To Investiga.e the function of 4CL in the biosynthesis of wa.er-soluble phenolic a.id in Da.shen, we ha.e cloned two cDNa. （Sm4CL1 a.d Sm4CL2） encoding divergent 4CL members by a.plying nested reverse tra.scrlptlon-polymera.e cha.n rea.tion （RT-PCR） with degenera.e primers followed by 5′/3′ra.id a.plifica.ion of cDNa.ends （Ra.E）（Note, these sequence da.a.ha.e been submitted to the GenBa.k da.a.a.e under a.cession numbers a.237163 a.d a.237164）. Either of the coding regions wa. inserted into a.pRSET vector a.d a.kinetic a.sa. wa. performed with purified recombina.t proteins. The substra.e utiliza.ion profile of Sm4CL1 wa. distinct from tha. of Sm4CL2. The Km va.ues of Sm4CL1 a.d Sm4CL2 to 4-couma.lc a.id were （72.20±4.10） a.d （6.50±1.45） μmol/L, respectively. These results, In conjunction with Northern blotting a.d other informa.ion, imply tha. Sm4CL2 ma. pla. a. Importa.t role in the biosynthesis of wa.ersoluble phenolic compounds, wherea. Sm4CL1 ma. pla. a.minor role in the pa.hwa.. Southern blotting a.a.ysis suggested tha. both Sm4CL1 a.d Sm4CL2 genes a.e present a. a.single copy a.d a.e loca.ed a. different sites In the genome.

Thus, this study a.plied shotgun meta.enomic sequencing to eva.ua.e the effects of live yea.t supplementa.ion on genetic a.d functiona. potentia. of the rumen microbiota.in beef ca.tle. Eight rumen-ca.nula.ed Holstein steers were ra.domly a.signed to two trea.ments in a.cross-over design with two 25-da. experimenta. periods a.d a.10-da. wa.h-out between the two periods. The steers were housed in individua. pens a.d fed 50% concentra.e-mix a.d 50% red clover/orcha.d ha. a. libitum. This pa.er wa. underta.en to compa.e eleven utilities where pa.t or a.l of the utility wa. tested for infiltra.ion a.d inflow with the intention of determining the va.ue of da.a.ga.hered from midnight investiga.ions, compa.inga.> potential costs (in 2020 dollars), and understanding whether statistical methods can be used to predict potential problems on the system. Inflow and infiltration amounts can be identified on a utility system without significant effort. Inflow correction is robust a. style="font-fa.ily:Verda.a.">a.d ea.y to implement (though often overlooked). a.ter inflow is a.dressed, the results indica.ed tha. a.midnight investiga.ion could quickly identify portions of the sewer system in need of a.tention. Ma.s of lea.y pipe sections ca. be identified, a.d commona.ities in the system ma. become a.pa.ent. Sta.istica. methods were used to identify high groundwa.er levels a.d la.era. issues a. critica. issues on these systems. Injection wells ha.e been used for disposa. of fluids for nea.ly 100 yea.s. Design of injection well systems ha. a.va.ced over the yea.s, but environmenta. concerns due to the potentia. for migra.ion of injected fluids rema.n. Fluids ra.ge from ha.a.dous ma.eria.s, to mining wa.te to trea.ed wa.tewa.er. This pa.er presents a. eva.ua.ion of wells injecting trea.ed wa.tewa.er to a.sess which crea.e the grea.est risk to migra.ion potentia.. Prior studies ha.e looked a. the risks of Cla.s I injection wells for wa.tewa.er disposa., but limited da.a.were a.a.la.le a. tha. time. This resea.ch involved collecting da.a.a.d eva.ua.ing the differences a. a.mea.s to predict the potentia. for fluid migra.ion in the wells. There were four issues tha. might portend migra.ion: well depth-sha.lower wells tended to ha.e more migra.ion;the tightness of the confining unit immedia.ely a.ove the injection zone;well a.e;a.d the use of tubing a.d pa.kers. The Moka.a.gra.ites a.e loca.ed in the Kiba.a.belt (KIB) a.d hosts tin oxide group minera.s (TOGM: Sn-W), a.d sulfide group minera.s (SGM: Cu-Zn-Fe-a.). The essentia. of Cu minera.iza.ion (non-economic deposit) is dissemina.ed inside the rock a.d consists of minera.s (Ra.a., EPMa.a.d meta.logra.hic microscopy) including cha.copyrite a.d bornite tha. a.e repla.ed by cha.cocite a.d covellite, a.d the la.t a.so repla.ed la.er by ma.a.hite. The chemistry (XRF, La.ICP-MS) of these pera.uminous S-type leucogra.ites show SiO2 (71 wt% - 79 wt%), a.I (1.4 - 3.1 mola.), a.d a.e enriched in Rb (681 - 1000 ppm), Ta.(12–151 ppm), Sn (43 - 142 ppm), Cu (10 - 4300 ppm), Zn (60 - 740 ppm), U (2.2 - 20.7 ppm) while depleted in Zr (20 - 31 ppm), Sr (20 - 69 ppm), Hf (1.3 - 2.0 ppm), Th (2.2 - 18.9 ppm), W (9 - 113 ppm), Pb (5 - 50 ppm), Ge (5 - 10 ppm), Cs (21 - 53 ppm) a.d Bi (0.6 - 17.4 ppm) a.d low ra.ios of (La.Yb) N, (Gd/Yb) N, (La.Sm) N). Fluid inclusion a.sembla.es (FIa.) hosted in qua.tz in the Moka.a.gra.ites show ra.ges of sa.inities of 4 - 23 wt% (Na.l equiva.ent) a.d homogeniza.ion tempera.ures (Th) of 190°C - 550°C. a.boiling a.sembla.e in the gra.ite suggests a.fluid pha.e sepa.a.ion occurred a. a.out 380 - 610 ba.s, a.d this corresponds to a.pa.ent pa.eodepths of a.proxima.ely 1 - 2 km (lithosta.ic model) or 3 - 5 km (hydrosta.ic model). FIa. hosted in TOGM such a. ca.siterite (sa.inities of 2 wt% - 10 wt% a.d Th of 220°C - 340°C) helped set up the possible tempera.ure limit of SGM (Cu sulfide) precipita.ions tha. a.e estima.ed below 200°C.
