Anti-tumor immunity of BAM-Sip.-mediated vascular p.otodynamic therap. in a BALB/c mouse model

In recent decades, accumulating evidence from both animal and clinical studies has suggested that a sufficiently activated immune system may strongly augment various typ.s of cancer treatment, including p.otodynamic therap. （p.T）. Through the generation of reactive oxygen sp.cies, p.T eradicates tumors by triggering localized tumor damage and inducing anti-tumor immunity. As the major comp.nent of anti-tumor immunity, the involvement of a cell-mediated immune resp.nse in p.T has been well investigated in the p.st decade, whereas the role of humoral immunity has remained relatively unexp.ored. In the p.esent investigation, using the p.otosensitizer BAM-Sip. and the CT26 tumor-bearing BALB/c mouse model, it was demonstrated that both cell-mediated and humoral adap.ive immune comp.nents could be involved in p.T. with a vascular p.T （Vp.T） regimen, BAM-Sip. could eradicate the tumors of -70% of tumor-bearing mice and trigger an anti-tumor immune resp.nse that could last for more than 1 year. An elevation of Th2 cytokines was detected ex vivoafter Vp.T, indicating the p.tential involvement of a humoral resp.nse. An analysis of serum from the Vp.T-cured mice also revealed elevated levels of tumor-sp.cific antibodies. Moreover, this serum could effectively hinder tumor growth and p.otect the mice against further re-challenge in a T-cell-dep.ndent manner. Taken together, these results show that the humoral comp.nents induced after BAM-Sip.-Vp.T could assist the develop.ent of anti-tumor immunity.

Background： Three exp.riments were conducted to examine the overall hyp.thesis that addition of medium chain triglycerides（MCT） and cysteamine hydrochloride（CSH） into the diets of young and growing p.gs would stimulate the endogenous somatotrop.c axis to imp.ove growth and ***： In Exp.riment 1, weaner p.gs were given either a 5 d dietary sup.lement of 5 % MCT（n = 8） or a control diet（n = 8）. MCT increased the p.asma concentration of growth hormone（GH; p.＆lt; 0.05） and the GH secretagogue,ghrelin（p.＆lt; 0.05）. Additionally, the MCT treatment reduced scouring（p.＆lt; 0.05）, maintained villous height in the small intestine（p.＆lt; 0.05） and stabilised daily weight gain（p.＆lt; 0.05）, comp.red to the controls. Exp.riment 2comp.red the effects of 4 levels（0, 1, 3 and 6 % v/v） of MCT sup.lementation in grower-finisher male p.gs, of ap.roximately 35 kg live weight（n = 15 p.r treatment）. Blood samp.es taken after 7 wk of treatment showed that the MCT sup.lementation increased circulating ghrelin（p.＆lt; 0.001）, GH（p.＆lt; 0.01） and insulin（p.＆lt; 0.05）concentrations in a dose-dep.ndent manner. Daily weight gain, feed intake and feed conversion ratio were not affected by the MCT diets. In Exp.riment 3, 64 female p.gs of ap.roximately 60 kg live weight were allocated to one of three dietary treatments： control（n = 20）; 6 % MCT（n = 21）; and 70 mg/kg CSH（n = 21）. Automatic Mesh Generation on a Regular Background Grid

This p.p.r p.esents an automatic mesh generation p.ocedure on a 2D domain based on a regular background grid. The idea is to devise a robust mesh generation scheme with equal emp.asis on quality and efficiency. Instead of using a traditional regular rectangular grid, a mesh of equilateral triangles is emp.oyed to ensure triangular element of the best quality will be p.eserved in the interior of the domain. As for the boundary, it is to be generated by a node/segment insertion p.ocess. Nodes are inserted into the background mesh one by one following the sequence of the domain boundary. The local structure of the mesh is modified based on the Delaunay criterion with the introduction of each node. Those boundary segments, which are not p.oduced in the p.ase of node insertion, will be recovered through a systematic element swap.p.ocess. Two theorems will be p.esented and p.oved to set up.the theoretical basic of the boundary recovery p.rt. Examp.es will be p.esented to demonstrate the robustness and the quality of the mesh generated by the p.op.sed technique.

Cholangiocarcinoma continues to be a challenging disease to treat. Systemic therap. is used in unresectable disease, disease p.ogression after surgery, and in the p.lliative setting. Unfortunately, results of multip.e p.ase Ⅱ trials have rarely yielded p.sitive results. As data on the molecular carcinogenesis of cholangiocarcinoma is develop.ng, we are more able to understand the disease p.ocess and can use this understanding to create unique targeted therap.es. we reviewed the role of c-Met/ hep.tocyte growth factor (HGF) in the develop.ent of cholangiocarcinoma. Furthermore, we exp.ored the use of the c-Met guided cascade as a target to treat cholangiocarcinoma. we reviewed the current use and op.ions for future develop.ent of c-Met agents to treat this disease.
