Variations in zonal fruit starch concentrations of apples – a developmental phenomenon or an indication of ripening?

MEF13 Requires MORF3 and MORF8 for RNA Editing at Eight Targets in Mitochondrial mRNAs in Arabidopsis thaliana

RNA editing sites in plant mitochondria and plastids are addressed by pentatricopeptide repeat （PPR） proteins with E or E and DYW domains, which recognize a specific nucleotide motif upstream of the edited nucleotide. In addition, some sites require MORF proteins for efficient RNA editing. Here, we assign the novel E domain-containing PPR protein, MEF13, as being required for editing at eight sites in Arabidopsis thaliana. A SNP in ecotype c24 altering the editing level at only one of the eight target sites was located by genomic mapping. An EMS mutant allele of the gene for MEF13 was identified in a SNaPshot screen of a mutated plant population. At all eight target sites of MEF13, editing levels are reduced in both morf3 and morf8 mutants, but at only one site in morfl mutants, suggesting that specific MEF13-MORF interactions are required. Yeast two-hybrid analyses detect solid connections of MEF13 with MORF1 and weak contact with MORF3 proteins. Yeast three-hybrid （Y3H） analysis shows that the presence of MORF8 enhances the connection between MEF13 and MORF3, suggesting that a MORF3-MORF8 heteromer may form stably or transiently to establish interaction with MEF13.

Vaccination with dendritic cells pulsed with hepatitis c pseudo particles induces specific immune responses in mice

AIM: To explore dendritic cells (Dcs) multiple functions in immune modulation. METHODS: We used bone-marrow derived dendritic cells from BALB/c mice pulsed with pseudo particles from the hepatitis c virus to vaccinate naive BALB/c mice. Hepatitis c virus (HcV) pseudo particles consist of the genotype 1b derived envelope proteins E1 and E2, covering a non-HcV core structure. Thus, not a single epitope, but the whole "viral surface" induces immunogenicity. For vaccination, mature and activated Dc were injected subcutaneously twice. RESULTS: Humoral and cellular immune responses measured by enzyme-linked immunosorbent assay and interferon-gamma enzyme-linked immunosorbent spot test showed antibody production as well as T-cellsdirected against HcV. Furthermore, T-cell responses confi rmed two highly immunogenic regions in E1 and E2 outside the hypervariable region 1. cONcLUSION: Our results indicate dendritic cells as a promising vaccination model for HcV infection that should be evaluated further.

The Effects of Using a Specially Designed Stirrup on Kinetic Energy Absorption by the Knee Joint of 12 Show Jumping/Eventing Riders

The use of Winderen Knee Protection Solution stirrups compared to standard iron stirrups, reveals the following benefits: 1) A reduction of stress or strain time in the order of 14 seconds per minute of activity whilst walking and 5 - 7 seconds less whilst trotting or cantering for muscles around the knee. 2) A reduction of stress or strain time in the order of 25 seconds per minute of activity whilst walking and 9 - 10 seconds less whilst trotting or cantering for ligaments around the knee. 3) A significant improvement in the E-score (less time exposed to stress and shock) and ST-score (lower force around the knee) whilst walking. 4) A considerable improvement in rider comfort and feeling of leg stability (self-assessment) compared with the owners current stirrups, whilst riding.

Application of Design of Experiments for AlloyDevelopment of an Aluminum copper casting Alloy

Design of experiments (DoE) based on a linear regression model was used to develop an Aluminum copper-based casting alloy. The main objectives of the development were the achievement of (1) a high strength at elevated temperatures with (2) a low hot tearing tendency. Within the DoE, 17 different chemical compositions of the newly developed alloy AlcuMnco(Ni) were cast, tested regarding hot tearing tendency and characterized in tensile tests up to 300 °c. Test results showed that the AlcuMnco(Ni)-alloys from the DoE have high mechanical properties from ambient temperature up to 300 °c and thus feature a high thermal stability. It was found that the alloying elements cu and co increase the yield strength whereas Mn and Ni tend to increase the attainable elongation. Furthermore, some of the alloys showed no or a very low tendency to hot tearing a remarkable feature for Al-cu alloys which are otherwise highly susceptible to hot tearing. The regression model that was developed from the test results fulfils a set of quality criteria and is therefore expected to provide reliable predictions. The predictive ability of the model was validated by casting and testing a sweet spot alloy. Results show that the model is sufficient for predicting the mechanical properties from ambient temperature to 250 °c. Furthermore, the sweet spot alloy surpasses the reference alloy AlcuNicoSbZr (RR30) in its mechanical properties up to 250 °c. It was shown that by applying design of experiments, time and effort for an alloy development can effectively be reduced and simultaneously a high degree of information density about the alloying system considered is generated.

Do U/Pb-SHRIMP Dating and Pb Stepwise Leaching (PbSL) Analyses confirm the Lack of Precambrian Basement Outcrops in Thailand?

U/Pb-SHRIMP dating and Pb stepwise leaching (PbSL) experiments on zircons and garnets, respectively have confirmed the lack of Precambrian outcrops within the crystalline basement of Thailand. The obtained data for the high grade metamorphism show Indosinian ages ranging from 225 - 200 Ma as previously suggested for the vast majority of outcrops in NW-Thailand ([1]?and references therein), as well as a small group of ages in the range of 445 Ma in the Lampang Province. Further, the age of a thermal imprint around 60 Ma was confirmed near Surat Thani, Peninsular Thailand, and only a few indications of older ages for the unknown source areas were detected in detrital components.

Development of an in Vitro Assay to Evaluate the Biological Impact of 5G Technology on Human Skin—Shield Effect of a Tannin-Rich Plant Extract

Background: The new 5G telecommunication technology has stirred concerns about potential negative effects on human health by radiofrequency electromagnetic fields. As to whether skin biology can be affected by 5G waves has remained an unsolved challenge despite recent studies dealing with this issue. In particular, a strategy for rational design of an assay allowing to 1) reproducibly evaluate and decipher the 5G effects on skin as well as 2) test the potential protective effects of cosmetic active ingredients, has yet to be found. Here we describe an in vitro model of human normal keratinocytes irradiated by 5G waves and show their impact on two biomarkers of inflammatory stress, i.e. interleukin-1β (IL-1β) and reactive oxygen species (ROS) production. In addition, the capacity of a tannin-rich plant extract to protect against 5G impact is evaluated. Materials and Methods: In the first series of experiments, monolayers of human normal keratinocytes were irradiated or not (control) by 5G waves (3.5 MHz) in an anechoic chamber and were incubated at 37˚c for 24 hours. At the end of the incubation period, extracellular IL-1β and intracellular ROS were quantified using specific ELISA and colorimetric assays, respectively. In the second series of experiments, the effect of an overnight pre-incubation with increasing concentrations of a tannin-rich plant extract was evaluated. Additionally, we studied in a prospective way the expression of a set of 88 genes selected for their relevance to keratinocyte homeostasis, in relation to the 5G challenge as well as the protective effect of a tannin-rich plant extract. Results: 5G waves significantly increased IL-1β production by 48.4% (p β and ROS production.
