Effect of Aerobic Exercise on Protein Expression in Muscle of Obese Mexican Adolescents: A Proteomic and Bioinformatic Analysis

The beneficial effects of exercise have been recognized for many years yet the molecular mechanisms by which exercise show benefits on health are still elusive. A combination of experimental and bioinformatics approaches can be an invaluable tool to increase the cellular understanding of the molecular mechanisms that underlie the aerobic exercise in obese adolescents. Muscle skeletal proteins were separated by 2d-PAGE and changes in protein expression were revealed by ImageMaster 2d Platinum analysis software. Proteins with expression changes after aerobic exercise were identified by comparison with 2d maps from SWISS-2dPAGE and interactions of proteins were analyzed with STRING and dAVId databases. After aerobic exercise, all participants decreased glucose, insulin, total cholesterol, body mass index and waist circumference. Proteomic differential analysis revealed 10 overexpressed proteins after aerobic training, which correspond to carbonic anhydrase III, beta enolase, creatine kinase, ATP synthase beta subunit, aldolase A, glyceraldehyde-3-phosphate dehydrogenase, triosephosphate isomerase, pyruvate kinase, lactate dehydrogenase and adenylate kinase. Exercise increased glycolysis and oxidative phosphorylation pathways, which is likely to be regulated by ubiquitin c protein. In conclusion, our analysis suggested that obese adolescents show changes in their body mass index, waist circumference and serum glucose after aerobic exercise through improved protein expression patterns that aid, especially, glucose metabolism and increase the oxidative phosphorylation. The most likely explanation for the observed pattern is that ubiquitin c protein keeps the glycolytic enzymes bound within skeletal muscle, obtaining a non-degradative role in cell signaling.

Childhood obesity seems to be increasing at a disturbing rate. Exercise and diet are recommended to combat this epidemic. Light and moderate physical activity is associated with health benefits, whereas intense physical activity is associated with normal BMI and low cardiovascular risk. The aim of this study was to assess the metabolic response to a graded exercise test (GXT) in children with different body weight. Eight boys and nine girls underwent a GXT on a treadmill according to Blake protocol. These children were 7-9 years old. The participants were divided into three groups according to their body weight: normal-weight, overweight, and obese. After GXT, lactate and HdL levels increased significantly in all three groups

Compost Input Effect on dryland Wheat and Forage Yields and Soil Quality

Organic agricultural systems rely on organic amendments to achieve crop fertility requirements, and weed control must be achieved without synthetic herbicides. Our obj.ctive was to determine the crop yield and soil quality as affected by a transition from grass to dryland organic agriculture in the Central Great Plains of North America. This study evaluated three beef feedlot compost(BFC)treatments in 2010–2015 following biennial application rates: 0(control), 22.9, and 108.7 t ha^(-1) on two dryland organic cropping systems: a wheat(Triticum aestivum)-fallow(WF) rotation harvested for grain and a triticale(Triticosecale)/pea(Pisum sativum)-fallow(T/P-F) rotation harvested for forage. The triticale + pea biomass responded positively to the 108.7-t ha^(-1) BFC treatment,but not the 22.9-t ha^(-1) BFC treatment. The wheat biomass was not affected by BFC addition, but biomass N content *** feedlot compost input did not increase wheat grain yields, but had a positive effect on wheat grain Zn content. Soil total C and N contents increased with the rate of 108.7 t ha^(-1) BFC after three applications, but not with 22.9 t ha^(-1) BFC. Soil enzyme activities associated with N and C cycling responded positively to the 108.7-t ha^(-1) BFC treatment. Saturated salts were high in the soil receiving 108.7 t ha^(-1) of BFC, but did not affect crop yields. These results showed that BFC was effective in enhancing forage yields, wheat grain quality, and soil C and N, as well as specific microbial enzymes important for nutrient cycling. However, the large rates of BFC necessary to elicit these positive responses did not increase grain yields, and resulted in an excessive buildup of soil P.

Multi-obj.ctive optimization design of bridge piers with hybrid heuristic algorithms

This paper describes one approach to the design of reinforced concrete (RC) bridge piers, using a three-hybrid multi- obj.ctive simulated annealing (SA) algorithm with a neighborhood move based on the mutation operator from the genetic algorithms (GAs), namely MOSAMO1, MOSAMO2 and MOSAMO3. The procedure is applied to three obj.ctive functions: the economic cost, the reinforcing steel congestion and the embedded CO 2 emissions. Additional results for a random walk and a descent local search multi-obj.ctive algorithm are presented. The evaluation of solutions follows the Spanish Code for structural concrete. The methodology was applied to a typical bridge pier of 23.97 m in height. This example involved 110 design variables. Results indicate that algorithm MOSAMO2 outperforms other algorithms regarding the definition of Pareto fronts. Further, the proposed procedure will help structural engineers to enhance their bridge pier designs.

Agglomerate behavior in a recirculating fluidized bed with sheds： Effect of ring baffles

The radioactive particle tracking technique was used to study the effect of internal ring baffles on wet agglomerate motion inside a cold flow recirculating fluidized bed. The study found that using such a baffle on its own or above the regular sheds helps reduce the fouling of the stripper section by increasing the residence time that the agglomerates spend above the baffle, thereby reducing the release of the vapors below the baffles that cause fouling of the sheds. Adding down-comers, or flux tubes, to the ring baffles degrades the performance of the baffles, Reducing the length of the flux tubes, so that they do not reach the bottom of the baffle lip results in a further degradation in baffle performance.

Photosynthetic Regulation of the Cyanobacterium Synechocystis sp. PCC 6803 Thioredoxin System and Functional Analysis of TrxB （Trx x） and TrxQ （Trx y） Thioredoxins

The expression of the genes encoding the ferredoxin-thioredoxin system including the ferredoxin-thioredoxin reductase （FTR） genes ftrC and ftrV and the four different thioredoxin genes trxA （m-type; sir0623）, trxB （x-type; sir1139）, trxC （sll1057） and trxQ （y-type; sir0233） of the cyanobacterium Synechocystis sp. PCC 6803 has been studied according to changes in the photosynthetic conditions. Experiments of light-dark transition indicate that the expression of all these genes except trxQ decreases in the dark in the absence of glucose in the growth medium. The use of two electron transport inhibitors, 3-（3,4-dichlorophenyl）-1,1-dimethylurea （dCMU） and 2,5-dibromo-3-methyl-6-isopropyl-p- benzoquinone （dBMIB）, reveals a differential effect on thioredoxin genes expression being trxC and trxQ almost unaffected, whereas trxA, trxB, and the ftr genes are down-regulated. In the presence of glucose, dCMU does not affect gene expression but dBMIB still does. Analysis of the single TrxB or TrxQ and the double TrxB TrxQ Synechocystis mutant strains reveal different functions for each of these thioredoxins under different growth conditions. Finally, a Synechocystis strain was generated containing a mutated version of TrxB （TrxBC34S）, which was used to identify the potential in-vivo targets of this thioredoxin by a proteomic analysis.

GSTARS computer models and their applications, part I:theoretical development

GSTARS is a series of computer models developed by the U.S. Bureau of Reclamation for alluvial river and reservoir sedimentation studies while the authors were employed by that agency. The first version of GSTARS was released in 1986 using Fortran IV for mainframe computers. GSTARS 2.0 was released in 1998 for personal computer application with most of the code in the original GSTARS revised, improved, and expanded using Fortran IV/77. GSTARS 2.1 is an improved and revised GSTARS 2.0 with graphical user interface. The unique features of all GSTARS models are the conj.nctive use of the stream tube concept and of the minimum stream power theory. The application of minimum stream power theory allows the determination of optimum channel geometry with variable channel width and cross-sectional shape. The use of the stream tube concept enables the simulation of river hydraulics using one-dimensional numerical solutions to obtain a semi-two-dimensional presentation of the hydraulic conditions along and across an alluvial channel. According to the stream tube concept, no water or sediment particles can cross the walls of stream tubes, which is valid for many natural rivers. At and near sharp bends, however, sediment particles may cross the boundaries of stream tubes. GSTARS3, based on FORTRAN 90/95, addresses this phenomenon and further expands the capabilities of GSTARS 2.1 for cohesive and non-cohesive sediment transport in rivers and reservoirs. This paper presents the concepts, methods, and techniques used to develop the GSTARS series of computer models, especially GSTARS3.
