Involvement and therapeutic implications of airway epithelial barrier dysfunction in type 2 inflammation of asthma

Type 2 inflammation is a complex immune response and primary mechanism for several common allergic diseases including allergic rhinitis,allergic asthma,atopic dermatitis,and chronic rhinosinusitis with nasal polyps.It is the predominant type of immune response against helminths to prevent their tissue infiltration and induce their expulsion.Recent studies suggest that epithelial barrier dysfunction contributes to the development of type 2 inflammation in asthma,which may partly explain the increasing prevalence of asthma in China and around the globe.The epithelial barrier hypothesis has recently been proposed and has received great interest from the scientific community.The development of leaky epithelial barriers leads to microbial dysbiosis and the translocation of bacteria to inter-and sub-epithelial areas and the development of epithelial tissue inflammation.Accordingly,preventing the impairment and promoting the restoration of a deteriorated airway epithelial barrier represents a promising strategy for the treatment of asthma.This review introduces the interaction between type 2 inflammation and the airway epithelial barrier in asthma,the structure and molecular composition of the airway epithelial barrier,and the assessment of epithelial barrier integrity.The role of airway epithelial barrier disruption in the pathogenesis of asthma will be discussed.In addition,the possible mechanisms underlying the airway epithelial barrier dysfunction induced by allergens and environmental pollutants,and current treatments to restore the airway epithelial barrier are reviewed.

【Objective】 The study was aimed to assess the role of polymorphisms of HTT-LPR and COMT genes in the occurrence of IBS with a glance to the psychoemotional state of children.【Method】 The study was conducted in the gastroenterology department of children clinical hospital № 1 named after B.Y.Reznik(Odessa,Ukraine).There were examined 122 children with IBS at the age of 6 to 17 years.Molecular genetic studies were performed in a specialized laboratory of the Center of Molecular and Genetic Medicine of ONMedU.The samples of genetic material were taken by using non-invasive methods.DNA was extracted using the bisulfate method.Genotyping was performed by PCR using specific oligonucleotide primers.The characteristics of mental and emotional state of children were tested by the questionnaire of "Profile of psychosomatic orientation".Statistical analysis was performed using nonparametric methods with software Statistics 8.0(StatSoft Inc.,USA).【Result】 There was demonstrated that the frequency of homozygote variant of СОМТ*2 polymorphism was 14.8%,and by 5-HTTLPR polymorphism-9.8%.Children suffering IBS has these genotypes in every third case whereas the severity anxiety is correlated with the presence of these functional polymorphisms.The presence of mutant alleles in the homozygous state is closely correlated with the severity of anxiety in children suffering IBS(r =0.84 P <0.01).【Conclusion】 IBS occurrence and anxiety severity is correlated with the presence of functional polymorphisms of HTT-LPR and COMT genes.

Objective:Although immunity to malaria is reduced in pregnancy,the maternal immune system still continues to respond to malaria infection by the production of antibodies.IgG has been reported to play significant role in immune response against P.falciparum.Anti-MSP-119 antibody and reactive oxygen species have been shown to be protective against malaria infection in children.This work assessed the response of anti-MSP-119 antibody（a promising blood stage vaccine candidate antigen） and oxidative stress in 250 pregnant women.Methods: Blood samples were collected in dry and wet seasons.Plasmodium falciparum infection was determined by microscopy, anti-MSP-119 IgG level was investigated using ELISA.Malondiadelhyde（MDA） and reduced glutathione （GSH） were used as indicators of oxidative stress and they were quantified spectrophotometrically.Results: Parasitaemia was significantly higher（Pdity.There was a significant increase（P19 IgG and MDA levels in the dry than wet season.Anti-MSP-119 IgG and MDA levels were significantly higher in P.falciparum positive primigravidae than P.falciparum negative primigravidae in both wet and dry seasons.In wet season anti-MSP-119 IgG level was significantly increased（P19 IgG and MDA were significant higher in P.falciparum positive multigravidae than primigravidae. Reduced glutathione（GSH） level was significantly reduced（P<0.05） among malaria positive than malaria negative patients in both seasons.Conclusion:This study suggests that IgG and MDA response were positively associated with the presence of malaria infection.

Marcinków(Lower Silesia, ?nie?nik Masiff,south-western Poland) has been a well-known mining centre since the mid-15 th century. Mineral extraction ended in 1949 with the ores of Pb, Ag, Cu and occasionally Au, U or graphite mined there. The mine is surrounded by metamorphic graphite and mica schist. It is not open to the public and no bats have been recorded living in. The aim of this study was the mycological analysis of the air, the rock surface and the water of the disused ore mine of Marcinków.Additionally, the chemical composition of rocks and water, and microclimate conditions of the adit were determined. Fungi were identified using a combination of phenotypic and molecular methods.Six taxa of fungi were cultured from the indoor air samples of the adit, whereas the outdoor air samples contained only five taxa. On the other hand, nine taxa were isolated from both the rocky walls and the water.The Aspergillus group was present only on the rocks and in the water, whereas Cladosporium herbarum,Dumontinia tuberosa, Epicoccum nigrum, Hirsutella sp., were found exclusively in the air. The mycobiota found in the ore mine were mostly similar to those recorded in other mines and natural caves. However,it should be noted that the internal microclimate and chemical compounds of the rocks.

Burkina Faso(West Africa) is a tropical country with a high incidence of infectious diseases.The uncontrolled use of antibiotics against bacterial pathogens has given rise to the emergence of antibiotic resistance in this country.The aims of this study were;i) to determine the prevalences of the most important pathogenic bacteria isolated in the town of Ouagadougou;ii) to identify the bacterial species which have acquired resistance as a result of antibiotic selection;iii) to compare antibiotic-resistances of Escherichia coli isolated from stool culture in the present study with results obtained in 2002 from strains collected in the same structure in Burkina Faso;iv) to determine the trend of antibiotic resistance in Burkina Faso in order to give local advice on the most appropriate empiric antibiotic therapy.Six thousand two hundred and sixty four samples of blood,stools, urine,sputum,pus and vaginal secretion were collected and analyzed in Saint Camille Medical Center(SCMC) laboratory from May 2001 to May 2006.Out of the 6264 samples tested no pathogen was identified in 1583(25.31%),whilst 4681(74.73%) were positive with the incidence of the microrganisms isolated being as follows;Escherichia coli 1291(27.6%),Staphylococcus aureus 922(19.7%),Salmonella spp 561(12. 0%),Streptococcus spp 499(10.7%),Klebsiella spp 359(7.7%),Shigella spp(6.3%),Acinetobacter spp 266(5.7%) and others 783(16.7%).Among the isolated pathogens,the highest resistance was found to amoxicillin:Proteus spp 95,6%,Escherichia coli 78,2%,Salmonella spp 62,2%,Shigella spp 73,4% and Klebsiella spp 89,9%,followed by resistance to ampicillin and cotrimoxazole.Comparing the prevalence of antibiotic resistance of Escherichia cob’from stool cultures isolated during 1999-2000 to that of 2001-2006, a significant reduction was found which could be due to the improved use of antibiotics in recent years.The reduced antibiotic-resistance observed in pathogens isolated in Burkina Faso during this study as compared to previous data,could be the result of sett

BACKGROUND Three-dimensional(3D) laparoscopic technique has gradually been applied to the treatment of carcinoma in the remnant stomach(CRS), but its clinical efficacy remains controversial.AIM To compare the short-term and long-term results of 3D laparoscopic-assisted gastrectomy(3DLAG) with open gastrectomy(og) for CRS.METHODS The clinical data of patients diagnosed with CRS and admitted to the First Medical Center of Chinese PLA General Hospital from January 2016 to January 2021 were retrospectively collected. A total of 84 patients who met the inclusion and exclusion criteria were enrolled. All their clinical data were collected and a database was established. All patients were treated with 3DLAG or og by experienced surgeons and were divided into two groups based on the different surgical methods mentioned above. By using outpatient and telephone follow-up,we were able to determine postoperative survival and tumor status. The postoperative short-term efficacy and 1-year and 3-year overall survival(OS) rates were compared between the two groups.RESULTS Among 84 patients with CRS, 48 were treated with og and 36 with 3DLAG. All patients successfully completed surgery. There was no significant difference between the two groups in terms of age, gender, body mass index, ASA score,initial disease state(benign or malignant), primary surgical anastomosis method,interval time of carcinogenesis, and tumorigenesis site. Patients in the 3DLAG group experienced less intraoperative blood loss(188.33 ± 191.35 mL vs 305.83 ± 303.66 mL;P =0.045) and smaller incision(10.86 ± 3.18 cm vs 20.06 ± 5.17 cm;P og group. 3DLAGC was a more minimally invasive method. 3DLAGC retrieved significantly more lymph nodes than og(14.0 ± 7.17 vs 10.73 ± 6.82;P = 0.036), whereas the number of positive lymph nodes did not differ between the two groups(1.56 ± 2.84 vs 2.35 ± 5.28;P = 0.413). We present how residual intensity modulation(RIM) affects the performance of a resonator fiber optic gyro(R-Fog) through a sinusoidal wave phase modulation technique. The expression for the R-Fog system's demodulation curve under RIM is obtained. Through numerical simulation with different RIM coefficients and modulation frequencies, we find that a zero deviation is induced by the RIM effect on the demodulation curve, and this zero deviation varies with the RIM coefficient and modulation frequency. The expression for the system error due to this zero deviation is derived. Simulation results show that the RIM-induced error varies with the RIM coefficient and modulation frequency. There also exists optimum values for the RIM coefficient and modulation frequency to totally eliminate the RIM-induced error, and the error increases as the RIM coefficient or modulation frequency deviates from its optimum value; however, in practical situations, these two parameters would not be exactly fixed but fluctuate from their respective optimum values, and a large system error is induced even if there exists a very small deviation of these two critical parameters from their optimum values. Simulation results indicate that the RIM-induced error should be considered when designing and evaluating an R-Fog system.

The present paper deals with a new Miocene tupaiid, Prodendrogale yunnanica gen. et sp. nov., which is the only record of fossil tree shrew outside the India-Pakistan subcontinent. The material was collected in 1983 by wet-sieving techniques from the deposits of Section D (see Qi, 1985) at the hominoid locality of Lufeng. Based on 17 isolated teeth, descriptions and comparison of the new taxon are made. The possible phyletic relations of the Lufeng tupaiid to other known genera of Tupaiidae are discussed in this paper as well.

AIM: To evaluate the radicalness and safety of laparoscopic D2 dissection for gastric cancer. METHODS: Clinicopathological data from 209 patients with gastric cancer, who underwent radical gastrectomy with D2 dissection between January 2007 and February 2011, were analyzed retrospectively. Among these patients, 131 patients underwent laparoscopyassisted gastrectomy (LAG) and 78 underwent open gastrectomy (og). The parameters analyzed included operative time, blood loss, blood transfusion, morbidity, mortality, the number of harvested lymph nodes (HLNs), and pathological stage.RESULTS: There were no significant differences in sex, age, types of radical resection [radical proximal gastrectomy (PG + D2), radical distal gastrectomy (DG + D2) and radical total gastrectomy (TG + D2)], and stages between the LAG and og groups (P > 0.05). Among the two groups, 127 cases (96.9%) and 76 cases (97.4%) had 15 or more HLNs, respectively. The average number of HLNs was 26.1 ± 11.4 in the LAG group and 24.2 ± 9.3 in the og group (P = 0.233). In the same type of radical resection, there were no signifi cant differences in the number of HLNs between the two groups (PG + D2: 21.7 ± 7.5 vs 22.4 ± 9.3; DG + D2: 25.7 ± 11.0 vs 22.3 ± 7.9; TG + D2: 30.9 ± 13.4 vs 29.3 ± 10.4; P > 0.05 for all comparisons). Tumor free margins were obtained in all cases. Compared with og group, the LAG group had signifi cantly less blood loss, but a longer operation time (P dity of the LAG group was 9.9%, which was not signifi cantly different from the og group (7.7%) (P = 0.587). The mortality was zero in both groups. CONCLUSION: Laparoscopic D2 dissection is equivalent to og in the number of HLNs, regardless of tumor location. Thus, this procedure can achieve the same radicalness as og.
