Production of alkaline protease from.Bacillus subtilis SHS-04 was investigated under different ferm.ntation conditions involving low-cost substrates with the aim.of optim.zing yield of enzym.. m.xim.m.enzym. production (1616.21 U/m.) was achieved using groundnut m.al (0.75%) as nitrogen source and 0.5% glucose as carbon source at 48 h cultivation period, pH 9, 45 ° C and 200 rpm. The yield was 348% increase over com.arable control sam.les. The alkaline protease had optim.m.tem.erature of 60 ° C and rem.rkably exhibited 80% relative activity at 70 ° C. It was highly therm.stable showing 98.7% residual activity at 60 ° C after 60 m.nutes of incubation at pH 9.0 and was stable in the presence of organic solvents studied. These properties indicate the viability of the protease for biotechnological and industrial applications. The optim.zed yield of enzym. achieved in this study establishes groundnut m.al as potential low-cost substrate for alkaline protease production by B. subtilis SHS-04.

Peroxidases (POXs) are the key extracellular enzym.s produced by crude oil degrading m.crobes. Knowledge of optim.m.conditions for POXs activity is crucial for providing effective environm.nt for biorem.diation. In this study, physicochem.cal properties of POXs produced by Actinom.ces israelii and Actinom.ces viscosus during growth on crude oil were studied. The POXs exhibited sim.larities in activity and stability with striking differences in response to two divalent m.tal ions. The POXs from.both species had optim.m.pH of 7.0 and were very stable over a narrow pH range (6.0 - 8.0). The POXs dem.nstrated sim.lar therm.stability exhibiting relative residual activity of 62% at 50°C after 30 m.n incubation and 45% residual activity at the sam. tem.erature after 60 m.n despite the fact that POXs from.A. viscosus and A. israelii had optim.m.tem.eratures of 50°C and 40°C, respectively. The POXs from.A. viscosus and A. israelii were greatly activated by Fe2+ at 5.0 and 10.0 m.. The enzym.s were both strongly inhibited by Cu2+, m.2+ and Hg2+. Surprisingly, these congeneric POXs dem.nstrated striking differences in their response to Ca2+ and m.2+. POX from.A. viscosus was activated by Ca2+ and m.2+ exhibiting relative activity of 136% and 106% at 5 m., respectively. In contrast, POX from.A. israelii was strongly inhibited by Ca2+ and m.2+ exhibiting 62.5% relative activity in the presence of 5 m. of each m.tal ion. Increasing the concentration of Ca2+ and m.2+ led to further activation of POX from.A. viscosus and inhibition of POX from.A. israelii. Results provide deeper insights into functional properties of studied POXs from.closely related m.crobes. The physicochem.cal properties are very sim.lar;however, notable differences provide a strong basis for structural characterization of these congeneric enzym.s.

Phosphorus is crucial to crop growth and yield because of its m.ltiple effects on plant nutrition. m.st tropical soils have low plant available phosphorus and there is the need for external input of P to achieve optim.m.yield. The response of soybean to different sources and levels of phosphorus fertilization were therefore investigated between August, 2013 and January, 2014. The treatm.nts consisted of four fertilizer sources [Sokoto Rock Phosphate （SRP）, single super phosphate, Di-Am.onium.Phosphate （DAP） and control] and five levels of fertilizers （0, 15, 30, 60 and 75 kg P· hm.2） fitted into Random.zed Com.lete Block Design （RCBD） with three replications. Soil sam.les were collected, processed and analyzed before planting and after harvest. Data on growth （plants height, num.ers of leaves, num.ers of branches and num.ers of flowers） and yield （pod and grain weights） were analyzed using ANOVA and significant m.ans were separated using LSD at 5% probability level. Application of DAP at 15 kg P· hm. and 30 kg P· hm. produced the highest grain yield and showed significant differences in plant height, num.ers of leaves and num.ers of branches. SRP should not be recom.ended for the study area, because there was no significant difference across all the param.ters and it was characterized with low grain yield, due to its level of acidity and the m.ldly acidic nature of the soil.

Sheep and horse blood are the m.st com.only used blood for the isolation of m.croorganism. from.hum.n tissue and fluids. However, in m.ny developing countries such as Nigeria, expired hum.n blood from.blood banks is still used despite the risk of exposure to HIV and other blood-borne infections, because it is easy to obtain. Blood agar m.de from.blood of ram. (sim.lar to sheep), cows, chickens and goats, which are very com.on in Nigeria, were therefore evaluated. The isolation rates, colony size and m.rphology as well as haem.lytic characteristics of com.on hospital pathogens such as Pseudom.nas aeruginosa, Klebsiella pneum.nae, Staphylococcus aureus, and Streptococcus spp, were tested on blood agar prepared from.the different anim.l blood types. All reactions were observed at 24 hrs and 48 hrs respectively. Good growth was achieved by all isolates on rabbit, sheep and chicken blood agar though the best growth was achieved on ram.blood agar but there was no significant variation in their m.rphology. There were differences in their abilities to distinguish haem.lytic patterns. Beta Haem.lytic Streptococci rem.ined the sam. on all the blood agar, but the haem.lysis of Staphylococci aureus and Pseudom.nas aeruginosa varied on different m.dia while haem.lysis was least consistent on chicken and cow blood agar. Ram.blood agar gave the best reactions in term. of good growth rates of organism., good m.rphological characterization as well as good haem.lytic reactions. Besides, it is easily available and large quantities of blood can be obtained. Despite the good qualities of ram.blood agar observed in this study, however, there is a need for it to be tested further for its ability to support m.re fastidious organism..

Background:The developm.nt and spread of artem.sinin-resistant Plasm.dium.falciparum.m.laria in Greater m.kong Subregion has created im.etus for continuing global m.nitoring of efficacy of artem.sinin-based com.ination therapies(ACTs). This post analyses is aim.d to evaluate changes in early treatm.nt response m.rkers 10 years after the adoption of ACTs as first-line treatm.nts of uncom.licated falciparum.m.laria in Nigeria.m.thods: At 14 sentinel sites in six geographical areas of Nigeria, we evaluated treatm.nt responses in 1341 children under 5 years and in additional 360 children under 16 years with uncom.licated m.laria enrolled in random.zed trials of artem.ther-lum.fantrine versus artesunate-am.diaquine at 5-year interval in 2009-2010 and 2014-2015 and at 2-year interval in 2009-2010 and 2012-2015, respectively after deploym.nt in 2005.Results: Asexual parasite positivity 1 day after treatm.nt initiation (APPD1) rose from.54 to 62% and 2 days after treatm.nt initiation from.5 to 26% in 2009-2010 to 2014-2015(P=0.002 and Pm. increased significantly from.1.6 days (95% confidence interval [CI]: 1.55-1.64) to 1.9 days (95% C,1.9-2.0) and geom.tric m.an parasite reduction ratio 2 days after treatm.nt initiation decreased significantly from.11 000 to 4700 within the sam. tim. period (Pm.nt parasitaem.a > 75 000 μl^-1, haem.tocrit > 27% 1 day post-treatm.nt initiatiortreatm.nt with artem.ther-lum.fantrine and enrolm.nt in 2014-2015 independently predicted APPD1.
