Mapping the Genetic Diversity of Castanea sativa: Exploiting Spatial Analysis for Biogeography and Conservation Studies

The current distribution of forest tree species is a result of natural or human mediated historical and contemporary processes. Knowledge of the spatial distribution of the diversity and divergence of populations is crucial for managing and conserving genetic resources in forest tree species. By combining tools from population genetics, landscape ecology and spatial statistics, landscape genetics thus represents a powerful method for evaluating the geographic patterns of genetic resources at the population level. In this study, we explore the possibility of combining genetic diversity data, spatial statistic tools and GIS technologies to map the genetic divergence and diversity of 31 Castanea sativa populations collected in Spain, Italy, Greece, and Turkey. The IDW technique was used to interpolate the diversity values and divergence indices as expected hetereozygosity (He), allelic richness (Rs), private allelic richness (PRs), and membership values (Q) of each population to different clusters. Genetic diversity maps and a synthetic map of the spatial genetic structure of European chestnut populations were produced. Spatial coincidences between landscape elements and statistically significant genetic discontinuities between populations were investigated. Evidence is provided of the significance of cartographic outputs produced in the study and on their usefulness in managing genetic resources.

Integration of Environmental and Social Values in Cultural Spaces:Sustainable and Inclusive Wayfinding Materials for Museums

In the context of use of large museum centers,numerous national and international methodological experiments show that the wayfinding project must consider the needs of both habitual users(user-centered design)and local communities(design for communities)and the importance of environmental protection(eco-design)as a priority interest of the community.This“double target”,“user-centered”and“environment-centered”can be applied during the selection process of materials to be used in the project.With respect to these possibilities,this contribution intends to present the results of research focused on material characterization of the reception and distribution spaces of large museum centers.This characterization is based on use of sensory materials and aims to evaluate their impact on the usability and sustainability of wayfinding systems.The paper directed towards a proposal for organization of integrated information on new generation so-called smart materials;within the design of a wayfinding system,these can balance the aesthetic-perceptual and performance and environmental impact,in order to allow designers to make informed decisions oriented towards inclusion and sustainability.The study was addressed by conducting two phases of systematic literature and library review of materials.The investigations conducted led to achievement of a first research result which consists in the identification of a“standard sheet”for the mapping and cataloging of the materials used for wayfinding.The“standard sheet”allows organizing the information on smart,sensorial,and eco-friendly materials,balancing the aesthetic-perceptive component with the performance on the environmental impact along the entire life cycle in a circular perspective.This tool could guide designers towards an environmental communication project oriented towards sustainability and is effective for usability and wayfinding.

Default rules in investment decision-making:trait anxiety and decision-making styles

This study investigates the role of default options in the relationship between trait anxiety,and decision-making styles and financial decisions.One hundred and ninety-four participants were divided into three groups and subjected to three different conditions.Under each experimental condition,they had to decide whether to accept or reject investment proposals.In the first group,they had been enrolled in investment plans by default(opt-out condition),in the second group,they had not been automatically enrolled in these plans(opt-in condition),and in the third group they had to choose whether to enroll or not(control condition).The results showed that the investment decisions of anxious,avoidant,rational and dependent individuals could be facilitated by default options.In conclusion,using default options as a“nudge”can support specific groups of people to improve their financial decisions.

X-linked hypophosphatemia(XLH)represents the most common form of familial hypophosphatemia.Although significant advances have been made in the treatment of bone pathology,patients undergoing therapy continue to experience significantly decreased oral health-related quality of life.The following study addresses this persistent oral disease by further investigating the effect of DMP1 expression on the differentiation of XLH dental pulp cells.Dental pulp cells were isolated from the third molars of XLH and healthy controls and stable transduction of full-length human DMP1 were achieved.RNA sequencing was performed to evaluate the genetic changes following the induction of odontogenic differentiation.RNAseq data shows the upregulation of inhibitors of the canonical Wnt pathway in XLH cells,while constitutive expression of full-length DMP1 in XLH cells reversed this effect during odontogenic differentiation.These results imply that inhibition of the canonical Wnt pathway may contribute to the pathophysiology of XLH and suggest a new therapeutic strategy for the management of oral disease.

The Partition of Unity Method for High-Order Finite Volume Schemes Using Radial Basis Functions Reconstruction

This paper introduces the use of partition of unity method for the development of a high order finite volume discretization scheme on unstructured grids for solving diffusion models based on partial differential equations.The unknown function and its gradient can be accurately reconstructed using high order optimal recovery based on radial basis functions.The methodology proposed is applied to the noise removal problem in functional surfaces and images.Numerical results demonstrate the effectiveness of the new numerical approach and provide experimental order of convergence.

Implicit Shape Reconstruction of Unorganized Points Using PDE-Based Deformable 3D Manifolds

In this work we consider the problem of shape reconstruction from an unorganized data set which has many important applications in medical imaging, scientific computing, reverse engineering and geometric modelling. The reconstructed surface is obtained by continuously deforming an initial surface following the Partial Differential Equation (PDE)-based diffusion model derived by a minimal volume-like variational formulation. The evolution is driven both by the distance from the data set and by the curvature analytically computed by it. The distance function is computed by implicit local interpolants defined in terms of radial basis functions. Space discretization of the PDE model is obtained by finite co-volume schemes and semi-implicit approach is used in time/scale. The use of a level set method for the numerical computation of the surface reconstruction allows us to handle complex geometry and even changing topology,without the need of user-interaction. Numerical examples demonstrate the ability of the proposed method to produce high quality reconstructions. Moreover, we show the effectiveness of the new approach to solve hole filling problems and Boolean operations between different data sets.

Hepatocellular carcinoma(HCC)is the most frequent liver neoplasm,and its incidence rates are constantly increasing.Despite the availability of potentially curative treatments(liver transplantation,surgical resection,thermal ablation),long-term outcomes are affected by a high recurrence rate(up to 70%of cases 5 years after treatment).HCC recurrence within 2 years of treatment is defined as“early”and is generally caused by the occult intrahepatic spread of the primary neoplasm and related to the tumor burden.A recurrence that occurs after 2 years of treatment is defined as“late”and is related to de novo HCC,independent of the primary neoplasm.Early HCC recurrence has a significantly poorer prognosis and outcome than late recurrence.Different pathogenesis corresponds to different predictors of the risk of early or late recurrence.An adequate knowledge of predictive factors and recurrence risk stratification guides the therapeutic strategy and post-treatment surveillance.Patients at high risk of HCC recurrence should be referred to treatments with the lowest recurrence rate and when standardized to combined or adjuvant therapy regimens.This review aimed to expose the recurrence predictors and examine the differences between predictors of early and late recurrence.

A direct enantio-,diastereo-,and chemo-selective high-performance liquid chromatographic method was developed for determining the content,enantiomeric purity,and related substances of the chiral antidepressant drug sertraline HCl in a single chromatographic run.The separation was achieved on a chiral stationary phase based on amylose tris(3-chloro-5-methylphenylcarbamate)under reversed-phase conditions.The method was optimized by evaluating the influence of the temperature and mobile phase composition on the retention and selectivity.The application of the single-run approach allowed to baseline resolve all investigated species in less than 15 min,without using buffers or tandem-coupled columns.The chromatographic method was validated according to the guidelines of the Official Medicines Control Laboratory and applied to control the content of sertraline HCl and related chiral substances in a generic antidepressant formulation.
