Anti-Candida and anti-Cryptococcus evaluation of 15 non-alkaloidal compounds from Pterogyne nitens

Objective: To evaluate anti-Candida and anti-Cryptococcus activities of 15 nonalkaloidal compounds from Pterogyne nitens Tulasne(Leguminosae), a South American medicinal ***: Compounds were submitted to antifungal assays, using microdilution method described by Clinical and Laboratory Standards Institute document, with minor modifications. Five species of Candida and two species of Cryptococcus, including clinical isolates were screened. Antifungal activity was expressed by minimum inhibitory concentration(mIC). Amphotericin B and fluconazole were used as standard antifungal ***: Among tested compounds, six substances presented fungal growth inhibition(mIC mg/m L) [three flavone derivatives(1–3), a glycosylated flavonol derivative(5)and two phenolic acids(10 and 12)]. Sorbifolin(1), exhibited potent antifungal activity,demonstrating mIC value of 3.90 mg/m L against Candida glabrata ATCC 90030, Cryptococcus gattii 118 and fluconazole-resistant clinical isolate of Cryptococcus neoformans var. grubii. Pedalin(2) and nitensoside B(3), two glycosylated flavone derivatives, were active against Cryptococcus neoformans ATCC 90012(mIC = 7.80 mg/m L).Conclusions: Flavone derivatives from Pterogyne nitens can serve as prototypes for the design and development of innovative anti-Candida and anti-Cryptococcus hits.

Dose of alendronate directly increases trabeculae expansivity without altering bone volume in rat femurs

AIm To evaluate the effects of sodium alendronate on bone repair in fractures created in appendicular bones. mETHODS Wistar rats(n = 36) were allocated into three distinct groups: group C(control), group B1(received 1 mg/kg of alendronate), and group B2(received 3 mg/kg of alendronate). The rats underwent femoral transversal linear fracture surgery using stable internal fixation with a 2.0 mm plate and screw system. Each animal randomly received intraperitoneal applications of sodium alendronate at a dose corresponding to group B1 or B2 three times a week, while the control group received a 0.9% saline solution. Drug administration was performed until euthanasia at 45 d. The femurs were removed and each surgical piece was sent for radiographic, tomographic and microtomographic analysis. Data were submitted to descriptive and inferential statistical analysis(95% confidence interval).RESULTS Quantitative evaluations of bone neoformation did not show differences among the groups in the radiographic(P = 0.341), microtomographic(P = 0.581) and tomographic evaluations(P = 0.171). In the qualitative microtomographic analysis, a smaller distance was observed between the internal bone trabeculae in the groups that used alendronate(P = 0.05). On the other hand, group B2 had a higher amount of bone trabeculae per unit length when compared to the other groups(P = 0.04).CONCLUSION It is likely that the use of alendronate did not have a direct influence on the amount of bone neoformation, however it did influence the bone quality in a dosedependent manner, ultimately affecting the distance and quantity of the trabeculae.

Analysis of the interaction between [Ru(phenanthroline)<sub>3</sub>]<sup>2+</sup>and bovine serum albumin

The interaction of compounds with potential use as pharmaceutical with a carrier protein as serum albumin is of great importance in their biodistribution. Albumin offers different sites for binding metallic compounds. Using a combination of spectropho-tometric and electrochemical techniques, the interaction between [Ru(phen)3]Cl2 (phen = phenantroline) and bovine serum albumin was evaluated. In particular, it was possible to calculate an apparent binding constant (Kb) of 4.4 × 103 (for concentrations expressed in m) for the main interaction site of the protein. A number of ca. 40 molecules of Ru-phen per molecule of BSA under saturation conditions, and a positive cooperative behavior towards association from the protein were found.

CYP1A1,CYP2E1 and EPHX1 polymorphisms in sporadic colorectal neoplasms

AIm To investigate the contribution of polymorphisms in the CYP1A1, CYP2E1 and EPHX1 genes on sporadic colorectal cancer(SCRC) risk. mETHODS Six hundred forty-one individuals(227 patients with SCRC and 400 controls) were enrolled in the study. The variables analyzed were age, gender, tobacco and alcohol consumption, and clinical and histopathological tumor parameters. The CYP1A1 *2A, CYP1A1 *2C CYP2E1 *5B and CYP2E1 *6 polymorphisms were analyzed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). The EPHX1 Tyr113 His, EPHX1 His139 Arg and CYP1A1 *2C polymorphisms were detected by real-time PCR. Chisquared test and binary logistic regression were used in the statistical analysis. Fe_(3)O_(4)magnetic xerogel composites were prepared by polycondensation of resorcinol(R)-formaldehyde reaction via a sol-gel process in an aqueous solution through varying the molar ratio of Fe_(3)O_(4)nanoparticles(mNPs),catalyst(C),and water(W)*** were obtained by co-precipitation(mC),oxidation of iron salts(mO),or solvothermal synthesis(mS).Both mNPs and magnetic xerogels were examined regarding the performance of arsenic and fluoride removal in a batch *** mC-based mNPs had higher adsorption capacities for both fluoride(202.9 mg/g)and arsenic(3.2 mg/g)than other mNPs in optimum *** X-ray diffraction,Fourier transform infrared spectroscopy,and energy-dispersive X-ray spectroscopy confirmed that Fe was composed into the polymeric matrix of magnetic xerogels that contained 0.59%-4.42%of Fe with a molar ratio of mNPs(m)to R between 0.01 and *** low R/C and optimum m/R ratios,an increase in the surface area of magnetic xerogels affected the fluoride and arsenic adsorption *** magnetic xerogel composites with the mC-based mNPs prepared at a fixed R/C ratio(100)and at different R/W(0.05-0.06)and m/R(0.07-0.10)ratios had a high arsenic removal efficiency of 100%at an As(V)concentration of 0.1 mg/L and pH of *** maximum adsorption capacities of magnetic xerogels were approximately five times higher than those of the xerogels without mNP ***,Fe_(3)O_(4)nanoparticles enhanced the adsorption of arsenate and *** variations of alkaline catalyst and water content significantly affected the resulting properties of textural and surface chemistry of magnetic xerogel composites.

Event-based hydrology and sedimentation in paired watersheds under commercial eucalyptus and grasslands in the Brazilian Pampa biome

Ecological and hydrological impacts are expected when watersheds are under commercial *** this study we evaluated event-based hydrology and sedimentation in paired watersheds under commercial eucalyptus and grasslands ***,suspended sediment concentration,and sediment production were measured and simulated with the Limburg Soil Erosion model(LISEm)in paired hydrographic watersheds located in southern Brazil;one occupied with 7-years old Eucalyptus saligna plantation(forest watershed-FW;0.83 km^(2))and another Pampa biome with native and exotic grasses used for beef cattle production(grassland watershed-GW;1.10 km^(2)).The effect of rainfall on hydrological and sedimentological variables was more evident in GW compared with *** shape of the hydrograph was steepest in GW,with rapid,marked increase and decrease in flow during *** parameters changed for model calibration in FW and GW were soil saturated hydraulic conductivity,initial soil moisture,and manning roughness coefficient,whereby the statistical performance was classified as good(NSE>0.75).Fourteen events different from those used in the calibration process were validated for FW and GW,with NSE index classified as“good”and“very good”for *** study shows limitations of LISEm in calibrating and validating the sediment yield for individual events due to the spatial variability of factors controlling soil *** validated modeling results of this study may be applied in simulating soil and crop scenarios in watersheds cultivated with commercial forest and grassland for grazing.

Antimicrobial Efficacy of Curcumin Formulations byPhotodynamic Therapy

URTI （upper respiratory tract infections） are caused by acute, chronic or recurrent infections including pharyngitis andtonsillitis. The etiologic agents involved are bacteria, virus, and fungi. The standard treatment for bacterial infections is the use ofantibiotics. The antibiotic resistance, side effects of drugs and URTI complications indicate the importance of new therapies. PDT（photodynamic therapy） is a treatment option with a fast onset of action, low side effects, and easy administration of PS（photosensitizer） based on this study. The main aim of this study was to develop a new modality for the treatment of infectiouspharyngotonsillitis by photodynamic therapy. In addition, this study aims to evaluate components of the therapy as the PS, curcumin,in two formulations and irradiation conditions in the effectiveness of treatment. The procedure consists of two steps： oraladministration of CFs （curcumin formulations） containing a PS and illumination （450 nm and 0-60 J/cm2）. Bacterial isolates wereobtained from patients in a routine clinical pratice. The new methodology has been developed from the principle ofPI （photodynamicinactivation）. The strain identification number of colony forming units determined the efficacy of PI using each component, lightingand PS. The use of curcumin formulations in PI presented microbial reduction of 5 log.

Adaptation and Validation of a Proteolytic Activity methodology Using Dye-Crosslinked Substrate Tablets

Enzymatic activities are important to be quantified in products as enzymatic cleaners, which are used in medical and surgical devices reprocessing. Enzymatic activities are critical for the proper chemical cleaning that intends to remove solid organic dirt from inaccessible sites. The most important enzyme for this purpose is the protease, which is able to dissolve the main dirt attached to medical and surgical instruments. In this context, this study contributes to the development of a new proteolytic activity quantification method and its validation. The methodology is based on colorimetry and uses a UV-Vis spectrophotometer to measure the substrate hydrolysis by the blue color intensity, employing Protazyme AK tablets as substrate.

Upregulation of the APE1 and H2AX genes and miRNAs involved in DNA damage response and repair in gastric cancer

Gastric cancer remains one of the leading causes of cancer-related death worldwide,and most of the cases are associated with Helicobacter pylori *** bacterium promotes the production of reactive oxygen species(ROS),which cause DNA damage in gastric epithelial *** this study,we evaluated the expression of important genes involved in the recognition of DNA damage(ATm,ATR,and H2AX)and ROS-induced damage repair(APE1)and the expression of some miRNAs(miR-15a,miR-21,miR-24,miR-421 and miR-605)that target genes involved in the DNA damage response(DDR)in 31 fresh tissues of gastric *** v3.1.1 was used to construct the postulated miRNA:mRNA interaction *** performed by real-time quantitative PCR exhibited significantly increased levels of the APE1(RQ=2.55,pmiR-421 and miR-605 in the gastric cancer *** addition,significantly elevated levels of miR-21,miR-24 and miR-421 were observed in diffuse-type gastric *** analysis reinforced some of the gene:gene(ATm/ATR/H2AX)and miRNA:mRNA relationships obtained also with the interaction ***,our findings show that tumor cells from gastric cancer presents deregulation of genes and miRNAs that participate in the recognition and repair of DNA damage,which could confer an advantage to cell survival and proliferation in the tumor microenvironment.
