BACKGROUNd Mesenchymal stem cells(MSC)effects on tissue regeneration are mainly mediated by their secreted substances(secretome),inducing their paracrine *** Conditioned medium(CM),including soluble factors(proteins,nucleic acids,lipids)and extracellular vesicles is emerging as a potential alternative to cell ***,the manufacturing of CM suffers from variable procedures and protocols leading to varying results between ***,there is no welldefined optimized procedure targeting specific applications in regenerative *** To focus on conditioned medium produced from dental MSC(dMSC-CM),we reviewed the current parameters and manufacturing protocols,in order to propose a standardization and optimization of these manufacturing *** We have selected all publications investigating the effects of dental MSC secretome in in vitro and in vivo models of tissue regeneration,in accordance with the PRISMA *** A total of 351 results were *** based on the inclusion criteria described above,118 unique articles were included in the systematic ***-CM production was considered at three stages:before CM recovery(cell sources for CM),during CM production(culture conditions)and after production(CM treatment).CONCLUSION No clear consensus could be recovered as evidence-based methods,but we were able to describe the most commonly used protocols:donors under 30 years of age,dental pulp stem cells and exfoliated deciduous tooth stem cells with cell passage between 1 and 5,at a confluence of 70%to 80%.CM were often collected during 48 h,and stored at-80°*** is important to point out that the preconditioning environment had a significant impact on dMSCCM content and efficiency.

Insulin Effect on Gene Expression of dental Pulp Cell during Osteodifferentiation

Insulin, an old but still actual molecule, has many roles in various cellular processes including bone formation and also angiogenesis. Insulin effects on different cell types were investigated, and we intended to check its effect on dental pulp cells (dPC) during osteoblast differentiation. First, bone differentiation ability of isolated dental pulp cells was assessed by alizarin red (AR) staining. Insulin increased dental pulp cell proliferation after 25 hours of culture. It increased mRNA expression of osteogenic markers such as Col1, RunX2, ALP, Osc, Mef2C and expression of genes involved in TGF b pathway such as Smad3, TSP1, VEGF at different time points.

Chemo-mechanical couplings in compacted argillite submitted to high-pH environment

In the french concept of deep nuclear wastes repository, the galleries should be backfilled with excavated argillite after the site exploitation period. After several thousands of years, the degradation of the concrete lining of the galleries will generate alkaline fluid (pH > 12) that will diffuse through the backfill. The objective of the paper is to describe the influence of such solute diffusion on the microstructure and the mechanical behavior of compacted argillite. Saturated-portlandite water was circulated through compacted samples for 3, 6 and 12 months at 20 °C or 60 °C, respectively. The microstructures before and after fluid circulation were determined with mercury intrusion porosimetry. Since it was planned to introduce additives (bentonite or lime) in the remoulded argillite to backfill the deep galleries, such mixtures were also studied. The results show that the influence of the alkaline fluid on the properties of the argillite is a function of the nature of the additive. The pure argillite undergoes slight modifications that can be related to a limited dissolution of its clayey particles. Conversely, intense alteration of the bentonite-argillite mixture was observed. Lime addition improves the mechanical characteristics of the argillite through the precipitation of cementitious compounds.

Allogenic banking of dental pulp stem cells for innovative therapeutics

Medical research in regenerative medicine and cellbased therapy has brought encouraging perspectives for the use of stem cells in clinical trials. Multiple types of stem cells, from progenitors to pluripotent stem cells, have been investigated. Among these, dental pulp stem cells(dPSCs) are mesenchymal multipotent cells coming from the dental pulp, which is the soft tissue within teeth. They represent an interesting adult stem cell source because they are recovered in large amount in dental pulps with non-invasive techniques compared to other adult stem cell sources. dPSCs can be obtained from discarded teeth, especially wisdom teeth extracted for orthodontic reasons. To shift from promising preclinical results to therapeutic applications to human, dPSCs must be prepared in clinical grade lots and transformed into advanced therapy medicinal products(ATMP). As the production of patient-specific stem cells is costly and time-consuming, allogenic biobanking of clinical grade human leukocyte antigen(HLA)-typed dPSC lines provides efficient innovative therapeutic products. dPSC biobanks represent industrial and therapeutic innovations by using discarded biological tissues(dental pulps) as a source of mesenchymal stem cells to produce and store, in good manufacturing practice(GMP) conditions, dPSC therapeutic batches. In this review, we discuss about the challenges to transfer biological samples from a donor to HLA-typed dPSC therapeutic lots, following regulations, GMP guidelines and ethical principles. We also present some clinical applications, for which there is no efficient therapeutics so far, but that dPSCs-based ATMP could potentially treat.
