The cRISPR/cas9 system has been tailored to a revolutionary genetic tool because of its remarkable simplicity and *** complex genome editing in the mouse since the 1990 s has been dominated by the use of embryonic stem（ES） cells,cRISPR/cas9 now offers a versatile and fast approach to precisely modify virtually any DNA regions directly in mouse ***,this relative simplicity does not preclude a conscientious preparatory work that is often neglected when initiating a ***,we describe the key steps leading to successful generation of a double knockout（KO） mouse by simultaneously targeting two homolog genes,Tmem176 a and Tmem176 b,which are located in the same genomic ***,we show that similar efficiency can be obtained in a mixed genetic background or directly in the c57BL/6 inbred ***,presented as a detailed case study that should be helpful to the non-specialists,we focus on the genotyping strategy to anticipate the various possibilities.

Purpose:The purposes of this study were to examine the trajectories of athlete burnout across a 2-month period characterized by high physical,psychological,and social demands to explore(1)whether several subgroups of athletes representing distinct burnout trajectories emerged from the analyses and(2)whether athlete burnout symptoms(reduced accomplishment,sport devaluation,and exhaustion)developed in tandem or whether some burnout dimensions predicted downstream changes in other dimensions(causal ordering model).Methods:One hundred and fifty-nine table tennis players in intensive training centers completed a self-reported athlete burnout measure across 3 time points within a 2-month period characterized by high *** were analyzed through latent class growth ***:Results of latent class growth analysis showed 3 distinct trajectories for each athlete burnout dimension,indicating not only linear or quadratic change but also stability in longitudinal athlete burnout *** also suggested that the 3 dimensions of athlete burnout did not develop in ***,the likelihood of belonging to particular emerging trajectories of sport devaluation and physical/emotional exhaustion was significantly influenced by the athletes’perception of reduced accomplishment assessed at Time ***,reduced accomplishment predicted downstream changes in the 2 other athlete burnout ***:As a whole,these results highlighted that the multinomial heterogeneity in longitudinal athlete burnout symptoms needs to be accounted for in future research.

AIM: To evaluate the efficacy and safety of the FOLFIRI regimen in patients with metastatic pancreatic adenocarcinoma (PAc) after the failure of gemcitabine and platinum salts. METHODS: All consecutive patients with histologically confirmed, metastatic PAc and World Health Organiza-tion performance status (PS) ≤ 2 received FOLFIRI-1 [irinotecan 180 mg/m2 on day 1 and leucovorin 400 mg/m2 followed by 5-fluorouracil (5-FU) 400 mg/m2 bolus, then 5-FU 2400 mg/m2 as a 46-h infusion, biweekly] or FOLFIRI-3 (irinotecan 100 mg/m2 on day 1 and leucovorin 400 mg/m2, then 5-FU 2400 mg/m2 as a 46-h infusion and irinotecan 100 mg/m2 repeated on day 3, biweekly) after failure of gemcitabine and platinum-based chemotherapies as a systematic policy in two institutions between January 2005 and May 2010. Tumor response, time to progression (TTP), overall survival rate (OS) and grade 3-4 toxicities were retrospectively studied. Subgroup analyses were performed to search for prognostic factors. RESULTS: Sixty-three patients (52.4% male, median age 59 years) were analyzed. Among them, 42.9% were PS 0, 38.1% were PS 1 and 19.0% were PS 2. Fifty one patients (81.0%) had liver metastases. Before the FOLFIRI regimen, patients had received 1 line (n = 19), 2 lines (n = 39) or 3 lines (n = 5) of chemotherapy. Median TTP obtained with the line before FOLFIRI was 3.9 mo (95% cI: 3.4-5.3 mo). A total of 480 cycles was completed (median: 6 cycles, range: 1-51 cycles). The main reason for discontinuing FOLFIRI was tumor progression (90.3%). Tumor control was achieved in 25 patients (39.7%) (partial response: n = 5, stable disease: n = 20) with FOLFIRI. Median TTP was 3.0 mo (95% cI: 2.1-3.9 mo) and median OS was 6.6 mo (95% cI: 5.3-8.1 mo). Dose adaptation was required in 36 patients (57.1%). Fifteen patients (23.8%) had grade 3-4 toxicities, mainly hematological (n = 11) or digestive (n = 4). Febrile neutropenia occurred in 3 patients. There was no toxic death. In the present study, physico-chemical investigations have been carried out on the possibility of using cameroonian volcanic or clay pozzolans as raw material for geopolymer or pozzolanic binder. The research had made some suggestive results and conclusions. Powders of less than 100 μm of five sampled pozzolans from volcanic or clay origins have been subjected to chemical and mineralogical analysis, BET specific surface, absolute density, granulometry and pozzolanic activity in solution tests. The results obtained showed that, geopolymers or pozzolanic binders can be produced from samples studied. The samples contain significant amounts of glassy or amorphous phase ready to dissolve in an alkaline solution. The high alkali content of volcanic pozzolans makes them more appropriate for geopolymer application. clay pozzolans are the easier to grind in order to obtain the appropriate fineness and can be used for both geo- polymers and pozzolanic binders.

拉幅机广泛应用于织物的染色,漂白和印花工序,作用是沿幅宽方向对织物进行拉伸。织物夹具(布铗)作为拉幅定型机、丝光机中广泛使用的部件,承担着使织物有较好的定型效果,避免出现“荷叶边”、“破边”等缺陷的作用。但是,布铗在使用过程中,处于高温、反复拉幅张力的作用,在未到使用期限前就出现了脱铗失效现象,提前报废。因此,无论是布铗的生产企业,还是使用厂家,都关注该部件的失效现象,研究延长使用寿命的措施。本论文结合试验数据,主要采用有限元法分析某机械制造公司提供的布铗及高温(230℃)、持续工作条件(260天以上)工艺条件,从材料、加工安装方式、使用工况等方面研究布铗的失效行为。主要工作如下:1.对2种不同类型的织物试样,分别沿经向、纬向进行不同张力的拉伸试验。以布铗对称模型为研究对象,假设轻微磨损会影响布铗轴孔和夹体之间的滑动接触,采用ABAQUS软件分析拉幅力(100N、200N、300N)在不同刀铗形状曲线下,对织物接触压力分布的影响,并分析夹持角与夹持牢度的关系。2.根据传热理论,确定高温布铗的热传导方式,边界条件及热载荷施加形式。通过瞬态温度—结构耦合方法模拟了定型温度下,布铗的温度场、热应力及热变形分布状况。分析不同的刀铗主体轴孔与销轴的装配方式(间隙配合和过盈配合)对刀铗和底板变形后空间位置的影响,进一步分析夹持牢度。3.在230℃高温及各种拉力(100N、200N、300N)条件下,模拟分析了布铗主体轴孔蠕变变形。仿真结果表明,主体轴孔的蠕变变形随时间的增加而增大。在不同时间段(50天,200天,260天),利用cMM试验机对同意类型的布铗进行了尺寸变化和变形测试。将布铗主体轴孔的尺寸变化测试数据与布铗主体轴孔的蠕变变形数值分析进行比较,测试结果和数值结果表现出很好的一致性。4.分析引起布铗失效的蠕变、机械载荷和热变形等主要因素,提出了延长布铗使用寿命的措施。在高温及恒定拉幅力300N的条件下,结合解析法和有限元法,计算了布铗的可能失效时间。
