Peripartal rumen-protected methionine supplementation to higher energy diets elicits positive effects on blood neutrophil gene networks, performance and liver lipid content in dairy cows

Background： Main objectives were to determine to what extent Smartamine M（SM） supplementation to a prepartal higher-energy diet could alter neutrophil（PMN） and liver tissue immunometabolic biomarkers, and whether those responses were comparable to those in cows fed a prepartal lower-energy diet（CON）.Results： Twenty-eight multiparous Holstein cows were fed CON（NEL= 1.24 Mcal/kg DM） during d-50 to d-22 relative to calving. From d-21 to calving, cows were randomly assigned to a higher-energy diet（OVE, n = 9; NEL= 1.54 Mcal/kg DM）, OVE plus SM（OVE ＋ SM, n = 10; SM = 0.07 % of DM） or remained on CON（n = 9）. Al cows received the same basal lactation diet（NEL= 1.75 Mcal/kg DM）. Supplementation of SM（OVE ＋ SM） continued until 30 d postpartum. Liver biopsies were harvested at d-10, 7, and 21 relative to parturition. Blood PMN isolated at-10, 3, and 21 d relative to calving was used to evaluate gene expression. As expected, OVE increased liver lipid content postpartum; however,cows fed OVE ＋ SM or CON had lower concentrations than OVE. Compared with OVE, cows in CON and OVE ＋ SM had greater DMI postpartum and milk production. Furthermore, cows fed OVE ＋ SM had the greatest milk protein and fat percentage and lowest milk SCC despite having intermediate PMN phagocytic capacity. Adaptations in PMN gene expression in OVE ＋ SM cows associated with the lower SCC were gradual increases from-10 to 21 d in genes that facilitate migration into inflammatory sites（SELL, ITGAM）, enzymes essential for reducing reactive oxygen metabolites（SOD1, SOD2）, and a transcription factor（s） required for controlling PMN development（RXRA）. The greater expression of TLR4 on d 3, key for activation of innate immunity due to inflammation, in OVE compared with CON cows suggests a more pronounced inflammatory state. Feeding OVE ＋ SM dampened the upregulation of TLR4, despite the fact that these cows had similar expression of the pro-inflammatory genes NFKB1 and TNF as OVE. Cows in CON had lower overall expr

A Randomized Controlled Trial of Mindfulness in Recovery from Colorectal Cancer

Objective:This study examined whether a 4-week group-based mindfulness intervention would be superior in reducing psychological distress in colorectal cancer(CRC)patients compared to a psychoeducation and cognitive behavioural skills learning support active control ***:Patients with CRC were randomized via Computerised Permuted Block Randomisation to mindfulness or active control groups(2-h weekly sessions over 4 weeks).Outcomes were measured pre-intervention,and 8 weeks and 6 months *** primary outcome was psychological distress measured by the Hospital Anxiety and Depression *** outcomes were generic quality of life(QoL),disease specific QoL,mindfulness,and intervention credibility and ***:Sixty-eight participants were randomized to mindfulness(n=35)or active control group(n=33).Uptake of potentially eligible patients consenting was low(28.0%)and the dropout rate was 33.8%.Depression scores were reduced in both groups at week 8(P=0.020).Control participants had greater improvement in generic mental QoL scores at week 8 than mindfulness(P=0.023).In disease specific QoL,there was reduction in impotence symptom in the mindfulness group(P=0.022)and reduction in faecal incontinence in the control group(P=0.019).The embarrassment symptom had a significantly lower increase in the mindfulness group at week 8 compared to the control group(P=0.009).Both groups rated the treatments as credible and ***:Mindfulness was not superior to the active control group in terms of alleviating psychological distress but both treatments were associated with some improvements in *** was low uptake of both interventions.(Trial registration number:ACTRN12616001033437)

Objective To evaluate the efficiency of silymarin(SMN) in modulating metabolic parameters and redox status in rats with type 1 diabetes mellitus(T1 DM).Methods Diabetes was induced by intraperitoneal injection of alloxan. The diabetic rats were administered with SMN at doses of 50 and 100 mg/kg body weight/d for 30 consecutive days. The rats were divided into the following four groups: vehicle control, diabetic(alloxan-treated), DS50(alloxan +50 mg/kg body weight/d of SMN), and DS100(alloxan + 100 mg/kg body weight/d of SMN) groups. The bodyweight and food and water intake were evaluated. After 30 d, the animals were euthanized and the blood was collected for measuring the serum levels of glucose, triacylglycerol(TAG), urea, and creatinine. The liver and pancreas were collected for measuring the activities of superoxide dismutase(SOD) and catalase(CAT), and the levels of carbonylated protein(PC). The pancreas sample was also used for histological *** SMN reduced hepatic(P < 0.001) and pancreatic(P < 0.001) protein damage and creatinine levels(P = 0.0141) in addition to decreasing food(P < 0.001) and water intake(P < 0.001). However,treatment with SMN did not improve beta-cell function or decrease blood glucose levels in diabetic *** SMN improved polyphagia and polydipsia, renal function, and protected the liver and pancreas against protein damage without affecting hyperglycemia in diabetic animals.

Purification,characterization and antiproliferative activity of L-asparaginase from Aspergillus oryzae CCT 3940 with no glutaminase activity

Objective:To explore the anti-proliferative activity of purified L-asparaginase from Aspergillus oryzae CCT 3940(***).Methods:L-asparaginase was produced by submerged fermentation and purified to electrophoresis homogeneity by ionic exchanged chromatography in a fast protein liquid chromatographic *** purified enzyme was characterized and used for the antiproliferative assay against nine tumor cell lines and one non-tumor cell ***:The free glutaminase L-asparaginase was purified 28.6 fold.L-asparaginase showed high stability under physiological condition,remaining stable in the p H range7.0–8.0 after 1 h incubation at temperature range 30–45℃.The Km and Vmax values of purified L-asparaginase were estimated as 0.66 mmol/L and 313 IU/mL,*** purified enzyme could inhibit the growth of a broad range of human tumor cell lines at the concentrations ***,the enzyme from *** CCT 3940 could inhibit tumor growth of leukemia cell line(K562) with a total growth inhibition value of(3.2 ± 2.5) IU/mL and did not inhibit the non-carcinogenic human cell line growth at the concentrations ***:The sensitivity of the cells lines to purified L-asparaginase from *** CCT 3940 appeared to be concentration dependent affording a more significant decrease in cell growth than that observed for the commercial L-asparaginase from Escherichia *** L-asparaginase from *** CCT 3940 has a high potential for pharmaceutical exploitation in the treatment of leukemia.

Injection of bone marrow mesenchymal stem cells by intravenous or intraperitoneal routes is a viable alternative to spinal cord injury treatment in mice

In spite of advances in surgical care and rehabilitation, the consequences of spinal cord injury （SCI） are still challenging. Several experimental therapeutic strategies have been studied in the SCI field, and recent advances have led to the development of therapies that may act on the inhibitory microenvironment. Assorted lineages of stem cells are considered a good treatment for SCI. This study investigated the effect of systemic transplantation of mesenchymal stem cells （MSCs） in a compressive SCI model. Here we present results of the intraperitoneal route, which has not been used previously for MSC administration after compressive SCI. We used adult female C57BL/6 mice that underwent laminectomy at the T9 level, followed by spinal cord compression for 1 minute with a 30-g vascular clip. The animals were divided into five groups： sham （anesthesia and laminectomy but without compression injury induction）, MSC i.p. （intraperitoneal injection of 8×10^5 MSCs in 500 μL of DMEM at 7 days after SCI）, MSC i.v. （intravenous injection of 8 × 10^5 MSCs in 500μL of DMEM at 7 days after SCI）, DMEM i.p. （intraperitoneal injection of 500μL of DMEM at 7 days after SCI）, DMEM i.v. （intravenous injection of 500 μL of DMEM at 7 days after SCI）. The effects of MSCs transplantation in white matter sparing were analyzed by luxol fast blue staining. The number of preserved fibers was counted in semithin sections stained with toluidine blue and the presence of trophic factors was analyzed by immunohistochemistry. In addition, we analyzed the locomotor performance with Basso Mouse Scale and Global Mobility Test. Our results showed white matter preservation and a larger number of preserved fibers in the MSC groups than in the DMEM groups. Furthermore, the MSC groups had higher levels of trophic factors （brain-derived neurotrophic factor, nerve growth factor, neurotrophin-3 and neurotrophin-4） in the spinal cord and improved locomotor performance. Use of SFC in Extraction of Adaptogens from Brazilian Plants

Bioactive substances found in plants are very interesting because of their importance in medicine and foods. Species of Pfaffia genre is common in Brazil, the largest center of collection of this species. Statistics report that 11,000 tons of roots and other plants were exported in 2011. Due to the large market demand, a growth of 10% per year is estimated. One of the promising areas of study involves the supercritical fluid extraction of active compounds present in plants. The current goal is to reduce dependence on foreign markets. The extraction of bioactive compound known as Beta-Ecdysone has been studied. Statistics show that profits of exports coming from the raw material is almost ten times smaller than the extract itself. Considering this fact, we emphasize the importance of an alternative method of industrial extraction in Brazil using supercritical technology, which proves to be environmentally safe.
