Age-related macular degeneration(AMD)is characterized by complex interactions between genetic and environmental *** we genotyped the selected 25 single-nucleotide polymorphisms(SNPs)in 983 cases with advanced AMD and 271 cases with intermediate AMD and build an AMD life-risk score model for assessment of progression from intermediate to advanced *** analyzed the performance of the prediction model for geographic atrophy progressors or choroidal neovascularization progressors versus non-progressors based on the 25 SNPs plus body mass index and smoking *** results suggest that a class prediction algorithm can be used for the risk assessment of progression from intermediate to late AMD *** algorithm could also be potentially applied for therapeutic response,and toward personalized care and precision medicine.

Empowered Stakeholders: Female University Students’ Leadership During the COVID-19-Triggered On-campus Evictions in Canada and the United States

Acute glaucoma is a sight-threatening condition characterized by a sudden and substantial rise in intraocular pressure(IOP) and consequent retinal ganglion cell(RGC) *** closure glaucoma,a common cause of glaucoma in Asia that affects tens of millions of people worldwide,often presents acutely with loss of vision,pain,and high *** when medical and surgical treatment is available,acute angle closure glaucoma can cause permanent and irreversible loss of ***-like receptor 4(TLR4) signaling has been previously implicated in the pathogenesis of IOP-induced RGC death,although the underlying mechanisms are largely *** the present study,we used an acute IOP elevation/glaucoma model to investigate the underlying mechanism of RGC *** found that TLR4 leads to increased caspase-8 expression;this elevation increases IL-1β expression and RGC death via a caspase-1–dependent pathway involving Nod-like receptor family,pyrin domain containing 1(NLRP1)/NLRP3 inflammasomes and a caspase-1–independent *** show that inhibition of caspase-8 activation significantly attenuates RGC death by down-regulating the activation of NLRP1 and NLRP3,thus demonstrating the pivotal role of caspase-8 in the TLR4-mediated activation of *** findings demonstrate collectively a critical role of caspase-8 in transducing TLR4-mediated IL-1β production and RGC death and highlight signal transduction in a caspase-1–dependent NLRP1/NLRP3 inflammasome pathway and a caspase-1–independent pathway in acute *** results provide new insight into the pathogenesis of glaucoma and point to a treatment strategy.

The study of disaster-specific leadership of female university students has been largely neglected,especially during on-campus emergency eviction and *** on the COVID-19-triggered,on-campus evictions across Canada and the United States,this cross-national partnership examined the out-of-province/state and international female university students’leadership during the entire eviction *** in-depth interviews,this study revealed the female university students’leadership behaviors during three stages:(1)pre-eviction:their self-preparedness formed an emotional foundation to support others;(2)peri-eviction:their attitude and leadership behavior enabled them to facilitate(psychologically and physically)their peers’eviction process;and(3)post-eviction:they continued to support their peers virtually and raised the general public’s awareness regarding the plight of vulnerable and marginalized *** article argues that the female university students’leadership that emerged during the eviction process became complementary to and even augmented the universities’official efforts and *** leadership represents empirical evidence that contributes to the existing literature on gender and leadership by demonstrating female youth as empowered stakeholders rather than as merely passive *** studies could develop detailed stratification of gender and age dimensions in order to portray a more comprehensive picture of the younger generation’s leadership in hazards and disaster research and practice.

The purpose of this study is to examine the feasibility and short-term impact of a fully manualized, telephone-delivered intervention for spouse caregivers, Taking Care of Her (TCH). A total of 12 study participants from the Pacific NW were enrolled whose wife was diagnosed with Stage III ovarian cancer within 8 months. Feasibility was confirmed by rates of recruitment and retention;the quality of delivery of the intervention by telephone;and through data obtained on program acceptability during follow up exit interviews. Outcomes from the within-group analysis revealed improvements on standardized measures of spouses’ and patients’ depressed mood and anxiety;marital communication about the cancer;caregivers’ skills and confidence to manage the emotional toll of the illness on themselves and wives;and wives’ positive appraisal of spousal support. Study results suggest that the TCH Program has the potential to positively affect both spouse caregiver and patients’ adjustment to recently diagnosed advanced ovarian cancer. Telephone delivery holds promise for sustainability. A future clinical trial with a larger study sample is warranted.

OBJECTIVE In order to reduce animal use and alleviate animal pain in in-life procedures, the potential application for micro-sampling in pharmacokinetic(PK) studies was investigated in *** CD-1 or BALB/c mice were orally or intravenously administered with model compounds at the dose of 1 mg·kg iv or 10 mg·kg *** samples were collected at 0.083(only for iv dosing),0.25,0.5,1,2,4,7 and 24 h post-dose(n =3 per time-point) via the saphenous vein for PK *** micro-sampling method,only 30 pl/sample of blood were collected using capillary tubes(Drummond Scientific).A 150μl/sample of blood was collected using the normal sampling method. The LC/MS/MS system consists of a Shimadzu Nexera UHPLC,RP -18 column at 25℃,and AB SCIEX TRIPLE QUAD 5500.A binary gradient program with a flow rate of 0.8 ml·min was used to analyze the plasma *** volume was 1μ*** and internal standards(0.2μmol·L tolbutamide) were detected by the MS using multiple reaction monitoring in positive electrospray ionization mode(Analyte 1.5.2).RESULTS For each of the five compounds,plasma concentration *** curves and PK parameters(AUC,C,T,clearance,T,bioavailability,etc.) were compared between the micro-sampling and normal sampling *** PK profiles of each compound generated by both methods were well *** addition,compared to the normal sampling methods(e.g. tail-vein bleeding,retro-orbital puncture),micro-sampling via the saphenous vein significantly alleviated pain in *** volume of blood withdrawn was reduced by 80%,and the number of mice required for a PK study was decreased by 65%.CONCLUSION The micro-sampling method can replace normal blood sampling *** this method,the animal welfare will be significantly improved in terms of alleviating animal pain and reducing animal use.

学界通常将美国国会于1906年颁布的《联邦食品与药品法》看作时任总统罗斯福、黑幕揭发者和哈维·维利这样的监管机构官僚共同合作推动的结果。相比之下,以基督教妇女禁酒联合会为代表的妇女改革团体为食品药品纯净运动所做的贡献却未能得到应有重视。事实上,现代意义上的社会运动无法离开女性参与,妇女改革团体的行动代表着一个社会追求进步和向上的努力。对此问题的深入展开要求研究者转换视角,关注女性在社会运动中的诉求,这也将有助于推动对进步时代美国社会的整体认识。
