Assessment of oxidative and UV-c treatments for inactivating bacterial biofilms from groundwater wells

Microorganisms are ubiquitous in natural environments and in water supply infrastructure including groundwater wells. Sessile-state microorganisms may build up on well surfaces as biofilms and, if excessive, cause biofouling that reduces well productivity and water quality. conditions can be improved using biocides and other traditional well rehabilitation measures; however, biofilm regrowth is inevitable given the continuous introduction of microorganisms from the surrounding environment. Alternative and less invasive well maintenance approaches are desirable for reducing biofilm densities while also minimizing harmful disinfection-by- products. The primary objective of this research was to evaluate effectiveness of alternative treatments for inacti- vating microorganisms comprising biofilms. A novel approach was designed for in situ growth of biofilms on steel coupons suspended from ＇chandeliers＇. After more than 100 days of in situ growth, biofilms were harvested, sampled, and baseline biofilm densities quantifed through cultivation. Ultraviolet-c （UV-c） and oxidative treatments including hydrogen peroxide （H2O2）, ozone （O3） and mixed oxidants were then applied to the biofilms in laboratory-scale treatments. Microbial inactivation was assessed by comparing treated versus baseline biofilm densities. H2O2 was the most effective treatment, and decreased density below baseline by as much as 3.1 orders of magnitude. Mixed oxidants were effective for the well having a lower density biofilm, decreasing density below baseline by as much as 1.4 orders of magnitude. Disparity in the response to treatment was apparent in the wells despite their spatial proximity and common aquifer source, which suggests that microbiological communities are more heterogeneous than the natural media from which they originate.

Building on the pioneering work of Jean-Marie Andre and working in the laboratory he founded, the authors have developed a code called FT-1D to make Hartree-Fock electronic structure computations for stereoregular polymers using Ewald-type con- vergence acceleration methods. That code also takes full advantage of all line-group symmetries to calculate only the minimal set of two-electron integrals and to optimize the computation of the Fock matrix. The present communication reports a bench- mark study of the FT-1D code using polytetrafluoroethylene （PTFE） as a test case. Our results not only confirm the algorith- mic correctness of the code through agreement with other studies where they are applicable, but also show that the use of con- vergence acceleration enables accurate results to be obtained in situations where other widely-used codes （e.g., PLH and crys- tal） fail. It is also found that full attention to the line-group symmetry of the PTFE polymer leads to an increase of between one and two orders of magnitude in the speed of computation. The new code can therefore be viewed as extending the range of electronic-structure computations for stereoregular polymers beyond the present scope of the successful and valuable code crystal.

Integrins and their potential roles in mammalian pregnancy

Integrins are a highly complex family of receptors that, when expressed on the surface of cells, can mediate reciprocal cell-to-cell and cell-to-extracellular matrix(EcM) interactions leading to assembly of integrin adhesion complexes(IAcs) that initiate many signaling functions both at the membrane and deeper within the cytoplasm to coordinate processes including cell adhesion, migration, proliferation, survival, differentiation, and metabolism. All metazoan organisms possess integrins, and it is generally agreed that integrins were associated with the evolution of multicellularity, being essential for the association of cells with their neighbors and surroundings, during embryonic development and many aspects of cellular and molecular biology. Integrins have important roles in many aspects of embryonic development, normal physiology, and disease processes with a multitude of functions discovered and elucidated for integrins that directly influence many areas of biology and medicine, including mammalian pregnancy, in particular implantation of the blastocyst to the uterine wall, subsequent placentation and conceptus(embryo/fetus and associated placental membranes) development. This review provides a succinct overview of integrin structure, ligand binding, and signaling followed with a concise overview of embryonic development, implantation, and early placentation in pigs, sheep, humans, and mice as an example for rodents. A brief timeline of the initial localization of integrin subunits to the uterine luminal epithelium(LE) and conceptus trophoblast is then presented, followed by sequential summaries of integrin expression and function during gestation in pigs, sheep, humans, and rodents. As appropriate for this journal, summaries of integrin expression and function during gestation in pigs and sheep are in depth, whereas summaries for humans and rodents are brief. Optimising workflow in andrology: a new electronic patient record and database

Aim： To improve workflow and usability by introduction of a new electronic patient record （EPR） and database. Methods： Establishment of an EPR based on open source technology （MySQL database and PI-IP scripting language） in a tertiary care andrology center at a university clinic. Workflow analysis, a benchmark comparing the two systems and a survey for usability and ergonomics were carried out. Results： Workflow optimizations （electronic ordering of laboratory analysis, elimination of transcription steps and automated referral letters） and the decrease in time required for data entry per patient to 71% ± 27%, P 〈 0.05, lead to a workload reduction. The benchmark showed a significant performance increase （highest with starting the respective system： 1.3 ± 0.2 s vs. 11.1 ± 0.2 s, mean ± SD）. In the survey, users rated the new system at least two ranks higher over its predecessor （P 〈 0.01） in all sub-areas. conclusion： With further improvements, today＇s EPR can evolve to substitute paper records, saving time （and possibly costs）, supporting user satisfaction and expanding the basis for scientific evaluation when more data is electronically available. Newly introduced systems should be versatile, adaptable for users, and workflow-oriented to yield the highest benefit. Gaseous emission （N2O, cH4 and NH3） from composting can be an important source of anthropogenic greenhouse gas and air pollution. A laboratory scale orthogonal experiment was conducted to estimate the effects of c/N ratio, aeration rate and initial moisture content on gaseous emission during the composting of pig faeces from chinese Ganqinfen system. The results showed that about 23.9% to 45.6% of total organic carbon （TOc） was lost in the form of cO2 and 0.8% to 7.5% of TOc emitted as cH4. Most of the nitrogen was lost in the form of NH3, which account for 9.6% to 32.4% of initial nitrogen. N20 was also an important way of nitrogen losses and 1.5% to 7.3% of initial total nitrogen was lost as it. Statistic analysis showed that the aeration rate is the most important factor which could affect the NH3 （p = 0.0189）, cH4 （p = 0.0113） and N20 （p = 0.0493） emissions significantly. Higher aeration rates reduce the cH4 emission but increase the NH3 and N20 losses. c/N ratio could affect the NH3 （p = 0.0442） and cH4 （p = 0.0246） emissions significantly, but not the N20. Lower c/N ratio caused higher NH3 and cH4 emissions. The initial moisture content can not influence the gaseous emission significantly. Most treatments were matured after 37 days, except a trial with high moisture content and a low c/N ratio.
