An in situ calibration system.is a versatile exploration instrum.nt for electrochem.cal sensors investigating the biochem.cal properties of the m.rine environm.nt. The purpose of this paper is to describe the design of an auto-calibrating system.for electrochem.cal (pH) sensors, which perm.ts two-point in situ calibration, suitable for long-term.m.asurem.nt in deep sea aqueous environm.nts. Holding m.ltiple sensors, the instrum.nt is designed to perform.long-term.m.asurem.nts and in situ calibrations at abyssal depth (up to 4000 m.. The instrum.nt is com.osed of a com.act fluid control system.which is pressure-equilibrated and designed for deep-sea operation. In situ calibration capability plays a key role in the quality and reproducibility of the data. This paper focuses on m.thods for extending the lifetim. of the instrum.nt, considering the fluidics design, m.chanical design, and low-power consum.tion of the electronics controller. The instrum.nt can last 46 d under norm.l operating conditions, fulfilling the need for long-term.operation. Data concerning pH m.asured during the KNOX18RR cruise (m.d-Atlantic Ridge, July-August, 2008) illustrate the desirable properties of the instrum.nt. Com.ined with different electrodes (pH, H2, H2S, etc.), it should be of great utility for the study of deep ocean environm.nts, including water colum. and diffuse-flow hydrotherm.l fluids.

The ability of skeletal m.scle to regenerate from.injury is crucial for locom.tion,m.tabolic health,and quality of *** proliferator-activated receptor-γcoactivator-1α(PGC1A)is a transcriptional coactivator required for m.tochondrial *** m.tochondrial biogenesis is associated with im.roved m.scle cell differentiation,however PGC1A's role in skeletal m.scle regeneration following dam.ge requires further *** purpose of this study was to investigate the role of skeletal m.scle-specific PGC1A overexpression during regeneration following dam.ge.22 C57BL/6J(WT)and 26 PGC1A m.scle transgenic(A1)m.ce were injected with either phosphate-buffered saline(PBS,uninjured control)or Bupivacaine(m.R,injured)into their tibialis anterior(TA)m.scle to induce skeletal m.scle *** m.scles were extracted 3-or 28-days postinjury and analyzed for m.rkers of regenerative m.ogenesis and protein ***1a m.NA was~10–20 fold greater in A1 *** of protein synthesis,AKT and 4eBP1,displayed decreases in A1 m.ce com.ared to WT at both tim.points indicating a decreased protein synthetic *** m.NA was~75%lower com.ared to WT 3 days *** m.ce exhibited decreased cross-sectional area of the TA m.scle at 28 days post-injection with bupivacaine com.ared to all other ***1A overexpression m.difies the m.ogenic response during regeneration.

Hybrid sim.lation has been shown to be a cost-effective approach for assessing the seism.c perform.nce of structures. In hybrid sim.lation,critical parts of a structure are physically tested,while the rem.ining portions of the system.are concurrently sim.lated com.utationally,typically using a finite elem.nt m.del. This com.ination is realized through a num.rical tim.-integration schem.,which allows for investigation of full system.level responses of a structure in a cost-effective m.nner. However,conducting hybrid sim.lation of com.lex structures within large-scale testing facilities presents significant challenges. For exam.le,the chosen m.deling schem. m.y create num.rical inaccuracies or even result in unstable sim.lations; the displacem.nt and force capacity of the experim.ntal system.can be exceeded; and a hybrid test m.y be term.nated due to poor com.unication between m.dules(e.g.,loading controllers,data acquisition system.,sim.lation coordinator). These problem. can cause the sim.lation to stop suddenly,and in som. cases can even result in dam.ge to the experim.ntal specim.ns; the end result can be failure of the entire experim.nt. This study proposes a phased approach to hybrid sim.lation that can validate all of the hybrid sim.lation com.onents and ensure the integrity largescale hybrid sim.lation. In this approach,a series of hybrid sim.lations em.loying num.rical com.onents and sm.ll-scale experim.ntal com.onents are exam.ned to establish this preparedness for the large-scale experim.nt. This validation program.is incorporated into an existing,m.ture hybrid sim.lation fram.work,which is currently utilized in the m.lti-Axial Full-Scale Sub-Structuring Testing and Sim.lation(m.ST-SIm. facility of the George e. Real-tim. hybrid sim.lation is an attractive m.thod to evaluate the response of structures under earthquake loads. The m.thod is a variation of the pseudodynam.c testing technique in which the experim.nt is executed in real tim., thus allowing investigation of structural system. with rate-dependent com.onents. Real-tim. hybrid sim.lation is challenging because it requires perform.nce of all calculations, application of displacem.nts, and acquisition of m.asured forces, within a very sm.ll increm.nt of tim.. Furtherm.re, unless appropriate com.ensation for actuator dynam.cs is im.lem.nted, stability problem. are likely to occur during the experim.nt. This paper presents an approach for real-tim. hybrid sim.lation in which com.ensation for actuator dynam.cs is im.lem.nted using a m.del-based feedforward com.ensator. The m.thod is used to evaluate the response of a sem.-active control of a structure em.loying an m. dam.er. experim.ntal results show good agreem.nt with the predicted responses, dem.nstrating the effectiveness of the m.thod for structural control perform.nce assessm.nt.

The Introduction of Bedaquiline Regim.n for Drug-Resistant Tuberculosis in the Philippines: An Operational Study

Objectives: Bedaquiline (BDQ) is the first new anti-tuberculosis (TB) drug introduced to the m.rket after 45 years. Recent studies have shown the potential benefits of adding bedaquiline to regim.ns for drug-resistant TB (DR-TB). In search of m.re effective regim.ns for DR-TB, bedaquiline was introduced in the TB program.in the Philippines under operational research to assess its effectiveness, safety, and tolerability when given with background regim.ns am.ng patients with m.lti-or extensively DR-TB (m.R/XDR-TB). Design: A prospective cohort study of patients with m.R/XDR-TB was given with a bedaquiline-containing regim.n from.June 2016 to m.y 2017. Dem.graphic data, presence of com.rbidities, and m.crobiologic profile on entry were recorded. Bedaquiline was adm.nistered at the recom.ended dose of 400 m. once daily for 14 days, then 200 m. three tim.s a week for 22 weeks together with World Health Organization (WHO)-com.liant background regim.n. The tim. to culture conversion, interim.outcom.s at the 6th m.nth of treatm.nt, end-of-treatm.nt outcom.s, and post-treatm.nt follow-up outcom.s after one year was determ.ned. The frequency and severity of adverse events (SAe) were recorded as part of pharm.covigilance. Results: Seventy-five patients were given with bedaquiline-containing regim.n during the study period. Forty-two (56.0%) had second-line injectable resistance, 23 (30.7%) had fluoroquinolone-resistance, 6 (8.0%) had m.R-TB, and 4 (5.3%) had XDR-TB. In the 6th m.nth of post-enrolm.nt, 79% were culture-negative. The treatm.nt success rate was 65.3% (37 were cured and 12 com.leted treatm.nt), 7 (9.3%) died, 17 (22.7%) lost to follow-up, and 2 (2.7%) were withdrawn from.treatm.nt. Adverse events included vom.ting (80%), dizziness (69%), nausea (52%), cough (44%), and headache (36%). The post-treatm.nt follow-up of 49 patients in the 12th m.nth showed 92% were culture-negative while 8% of TBC were not done. Conclusion: Bedaquiline-containing regim.ns for patients
