In an attempt to improve immune responses and protective efficacy,we constructed tworecombinant bacille Calmette-Guérin(rBCG)strains expressing an 85B antigen(Ag85B)and early secretedantigenic target-6 kDa antigen(ESAT6)of M vcobacterium tuberculosis(MTB)fusion *** rBCGstrains have the same protein insertion but in a different order(Ag85B-ESAT6 and ESAT6-Ag85B).Thecultured supernatant of rBCG strains and the sera from the mice immunized with the fusion protein Ag85B-ESAT6 or ESAT6-Ag85B formed a band with a fraction size of 37 kDa,equalivalent to the sum of Ag85B *** weeks after BALB/c mice were immunized with BCG or rBCG,spleen lymphocytes showedsignificant proliferation in response to culture filtrate protein of *** with the BCG group,micevaccinated with rBCG elicited a high level increase of immunoglobulin G antibodies to culture filtrate proteinin the *** γ-interferon levels in the lymphocyte culture medium supernatants increased remarkably inthe rBCG1 group,significantly higher than that of the BCG immunized group(P0.05).

AIM: To identify hepatitis C virus(HCV) core protein epitopes recognized by HLA-A2 restricted cytotoxic T lymphocyte (CTL). METHODS: Utilizing the method of computer prediction followed by a 4h(51)Cr release assay confirmation. RESULTS: The results showed that peripheral blood mononuclear cells (PBMC) obtained from two HLA-A2 positive donors who were infected with HCV could lyse autologous target cells labeled with peptide "ALAHGVRAL (core 150-158)". The rates of specific lysis of the cells from the two donors were 37.5% and 15.8%, respectively. Blocking of the CTL response with anti-CD4 mAb caused no significant decrease of the specific lysis. But blocking of CTL response with anti-CD8 mAb could abolish the lysis. CONCLUSION: The peptide (core 150-158) is the candidate epitope recognized by HLAA2 restricted CTL.

To investigate the characteristics of expression and distribution of 5 subtypes of somatostatin receptors (SSTR1～5) in lung cancer, in situ hybridization was used to examine the expression patterns of SSTR mRNAs in 21 cases of different pathologic types of lung cancer tissues with [a-35S]dATP labeled oligonucleotides of the 5 SSTR subtypes as probes. Additionally, Leica Q-500 image analysis processing system was employed for the semi-quantitatively analysis of the hybridization signals. Patterns of SSTR1～5 expression in lung cancer tissues were found as follows. SSTR2 was prominent in small cell lung cancer (SCLC), whereas in non-small cell lung cancer (NSCLC) including the adenous cancer (Ad) and the squamous cancer (Sq), the expression of SSTR1 mRNA was stronger than that of the other 4 types. The expression density of SSTR1～5 in the NSCLC was higher than the SCLC (p <0.01). The expression patterns and densities of the SSTR subtypes showed heterogeneity in different pathologic types of lung cancer. The expressions of the SSTR mRNAs in both SCLC and NSCLC indicated the positive prospects for somatostatin analog(SSA)--oriented agents in the treatment of both types of the lung cancer.

Activating transcription factor 3 (ATF3),a member of the activating transcription factor/cAMPresponsive element binding protein (ATF/CREB) family of transcription factors,is induced by manyphysiological *** investigate the activity of ATF/CREB in cells with physiological stresses,we developeda practical reporter vector,the plasmid pATF/CRE-luc,bearing activating transcription factor/cAMP responsiveelement (ATF/CRE) binding *** plasmid was constructed by inserting three repeats of the ATF/CREbinding element into the plasmid pG51uc,replacing the GAL-4 binding *** plasmids pACT/ATF3 andpATF/CRE-luc were transfected into HeLa and NIH3T3 cells,respectively,and the results showed that theexpression of luciferase .was increased in a dose-dependent manner on plasmid pACT/*** datasuggested that the plasmid pATF/CRE-luc could be used as a sensitive and convenient reporter system ofATF3 activity.

Protective effect of inosine against cell death after photochemically induced focal cerebral ischemia was investigated in adult rats. Intraperitoneal injection of inosine was started 24 h before operation and continued till the animals were *** TUNEL method and Nissl staining were used to identify apoptosis and necrosis,***-positive cells were located in the transitional zone between the necrotic core and normal *** mean maximum area of the necrotic cores and mean number of TUNEL-positive cells 24 h after operation were reduced significantly in the inosine-treated rats when compared with those in the saline-treated control animals. We suggest that inosine can significantly reduce both necrosis and apoptosis following cerebral ischemia induced by photothrombosis in adult rats.

Human phonation does not merely depend on the vibration of the vocal folds. Research by clinical and computer simulations has demonstrated that the false vocal fold (FVF) is an important laryngeal con-striction that plays a vital role during human voice production. This study explored the effects of the FVF gaps using both the three-dimensional Plexiglas model and the numerical computation methods. Twelve FVF gaps (ranging from 0.02 to 2.06 cm) were used in this study at three glottal angles (uniform and convergent/divergent 40°), two minimal glottal diameters (Dg) (0.04 cm and 0.06 cm) separately, and the constant subglottal pressure (8 cm H2O). The results suggested that (1) the intralaryngeal pressure was the lowest and the flow was the highest (least flow resistance) when the FVF gap was 1.5-2 times greater than Dg; (2) the divergent glottal angle gave lower pressure and greater flow than the conver-gent and uniform glottal angle as there were no FVF conditions; (3) the presence of the FVF decreased the effects of the glottal angle to a certain extent; and more importantly, (4) the presence of the FVF also moved the separation points downstream, straightened the glottal jet for a longer distance, decreased the overall laryngeal resistance, and reduced the energy dissipation, suggesting the significance of FVF in efficient voice production. These results may be incorporated in the phonatory models (physical or computational) for better understanding of vocal mechanics. The results might also be helpful in exploring the surgical and rehabilitative intervention of related voice problems.
