Upregulation of the APE1 and H2AX genes and miRNAs involved in DNA damage response and repair in gastric cancer

Gastric cancer remains one of the leading causes of cancer-related death worldwide,and most of the cases are associated with Helicobacter pylori *** bacterium promotes the production of reactive oxygen species(ROs),which cause DNA damage in gastric epithelial *** this study,we evaluated the expression of important genes involved in the recognition of DNA damage(ATM,ATR,and H2AX)and ROs-induced damage repair(APE1)and the expression of some miRNAs(miR-15a,miR-21,miR-24,miR-421 and miR-605)that target genes involved in the DNA damage response(DDR)in 31 fresh tissues of gastric *** v3.1.1 was used to construct the postulated miRNA:mRNA interaction *** performed by real-time quantitative PCR exhibited significantly increased levels of the APE1(RQ=2.55,ps beyond the miR-421 and miR-605 in the gastric cancer *** addition,significantly elevated levels of miR-21,miR-24 and miR-421 were observed in diffuse-type gastric *** analysis reinforced some of the gene:gene(ATM/ATR/H2AX)and miRNA:mRNA relationships obtained also with the interaction ***,our findings show that tumor cells from gastric cancer presents deregulation of genes and miRNAs that participate in the recognition and repair of DNA damage,which could confer an advantage to cell survival and proliferation in the tumor microenvironment.

The prostate is an accessory sex gland that develops under precise androgenic control. It is known that hormonal imbalance may disrupt its development predisposing this gland to develop diseases during aging. Although the hypothesis regarding earlier origins of prostate diseases was proposed many years ago, the mechanisms underlying this complex phenomenon are poorly understood. Therefore, the aim of this study was to evaluate the prostates of old male gerbils exposed to testosterone during intrauterine and postnatal life using morphological, biometrical, stereological, Kariometric, immunohistochemical, and immunofluorescence analyses. Our findings demonstrate that prenatal and pubertal exposure to testosterone increases the susceptibility to the development of prostate diseases during aging. The presence of a more proliferative gland associated with foci of adenomatous hyperplasia in animals exposed to testosterone during the prenatal and pubertal phase show that the utero life and the pubertal period are important phases for prostatic morphophysiology establishment, which is a determinant for the health of the gland during aging. Therefore, these findings reinforce the idea that prostate disease may result from hormonal disruptions in early events during prostate development, which imprint permanently on the gland predisposing it to develop lesions in later stages of life.

The world is facing dramatic challenges related to environmental sustainability at an accelerating *** this context,the field of economic geography(EG)has been playing an important role in understanding both the socioeconomic and technological dimensions of these challenges,as it deals with a variety of complementary notions and *** from this lens,our aim is to explore a conceptual framework that can help us to understand environmental changes relating to multi-dimensional territorial development,notably in eco-nomic contexts where inequality is high,and stratification based on hierarchies regulate social and economic *** on the territory concept,we propose the original notion of a hierarchical regional innovation system(HRIs)that emphasises the pervasive role of hierarchies(powers)in regional innovation systems and illustrate its value with evidence and case studies from extant literature on sustainability *** HRIs can help us understand and promote development paths considering the contribution of inclusive eco-innovations(another original conceptual amalgam).Through some empirical cases from other studies in low-carbon transitions,we show the application of the HRIs(and inclusive eco-innovation)*** conclusion,we provide incen-tives to explore new regional innovation systems,alongside the HRIs,adapted to different regions worldwide and centred on the inclusiveness of people and places.

Differences in viral kinetics between genotypes 1 and 3 of hepatitis C virus and between cirrhotic and non-cirrhotic patients during antiviral therapy

AIM： TO evaluate the impact of hepatitis C virus （HCV） infection with genotype 1 or 3 and the presence or absence of liver cirrhosis （LC） in the early viral kinetics response to treatment. METHODs： Naive patients （n = 46） treated with interferon-α （IFN-α） and ribavirin and followed up with frequent early HCV-RNA determinations were analysed. Patients were infected with genotype 1 （n = 28, 7 with LC） or 3 （n = 18, 5 with LC）. REsULTs： The first phase decline was larger in genotype 3 patients than in genotype 1 patients （1.72 vs 0.95 log IU/mL, P 〈 0.001）. The second phase slope decline was also larger in genotype 3 patients than in genotype 1 patients （0.87 vs 0.15 log/wk, P 〈 0.001）. Differences were found in both cirrhotic and non-cirrhotic patients. Genotype 1 cirrhotic patients had a slower 2^nd phase slope than non-cirrhotic patients （0.06 vs 0.18 log/wk, P 〈 0,02）. None of genotype 1 cirrhotic patients had a 1^st phase decline larger than 1 log （non-cirrhotic patients： 55%, P 〈 0.02）. A similar trend toward a slower 2nd phase slope was observed in genotype 3 cirrhotic patients but the 1^st phase slope decline was not different. sustained viral response was higher in genotype 3 patients than in genotype 1 patients ,（72% vs 14%, P 〈 0.001） and in genotype 1 non-cirrhotic patients than in genotype 1 cirrhotic patients （19% vs 0%）. A second phase decline slower than 0.3 log per week was predictive of non-response in all groups. CONCLUsION： Genotype 3 has faster early viral decline than genotype 1. Cirrhosis correlates with a slower 2nd phase decline and possibly with a lower 1^st phase slope decline in genotype 1 patients.

Critical bone defects are considered one of the major clinical challenges in reconstructive bone *** combination of 3D printed conductive scaffolds and exogenous electrical stimulation(Es)is a potential favorable approach for bone tissue *** this study,3D conductive scaffolds made with biocompatible and biodegradable polycaprolactone(PCL)and multi-walled carbon nanotubes(MWCNTs)were produced using the extrusion-based additive manufacturing to treat large calvary bone defects in *** results show that the use of PCL/MWCNTs scaffolds and Es contributes to thicker and increased bone tissue formation within the bone *** and mineralization are also significantly promoted using high concentration of MWCNTs(3 wt%)and ***,scaffolds favor the tartrate-resistant acid phosphatase(TRAP)positive cell formation,while the addition of MWCNTs seems to inhibit the osteoclastogenesis but present limited effects on the osteoclast functionalities(receptor activator of nuclear factor κβ ligand(RANKL)and osteoprotegerin(OPG)expressions).The use of Es promotes the osteoclastogenesis and RANKL expressions,showing a dominant effect in the bone remodeling *** results indicate that the combination of 3D printed conductive PCL/MWCNTs scaffold and Es is a promising strategy to treat critical bone defects and provide a cue to establish an optimal protocol to use conductive scaffolds and Es for bone tissue engineering.

Knowing what native trees can recr uit on degraded areas allows selecting the best species to restore these ***,as this information is not often available,experimentation is required before large-scale *** study used ex situ experiments to make these decisions on *** with r-strategist plants,excessive solar radiation and water shortage commonly impair tree recruitment in open *** experiments focused on the interactions among these factors and were conducted with three pioneer species from seasonally dry forests of northwest Argentina,Anadenanthera colubrina,Ceiba chodatii and Jacaranda *** of each species were sown at two light levels(sunlight/shade),two rainfall levels(full/reduced)and two levels of interspecific competition(with/without competitor)in a tree *** emergence and survival were monitored over a year and the results indicate that species differentially respond to varying levels of light and *** of *** tolerated water shortages under elevated solar radiation,which are desirable features for forest *** of *** tolerated shade and drought,suggesting that they require shading for establishing in open ***,*** seedlings neither tolerated full sunlight nor water shortages,suggesting that this species requires shading and regular watering if used in *** of the effects of light and water,the survival of all species was reduced by interspecific *** results highlight the importance of experimentation for selecting the best species for forest restoration and can enhance the cost/benefit ratios of these actions.

Internal migrations as phenomenon of social mobility and territorial experience among architecture students, University of Tolima,Ibagué, Colombia

Internal migration motivated by access to higher education is a relatively unexplored topic in Colombia, leading to a lack of data on its dimensions, characteristics, and impacts on urban space and the emotional conditions of migrants. For this reason, an academic program of architecture in a public university was selected to comprehend the internal migrations of students, exploring their territorial experiences, living conditions, and social mobility capacity. An exploratory methodology was designed to recognize the phenomenon through a descriptive analysis, utilizing institutional data, surveys, and interviews. The research identifies the primary reasons, barriers, and impacts experienced by the migrant population in Ibagué, Colombia. The study concludes with recommendations pertaining to both institutional and urban planning to address these identified challenges.
