Objective:To evaluate the potential prophylactic activity of traditional polyherbal remedy against ***:A traditional polyherbal remedy against malaria from Odisha,India was evaluated for its potential prophylactic activity using in vitro hepatic cell lines assay and the murine malaria system Plasmodium yoelii yoelii/Anopheles ***:The polyherbal extract inhibited the Plasmodium yoelii hepatic stages in vitro(IC500.74 mg/m L),a therapeutic index of *** mice treated with the aqueous extract(2 000 mg/kg/day),peak parasitaemia values were 81%lower in the experimental2.35%±0.14%as compared to controls 12.62%±0.52%(P<0.001),suggesting significant prophylactic ***:The observations provide a proof of concept for a traditional malaria prophylactic remedy used by tribal populations in India.

Background： Plasmodium falciparum malaria remains a major life-threatening disease. Recently, the Plasmodium apoptosis-linked pathogenicity factors （PALPF） have been identified. These antigens PALPF are expressed only by P falciparum-infected erythrocytes triggering endothelial cell apoptosis （apoptogenic）. Methods： We designed ten synthetic peptides （PI to P10） from PALPF： PF07 0032, PF10_0226, PFI0130c, PFD0875c and MAL13P1.206, and analyzed their antigenicity with an ELISA method using plasma samples from subjects living in Dienga, Gabon. Results： Four peptides showed good reactivity with human antibodies. The prevalence rate of specific IgG was 61%, 51%, 44% and 34% for P5, P6, P4 and P2, respectively. The median optical density of total IgG anti-P2 was higher than that directed against P4 and P6 （P = 0.009; P = 0.012 respectively）. The prevalence rate oflgG subclasses determined with plasma samples recognizing peptide 5 for IgGl, 2, 3 and 4 isotypes was 69%, 45%, 76% and 62%, respectively. All the subjects had at least one immunoglobulin subclass, while 13 （44%） had both IgG1 and IgG3 antibodies. There was no significant difference in the prevalence rate of anti-P5 IgG1, IgG3 and IgG4. Conclusion： These results warrant further immunogenicity studies of peptides 2, 4, 5 and 6 with a view of a tentative to antimalarial vaccine development.

Preferences for pre-exposure prophylaxis among U.S. military men who have sex with men:results of an adaptive choice based conjoint analysis study

Background: Pre-exposure prophylaxis(PrEP) prevents human immunodeficiency virus(HIV) infection, but its use remains low among U.S. military men who have sex with men(MSM), likely due to mis-matching with personal preferences. We conducted a study to characterize preferences to PrEP measures within this ***: HIV-negative military MSM were recruited through a closed, Lesbian, gay, Bisexual, and Transgendered(LGBT) military social media group. The survey was anonymous, and consisted of five experimentally varied attributes in service delivery: dosing method, provider type, visit location, lab work evaluation location, and dispensing *** importance and part-worth utility scores were generated using hierarchical bayes(HB) estimation, and the randomized first choice model was used to examine participation interest across eight possible PrEP program ***: A total of 429 participants completed the survey. Among the eight scenarios with varying attributes, the most preferred scenario featured a daily tablet, PrEP injection or implant, along with a military provider, smartphone/telehealth visit, and on-base locations for lab evaluation and medication pick-up. The results also emphasized the importance for providers to be familiar with PrEP prescription knowledge, and to provide interactions sensitive to sexual identity and mental ***: A PrEP program consisting of daily tablet is preferred in military healthcare settings is preferred. Longacting implants and injections are also desired.
