Olfactory ensheathing glia promote axonal regeneration in the mammalian central nervous system,including retinal ganglion cell axonal growth through the injured optic ***,it is unknown whether olfactory ensheathing glia also have neuroprotective *** ensheathing glia express brain-derived neurotrophic factor,one of the best neuroprotectants for axotomized retinal ganglion ***,we aimed to investigate the neuroprotective capacity of olfactory ensheating glia after optic nerve *** ensheathing glia cells from an established rat immortalized clonal cell line,TEG3,were intravitreally injected in intact and axotomized retinas in syngeneic and allogeneic mode with or without microglial inhibition or immunosuppressive *** and gene expression analyses were *** bulb-derived primary olfactory ensheathing glia and TEG3 express major histocompatibility complex classⅡ*** and syngenically transplanted TEG3 cells survived in the vitreous for up to 21 days,forming an *** axotomized retinas,only the allogeneic TEG3 transplant rescued retinal ganglion cells at 7 days but not at 21 *** these retinas,microglial anatomical activation was higher than after optic nerve crush *** intact retinas,both transplants activated microglial cells and caused retinal ganglion cell death at 21 days,a loss that was higher after allotransplantation,triggered by pyroptosis and partially rescued by microglial inhibition or ***,neuroprotection of axotomized retinal ganglion cells did not improve with these *** different neuroprotective properties,different toxic effects,and different responses to microglial inhibitory treatments of olfactory ensheathing glia in the retina depending on the type of transplant highlight the importance of thorough preclinical studies to explore these variables.

An optimal validation of a thematic map would ideally require in-situ observations of a large sample of units specifically conceived for the map under *** is often not possible due to budget *** alternative can be using photo-interpretation of high or very high resolution images instead of in-situ observations or using available data sets that do not fully comply with the ideal characteristics:unit size,reference date or sampling *** paper illustrates some examples of use of available data in the European *** land cover maps,the best existing data set is probably Land Use/Cover Areaframe Survey(LUCAS)that has been conducted by Eurostat on four occasions since *** LUCAS is based on systematic sampling,advantages and limitations of systematic sampling are discussed.A fine-scale population density map is presented as an example of a situation in which reference data on a statistical sample cannot be collected.

Numerical modeling of nutrient transport to assess the agricultural impact on the trophic state of reservoirs

A trophic state increase relates to surface water bodies nutrient enrichment,due to the chemical products used such as fertilizers in agriculture and residues from cattle raising *** research consists of nutrient transport numerical modeling to analyze the Betancíreservoir trophic state in Colombia;Water samples were collected to analyze total nitrogen,total phosphorus,ammonia,nitrates,nitrites,phosphates,chlorophyll-a,dissolved oxygen,BOD,COD,suspended solids,and water *** water quality model MOHID Studio was implemented and after its calibration,scenarios of increase and decrease of nutrients and inflows to the reservoir through its main tributaries were simulated to reproduce the agricultural activity changes in the basin and the effects that would have on the reservoir's trophic state dynamics;Therefore,the Carlson Trophic Status Index was calculated for each *** results show that variations in total nitrogen concentration and increased inflows present short-term consequences on the reservoir's trophic *** the incoming total nitrogen con-centrations by 100%causes the reservoir to change from a light eutrophic to a hypereutrophic *** results of this research provide a starting tool to water resources integrated management in reservoirs.

The Study of Carbamoyl Phosphate Synthetase 1 Deficiency Sheds Light on the Mechanism for Switching On/Off the Urea Cycle

Carbamoyl phosphate synthetase i （CPS1） deficiency （CPS1D） is an inborn error of the urea cycle having autosomal （2q34） recessive inheritance that can cause hyperammonemia and neonatal death or mental retardation. We analyzed the effects on CPS1 activity, kinetic parameters and enzyme stability of missense mutations reported in patients with CPS1 deficiency that map in the 20-kDa C-terminal domain of the enzyme. This domain turns on or off the enzyme depending on whether the essential allosteric activator of CPS 1, N-acetyl- L-glutamate （NAG）, is bound or is not bound to it. To carry out the present studies, we exploited a novel system that allows the expression in vitro and the purification of human CPS1, thus permitting site-directed mutagenesis. These studies have clarified disease causation by individual mutations, identifying functionally important residues, and revealing that a number of mutations decrease the affinity of the enzyme for NAG. Patients with NAG affinity-decreasing mutations might benefit from NAG site saturation therapy with N-carbamyl-L- glutamate （a registered drug, the analog of NAG）. Our results, together with additional present and prior site-directed mutagenesis data for other residues mapping in this domain, suggest an NAG-triggered conformational change in the 134-~4 loop of the C-terminal domain of this enzyme. This change might be an early event in the NAG activation process. Molecular dynamics simulations that were restrained according to the observed effects of the mutations are consistent with this hypothesis, providing further backing for this structurally plausible signaling mechanism by which NAG could trigger urea cycle activation via CPS1.

An overview of animal models for investigating the pathogenesis and therapeutic strategies in acute hepatic failure

Acute hepatic failure (AHF) is a severe liver injury accompanied by hepatic encephalopathy which causes multiorgan failure with an extremely high mortality rate, even if intensive care is provided. Management of severe AHF continues to be one of the most challenging problems in clinical medicine. Liver transplantation has been shown to be the most effective therapy, but the procedure is limited by shortage of donor organs. Although a number of clinical trials testing different liver assist devices are under way, these systems alone have no significant effect on patient survival and are only regarded as a useful approach to bridge patients with AHF to liver transplantation. As a result, reproducible experimental animal models resembling the clinical conditions are still needed. The three main approaches used to create an animal model for AHF are: surgical procedures, toxic liver injury and infective procedures. Most common models are based on surgical techniques (total/partial hepatectomy, complete/transient devascularization) or the use of hepatotoxic drugs (acetaminophen, galactosamine, thioacetamide, and others), and very few satisfactory viral models are available. We have recently developed a viral model of AHF by means of the inoculation of rabbits with the virus of rabbit hemorrhagic disease. This model displays biochemical and histological characteristics, and clinical features that resemble those in human AHF. In the present article an overview is given of the most widely used animal models of AHF, and their main advantages and disadvantages are reviewed.

Testing the effectiveness of conservation management within biosphere reserves:the case of the Mexican deer mouse(Peromyscus mexicanus)as a bioindicator

Biosphere reserves are protected areas whose purpose is to combine conservation and sustainable ***,their effectiveness has not been tested sufficiently,especially from an ecological and genetic ap*** this sense,the Peromyscus genus represents an excellent bioindicator to address these questions,due to its short life and high evolutionary rate and *** conservation managers,genetic structure can increase the rate of loss of genetic diversity because alleles exclusive of a subpopulation are more likely to disappear as a consequence of genetic drift in comparison with a panmictic *** we analyzed the abundance,movement distances,morphology/morphometry and genetic structure of 3 populations of Mexican deer mouse(Peromyscus mexicanus)located in different protected zones of La Tigra National Park(Honduras).Our results are consistent among the 3 approaches and showed the highest values of abundance,morphometry and genetic diversity in the population located at the core zone,whereas non-statistically significant differences were found between buffer and transition zone populations,suggesting suitable effectiveness of conservation management in the core zone but a lack of ecological buffering function of the other *** addition,the low movement distances and high genetic structure among the studied populations provide evidence of poor conservation management in the buffer and transition ***,we discuss the utility of the novel methodology used in this work,combining morphometry,abundance and genetics,in testing the effectiveness of conservation strategies in biosphere reserves,and the value of the Peromyscus genus as a bioindicator.

A sedentary lifestyle and obesity are important risk factors for severe COVID-19 complications and death[1,2].Excessive adipose tissue(AT)might contribute to more extensive viral spread with increased shedding,immune activation,and cytokine amplification[3].Several factors secreted by contracting muscle,termed myokines,mediate the beneficial effects of exercise in a wide range of diseases,including obesity[4].Thus,we conducted a study to explore the potential role of myokines of the fibronectin type III domain-containing family,FNDC4 and FNDC5,in mechanisms underlying the increased susceptibility to COVID-19 complications in obesity.

As the 5G ecosystem continues its consolidation,the testing and validation of the innovations achieved by integrators and verticals service providers is of preponderant *** this line,5GASP is a European H2020-funded project that aims at easing the idea-to-market process through the creation of an European testbed that is fully automated and self-service,in order to foster rapid development and testing of new and innovative 5G Network Applications(NetApps).The main objective of this paper is to present the 5GASP’s unified methodology to design,develop and onboard NetApps within the scope of different vertical services,letting them use specific 5G ***,we examine the whole 5GASP process in a tutorial fashion by adopting a specific use case focusing on the integration of a virtual On-Board Unit(vOBU)service that permits offloading processing from the attached vehicle and serving data-access *** demonstrated,the presented workflow permits the agile,rigorous,and safe development,testing and certification of NetApps,which will enable valuable in-network services for 5G and beyond infrastructures.

BACKGROUND The combination of alpha-fetoprotein(AFP)and squamous cell carcinoma antigen immunocomplex(SCCA-IgM)have been proposed for its use in the screening of hepatocellular carcinoma(HCC).Current screening programs for all cirrhotic patients are controversial and a personalized screening is an unmet need in the precision medicine *** To determine the role of the combination of SCCA-IgM and AFP in predicting mid-and long-term appearance of *** Two-hundred and three cirrhotic patients(Child A 74.9%,B 21.2%,C 3.9%)were followed-up prospectively every six months to screen HCC by ultrasound and AFP according to European Association for the Study of the Liver *** estimation cohort was recruited in Italy(30.5%;62/203)and validation cohort from Spain(69.5%;141/203).Patients underwent to evaluate SCCA-IgM by enzyme-linked immunosorbent assay(Hepa-IC,Xeptagen,Italy)and AFP levels at *** were followed-up for 60 mo,being censored at the time of the appearance of *** There were 10.8%and 23.1%of HCC development at two-and five-years *** with HCC showed higher levels of SCCA-IgM than those without it(425.72±568.33 AU/mL vs 195.93±188.40 AU/mL,P=0.009)during the fiveyear *** multivariate analysis,after adjusting by age,sex,aspartate transaminase and Child-Pugh,the following factors were independently associated with HCC:SCCA-IgM[Hazard ratio(HR)=1.001,95%CI:1.000-1.002;P=0.003],AFP(HR=1.028,95%CI:1.009-1.046;P=0.003)and creatinine(HR=1.56495%CI:1.151-2.124;P=0.004).The log-rank test of the combination resulted in 7.488(P=0.024)in estimation cohort and 11.061(P=0.004)in the validation cohort,and a 100%of correctly classified rate identifying a low-risk group in both cohorts in the two-year *** We have constructed a predictive model based on the combination of SCCA-IgM and AFP that provides a new HCC screening method,which could be followed by tailored HCC surveillance for individual patients,esp
