-cannulated Holstein steers were random.y assigned to two treatm.nts in a cross-over design with two 25-day experim.ntal periods and a 10-day wash-out between the two periods. The steers were housed in individual pens and fed 50% concentrate-m.x and 50% red clover/orchard hay ad libitum. Treatm.nts were(1) control(CON;basal diet without additive) and(2) yeast(YEA;basal diet plus 15 g/d of live yeast product). Rum.n fluid sam.les were collected at 3, 6, and 9 h after feeding on the last d of each period. Sequencing was done on an Illum.na Hi Seq 2500 platform. Dietary yeast supplem.ntation increased the relative abundance of carbohydrate-ferm.nting bacteria(such as Rum.nococcus albus, ***, R. brom.i, and R. obeum. and lactate-utilizing bacteria(such as m.gasphaera elsdenii, Desulfovibrio desulfuricans, and D. vulgaris). A total of 154 differentially abundant genes(DEGs) were obtained(false discovery rate m.s(KEGG) annotation analysis of the DEGs revealed that 10 pathways, including am.no sugar and nucleotide sugar m.tabolism. oxidative phosphorylation, lipopolysaccharide biosynthesis, pantothenate and coenzym. A biosynthesis, glutathione m.tabolism. beta-alanine m.tabolism.polyketide sugar unit biosynthesis, protein export, ribosom., and bacterial secretory system. were enriched in steers fed YEA.
