Peripartal rumen-protected methionine supplementation to higher energy diets elicits positive effects on blood neutrophil gene networks, performance and liver lipid content in dairy cows

Background： main objectives were to determine to what extent Smartamine m（Sm） supplementation to a prepartal higher-energy diet could alter neutrophil（PmN） and liver tissue immunometabolic biomarkers, and whether those responses were comparable to those in cows fed a prepartal lower-energy diet（CON）.Results： Twenty-eight multiparous Holstein cows were fed CON（NEL= 1.24 mcal/kg Dm） during d-50 to d-22 relative to calving. From d-21 to calving, cows were randomly assigned to a higher-energy diet（OVE, n = 9; NEL= 1.54 mcal/kg Dm）, OVE plus Sm（OVE ＋ Sm, n = 10; Sm = 0.07 % of Dm） or remained on CON（n = 9）. Al cows received the same basal lactation diet（NEL= 1.75 mcal/kg Dm）. Supplementation of Sm（OVE ＋ Sm） continued until 30 d postpartum. Liver biopsies were harvested at d-10, 7, and 21 relative to parturition. Blood PmN isolated at-10, 3, and 21 d relative to calving was used to evaluate gene expression. As expected, OVE increased liver lipid content postpartum; however,cows fed OVE ＋ Sm or CON had lower concentrations than OVE. Compared with OVE, cows in CON and OVE ＋ Sm had greater DmI postpartum and milk production. Furthermore, cows fed OVE ＋ Sm had the greatest milk protein and fat percentage and lowest milk SCC despite having intermediate PmN phagocytic capacity. Adaptations in PmN gene expression in OVE ＋ Sm cows associated with the lower SCC were gradual increases from-10 to 21 d in genes that facilitate migration into inflammatory sites（SELL, ITGAm）, enzymes essential for reducing reactive oxygen metabolites（SOD1, SOD2）, and a transcription factor（s） required for controlling PmN development（RXRA）. The greater expression of TLR4 on d 3, key for activation of innate immunity due to inflammation, in OVE compared with CON cows suggests a more pronounced inflammatory state. Feeding OVE ＋ Sm dampened the upregulation of TLR4, despite the fact that these cows had similar expression of the pro-inflammatory genes NFKB1 and TNF as OVE. Cows in CON had lower overall expr

The prostate is an accessory sex gland that develops under precise androgenic control. It is known that hormonal imbalance may disrupt its development predisposing this gland to develop diseases during aging. Although the hypothesis regarding earlier origins of prostate diseases was proposed many years ago, the mechanisms underlying this complex phenomenon are poorly understood. Therefore, the aim of this study was to evaluate the prostates of old male gerbils exposed to testosterone during intrauterine and postnatal life using morphological, biometrical, stereological, Kariometric, immunohistochemical, and immunofluorescence analyses. Our findings demonstrate that prenatal and pubertal exposure to testosterone increases the susceptibility to the development of prostate diseases during aging. The presence of a more proliferative gland associated with foci of adenomatous hyperplasia in animals exposed to testosterone during the prenatal and pubertal phase show that the utero life and the pubertal period are important phases for prostatic morphophysiology establishment, which is a determinant for the health of the gland during aging. Therefore, these findings reinforce the idea that prostate disease may result from hormonal disruptions in early events during prostate development, which imprint permanently on the gland predisposing it to develop lesions in later stages of life.

The renin-angiotensin system （RAS） has been known for more than a century as a cascade that regulates body fuid balance and blood pressure. Angiotensin Ⅱ（Ang Ⅱ） has many functions in different tissues; how-ever it is on the kidney that this peptide exerts its main functions. New enzymes, alternative routes for Ang Ⅱformation or even active Ang Ⅱ-derived peptides have now been described acting on Ang Ⅱ AT1 or AT2 recep-tors, or in receptors which have recently been cloned, such as mas and AT4. Another interesting observation was that old members of the RAS, such as angioten-sin converting enzyme （ACE）, renin and prorenin, well known by its enzymatic activity, can also activate intra-cellular signaling pathways, acting as an outside-in sig-nal transduction molecule or on the renin/（Pro）renin re-ceptor. moreover, the endocrine RAS, now is also known to have paracrine, autocrine and intracrine action on different tissues, expressing necessary components for local Ang Ⅱ formation. This in situ formation, especially in the kidney, increases Ang Ⅱ levels to regulate blood pressure and renal functions. These discoveries, such as the ACE2/Ang-（1-7）/mas axis and its antangonistic effect rather than classical deleterious Ang Ⅱ effects, improves the development of new drugs for treating hypertension and cardiovascular diseases.

Objective:We describe the first-year implementation experience of an Instrumented Sensory Integration Therapy Program in Audiological&Balance Center ***:This is a retrospective descriptive *** included Seventy-three adults with diagnoses of acute,episodic,or chronic vestibular *** were classified into the following two groups:group 1 included 46 individuals treated with ISIT plus VRT,and group 2 included 27 individuals treated only with ***:The Sensory Organization Test(SOT)for both groups showed a statistical significance for all three sensory inputs;visual systems(G1:p=0.0003;G2:p=0.0337),vestibular system(G1:pmonstrated by compound balance score(G1:pmplexity of functional neurological disorders in the context of vestibular syndromes seem to require between 10 and 20 therapy sessions,and combined ISIT plus VRT appears to be more effective than ISIT as a monotherapy.

Leishmaniasis is a set of diseases with a worldwide distribution that affects mainly economically underprivileged populations in developing countries. It has a major impact on public health, with a global cost of billions of dollars per year. The treatment and control of leishmaniasis vary according to the Leishmania species involved, which require reliable methods for species identification. Since most of the currently used methods have limitations, there is a need for assays that allow rapid, precise identification of the offending species. Real-time polymerase chain reactions in conjunction with dissociation curve analysis have been used to detect differences in the DNA composition of selected genes of Leishmania spp. Kinetoplast DNA is the main molecular target used because of its high copy number per parasite, but other targets have also been studied. As part of an effort to establish melting temperature standards for each target gene, we have reviewed the pertinent literature available in public databases, including Pub med, Web of Science, Sci ELO and LILACS, using the keywords "Leishmania", "leishmaniasis", "realtime PCR", "melting temperature", and "melting curve", alone or in combination. After applying eligibility criteria, 27 articles were selected for analysis. A considerable variation in the methodologies analyzed was found regarding molecular targets, standardization of the methods, reproducibility and specificity. Because of this, statistical analysis was not performed. In most cases, the methods were able to differentiate the parasite at the subgenus level or few species regardless of the target chosen.

The consumption of marine macroalgae has been linked with health benefits,albeit some actions,including genoprotection,remain ***,we evaluated the genoprotective potential of a mixture of Ulva rigida,Fucus vesiculosus and Gracilaria gracilis,through direct consumption or via indirect intake using fish(Sparus aurata)as a vector of algal ***(mus musculus)were subjected to a 1-month dietary supplementation with 5%of macroalgae mix or 10%of fillet of fish previously fed with an algae-supplemented aquafeed(for comparison purposes,10%supplementation with fillet of fish standardly fed was also considered).Thereafter,mice were challenged by the genotoxicant methyl methanesulfonate(mmS),and the genetic damage was evaluated through the micronuclei ***,a wider evaluation was carried out encompassing the assessment of energy metabolism,haematological and histological parameters,as well as growth *** unequivocally protected against mmS-induced *** activity profile of hepatic lactate and isocitrate dehydrogenases promoted by the supplemented diets reflected an improved energy *** properties were not transferred via fish fed with the algae-supplemented aquafeed,though the transference of phytocomponents with beneficial properties(*** balance improvement)should not be ***,these findings highlighted the genoprotection afforded by marine macroalgae,likely promoted by desmutagenic factors,reinforcing their definition as functional *** possibility of using fish as an indirect source of algae-borne phytocomponents proved to be plausible and worthy of further investigations.

The immune responses of humans and animals to insults(i.e., infections, traumas, tumoral transformation and radiation) are based on an intricate network of cells and chemical messengers. Abnormally high inflammation immediately after insult or abnormally prolonged proinflammatory stimuli bringing about chronic inflammation can lead to life-threatening or severely debilitating diseases. mesenchymal stem cell(mSC) transplant has proved to be an effective therapy in preclinical studies which evaluated a vast diversity of inflammatory conditions. mSCs lead to resolution of inflammation, preparation for regeneration and actual regeneration, and then ultimate return to normal baseline or homeostasis. However, in clinical trials of transplanted mSCs, the expectations of great medical benefit have not yet been fulfilled. As a practical alternative to mSC transplant, a synthetic drug with the capacity to boost endogenous mSC expansion and/or activation may also be effective. Regarding this, ImT504, the prototype of a major class of immunomodulatory oligonucleotides, induces in vivo expansion of mSCs, resulting in a marked improvement in preclinical models of neuropathic pain, osteoporosis, diabetes and sepsis. ImT504 is easily manufactured and has an excellent preclinical safety record. In the small number of patients studied thus far, ImT504 has been well-tolerated, even at very high dosage. Further clinical investigation is necessary to demonstrate the utility of ImT504 for resolution of inflammation and regeneration in a broad array of human diseases that would likely benefit from an immunoprotective/immunoregenerative therapy.
