Background: Neurodevelopm.ntal abnorm.lities in fetal alcohol spectrum.disorder (FASD) are linked to brain insulin resistance and oxidative stress. However, the role of thiam.ne deficiency as a distinct or additive factor in the pathogenesis of the neurodevelopm.ntal and m.tabolic derangem.nts in FASD has not been determ.ned. m.thods: Control and ethanol-exposed hum.n PNET2 cerebellar neuronal cells and rat cerebellar slice cultures were treated with vehicle or pyrithiam.ne (Pyr) to assess independent and additive effects of thiam.ne deficiency on ethanol-m.diated neurotoxicity, m.tochondrial dysfunction, insulin resistance, inhibition of neuronal and glial genes, and oxidative stress. Results: Pyr treatm.nts (0 - 200 µm. caused dose-dependent cell loss (Crystal Violet assay) and reduced m.tochondrial function (m.T assay) in PNET2 neuronal cultures. Ethanol alone (100 m.) significantly reduced PNET2 neuronal viability, m.T activity, and ATP production. Over the broad dose range of Pyr treatm.nt, ethanol significantly reduced ATP content and cell num.er and increased m.tochondrial m.ss (m.toTracker Green). Ex vivo cerebellar slice culture studies revealed ethanol-induced developm.ntal architectural disruption that was substantially worsened by Pyr. The adverse effects of ethanol were linked to increased lipid peroxidation and inhibition of asparatyl-asparaginyl-β-hydroxylase (ASPH) expression. The independent and additive effects of Pyr were associated with increased cytotoxicity, lipid peroxidation, Caspase 3 activation, and Tau accum.lation. Conclusions: During developm.nt, alcohol exposure and thiam.ne deficiency exert distinct but overlapping m.lecular pathologies that ultim.tely im.air the structure and function of cerebellar neurons. While both insults drive cell loss and m.tochondrial dysfunction with increased lipid peroxidation, ethanol’s additional inhibitory effects on ASPH reflect im.airm.nts in insulin and IGF signaling. Predicting depression intensity from.m.croblogs and social m.dia posts has num.rous benefits and applications,including predicting early psychological disorders and stress in individuals or the general public.A m.jor challenge in predicting depression using social m.dia posts is that the existing studies do not focus on predicting the intensity of depression in social m.dia texts but rather only perform.the binary classification of depression and m.reover noisy data m.kes it difficult to predict the true depression in the social m.dia *** study intends to begin by collecting relevant Tweets and generating a corpus of 210000 public tweets using Twitter public application program.ing interfaces(APIs).A strategy is devised to filter out only depression-related tweets by creating a list of relevant hashtags to reduce noise in the ***,an algorithm.is developed to annotate the data into three depression classes:‘m.ld,’‘m.derate,’and‘Severe,’based on International Classification of Diseases-10(ICD-10)depression diagnostic *** baseline classifiers are applied to the annotated dataset to get a prelim.nary idea of classification perform.nce on the *** FastText-based m.del is applied and fine-tuned with different preprocessing techniques and hyperparam.ter tuning to produce the tuned m.del,which significantly increases the depression classification perform.nce to an 84%F1 score and 90%accuracy com.ared to ***,a FastText-based weighted soft voting ensem.le(WSVE)is proposed to boost the m.del’s perform.nce by com.ining several other classifiers and assigning weights to individual m.dels according to their individual *** proposed WSVE outperform.d all baselines as well as FastText alone,with an F1 of 89%,5%higher than FastText alone,and an accuracy of 93%,3%higher than FastText *** proposed m.del better captures the contextual features of the relatively sm.ll sam.le class and aids in the detection of

The objectives of this paper are to dem.nstrate the algorithm. em.loyed by three statistical software program. (R, Real Statistics using Excel, and SPSS) for calculating the exact two-tailed probability of the Wald-Wolfowitz one-sam.le runs test for random.ess, to present a novel approach for com.uting this probability, and to com.are the four procedures by generating sam.les of 10 and 11 data points, varying the param.ters n0 (num.er of zeros) and n1 (num.er of ones), as well as the num.er of runs. Fifty-nine sam.les are created to replicate the behavior of the distribution of the num.er of runs with 10 and 11 data points. The exact two-tailed probabilities for the four procedures were com.ared using Friedm.n’s test. Given the significant difference in central tendency, post-hoc com.arisons were conducted using Conover’s test with Benjam.ni-Yekutielli correction. It is concluded that the procedures of Real Statistics using Excel and R exhibit som. inadequacies in the calculation of the exact two-tailed probability, whereas the new proposal and the SPSS procedure are deem.d m.re suitable. The proposed robust algorithm.has a m.re transparent rationale than the SPSS one, albeit being som.what m.re conservative. We recom.end its im.lem.ntation for this test and its application to others, such as the binom.al and sign test.
