The use of magnesium alloys,as a biodegradable medical device,is an interesting challenge for the biomaterials *** rapid degradation and the release of hydrogen,when exposed to biological fluids,are the main drawbacks for clinical *** this work,a coating made of polydopamine(PDOPA),is used as an intermediate layer to decrease the degradation rate of AZ31 magnesium alloy/polymeric coating system,when exposed to Hank’s *** results highlighted:(i)the formation of a thin PDOPA layer,(ii)an increased adhesion in the organic coating/metallic substrate system,(iii)a decrease of two orders of magnitude of the corrosion rate when the PDOPA film is used together with an external organic coating,(iv)the efficacy in the use of PDODA due to the synergistic effect of both,physical and chemical,interactions between the PDOPA layer and the organic coating.

The effect of sand-blasting and hydrofluoric acid etching on Ti CP2 and Ti CP4 surface topography

Titanium is widely used in biomedical applications from many years for its interesting properties, nevertheless there are a large number of researchers studying the way to improve the performances of biomedical devices. A large number of papers concern the study of “commercially pure titanium” (Ti CP) but, very often, no details are indicated regarding: 1) the type of Ti CP used;2) the sandblasting process;and 3) the analytic evaluation of surface topography. In this paper, the effect of sandblasting duration (1, 2, 4, 8 min) and hydrofluoric acid (HF) etching, as well as their combined or synergic effect, on the surface topography of CP grade 2 and grade 4, have been investigated. Results obtained show that sandblasting treatment duration should be different for Ti CP grade 2 and Ti CP grade 4 to achieve similar topography. The shape of the surface is highly modified after acid etching. The latter produces “peaks” when applied to not sand- blasted coupons, whilst cuts the “top of the peaks” in the sandblasted samples. Further we confirm that, the roughness parameter Ra (or Sa), is not sufficient to describe “the surface status” and we propose to use additional para- meters like: Sq, Sku and Ssk, to unequivocally describe surface topography of implants as well as well defined test procedures.

Anionic liposomes for small interfering ribonucleic acid （siRNA） delivery to primary neuronal cells： Evaluation of alpha-synuclein knockdown efficacy

Alpha-synuclein （a-syn） deposition in Lewy bodies （LB） is one of the main neuropathological hallmarks of Parkinson＇s disease （PD）. LB accumulation is considered a causative factor of PD, which suggests that strategies aimed at reducing a-syn levels could be relevant for its treatment. In the present study, we developed novel nanocarriers suitable for systemic delivery of small interfering ribonucleic acid （siRNA） that were specifically designed to reduce neuronal α-syn by RNA interference. Anionic liposomes loaded with an siRNA-protamine complex for α-syn gene silencing and decorated with a rabies virus glycoprotein （RVG）-derived peptide as a targeting agent were prepared. The nanoparticles were characterized for their ability to load, protect, and deliver the functional siRNA to mouse primary hippocampal and cortical neurons as well as their efficiency to induce gene silencing in these cells. Moreover, the nanocarriers were evaluated for their stability in serum. The RVG-decorated liposomes displayed suitable characteristics for future in vivo applications and successfully induced α-syn gene silencing in primary neurons without altering cell viability. Collectively, our results indicate that RVG-decorated liposomes may be an ideal tool for further studies aimed at achieving efficient in vivo α-syn gene silencing in mouse models of PD.
