Interplay of autophagy and innate immunity in Crohn's disease: A key immunobiologic feature

Crohn's disease representing a clinical phenotype of inflammatory bowel disease is a polygenic immune disorder with complex multifactor etiology. Recent genome-wide association studies of susceptibility loci have highlighted on the importance of the autophagy pathway, which previously had not been implicated in disease pathology. Autophagy represents an evolutionarily highly conserved multi-step process of cellular self-digestion due to sequestration of excessive, damaged, or aged proteins and intracellular organelles in double-membranous vesicles of autophagosomes, terminally self-digested in lysosomes. Autophagy is deeply involved in regulation of cell development and differentiation, survival and senescence, and it also fundamentally affects the inflammatory pathways, as well as the innate and adaptive arms of immune responses. Autophagy is mainly activated due to sensors of the innate immunity, i.e., by pattern recognition receptor signaling. The interplay of genes regulating immune functions is strongly influenced by the environment, especially gut resident microbiota. The basic challenge for intestinal immune recognition is the requirement of a simultaneous delicate balance between tolerance and responsiveness towards microbes. On the basis of autophagy-related risk genetic polymorphisms (ATG16L1, IRGM , NOD2 , XBP1 ) impaired sensing and handling of intracellular bacteria by innate immunity, closely interrelated with the autophagic and unfolded protein pathways seem to be the most relevant immunobiologic events. Autophagy is now widely considered as a key regulator mechanism with the capacity to integrate several aspects of Crohn's disease pathogenesis. In this review, recent advances in the exciting crosstalk of susceptibility coding variants-related autophagy and innate immunity are discussed.

Insights into hepatitis E virus epidemiology in Croatia

＜正＞The snap bean is a significant food crop. Under the Hungarian climate conditions, the supply of raw materials of good quality and appropriate quantity to canning factories can be ensured only with irrigation. The present paper evaluates the result of irrigation experiments carried out on snap beans sown in spring and summer and grown with or without irrigation. The experiments were run over the course of 12 years. The average of 12 years shows that the yield was 2.81 ha-1 for spring sown and 1.91 ha-1 in summer-sown plants without irrigation. The lowest level of profitable production i.e. 5.51 ha-1 was reached twice in the case of spring sowing and only once in the case of summer sowing. Profitable yield production can be ensured only with regular irrigation when the yield may be increased by 4-5 times. In four of the twelve years we determined the canopy surface temperature of snap bean stands with and without irrigation. A Raynger II infrared remote thermometer determined the canopy surface temperature each day at 13.00 hours. The canopy temperature can well characterize the water supply to plant stands. This parameter may be used for describing the degree of drought and the water turnover of plant stands with different water supply. The positive values of foliage-air temperature differences （SDD） numerically express the degree of drought and the water supply of the crops. The results indicated that a 1℃higher SDD value may cause 90-130 kg/ ha yield loss.

BACKGROUND Hepatitis E virus(HEV)is an emerging virus of global health *** seroprevalence rates differ greatly according to geographic region and population *** To analyze the seroprevalence of HEV in exposed(animal-related professions)and nonexposed populations,as well as solid organ and hematopoietic stem cell transplant *** Forestry workers(n=93),hunters(n=74),and veterinarians(n=151)represented the exposed *** general population(n=126)and pregnant women(n=118)constituted the control *** patients included liver transplant recipients(LTRs)(n=83),kidney transplant recipients(KTRs)(n=43),and hematopoietic stem cell transplant recipients(HSCRs)(n=39).HEV immunoglobulin G antibodies were detected using the enzyme-linked immunosorbent assay and confirmed by the immunoblot *** The HEV seroprevalence significantly differed between groups:Veterinarians 15.2%,hunters 14.9%,forestry workers 6.5%,general population 7.1%,and pregnant women 1.7%.In transplant patients,the seropositivity was highest in LTRs(19.3%),while in KTRs and HSCRs,the seroprevalence was similar to the general population(6.9%and 5.1%,respectively).A significant increase in seropositivity with age was observed from 2.9%in individuals less than 30 years to 23.5%in those older than 60 *** characteristics(sex,educational level,area of residence,and number of household members),eating habits(game meat,offal,and pork products consumption),and environmental and housing conditions(drinking water supply,type of water drainage/sewer,waste disposal,domestic animals)were not associated with HEV ***,individuals who reported a pet ownership were more often seropositive compared to those who did not have pet animals(12.5%vs 7.0%).CONCLUSION The results of this study showed that individuals in professional contact with animals and LTRs are at higher risk for HEV *** addition,age is a significant risk factor for HEV

The primary objective to these study was to test NPKCaMg-fertilization induced ＂Zn（2＋）＂ bioavailability in triticale rhizosphere soils. Soil and plant samples were taken from experimental sites randomly, and were analysed in laboratory to plant available Zn^2＋） content. Zinc ＂Zn（2＋）＂ concentrations showed a large variability to interactions with soil＇s pH values. It ranged from 0.4 and 1.36 mg kg1. Soil＇s ＂Zn（2＋）＂ contents had been significantly lower than International Soil Limit （ISL） value （70 mg kgl）, consequently. Plant available ＂Zn（2＋）＂ contents from triticale rhizosphere soils in Leaf＋Straw ranged from 10.1 to 38.4 mg kgl, and in Seed from 26.9 to 52.0 mg kg1. ＂Zn（2＋）＂s Actual Translocation Index （ATI） from rhizosphere soils to Leaf＋Straw had as average 23.6, and to grain 43. 1. ＂Zn（2＋）＂ Leaf＋Straw bioacummulation （g hal） had as average 13.4, Grain 23.2, and total （Leaf＋Straw＋Grain） 36.6. ＂Zn（2＋）＂translocation from soils to Leaf＋Straw was 37% lower than to Grain. Ultimately, within the range of experimental conditions tested these results are present that ＂Zn（2＋）＂ tends to accumulate to triticale organs to create the conditions for toxicity effects in Food Chane.

Combined Treatment with Buserelin+Cabergoline in Patient with Prostate Cancer and Pituitary Macroprolactinoma

Twelve years following hemicolectomy for colon adenocarcinoma, a 75-year-old patient with prostate cancer was treated for 4 weeks with the antiandrogen nilutamide and then with the long-acting GnRH agonist buserelin. The serum testosterone and prostate-specific antigen levels had decreased dramatically after 3 months of treatment. After 2 years of buserelin administration, the hormonal state was examined. Serum estradiol, testosterone, DHEA, DHEAS, FSH and LH levels proved to be suppressed, but the serum PRL concentration was extremely high (3 365 mIU/l). The pituitary MRI revealed a macroadenoma. The patient was treated with the dopamine agonist cabergoline, together with buserelin. After 9 months of this combined treatment, the prostate-specific antigen and testosterone levels were very low;the serum estradiol, DHEA, DHEAS, FSH and LH concentrations remained suppressed. The serum PRL level fell dramatically to 6.95 mIU/l, and a significant reduction in tumor size was observed on MRI. In conclusion: Combined buserelin + cabergoline treatment proved a highly successful procedure to cure this patient with prostate carcinoma and subsequent pituitary macroprolactinoma.

Automotive exhaust gas contains high concentrations of NOx, CO, and hydrocarbons, which are harmful to human health. Although these components can be reduced drastically by the use of a three-way catalyst, nitrous oxide, N2O, which has a strong greenhouse effect, is produced during the catalytic reaction. The present work is aimed at developing a novel family of catalysts, based on gold and silver, to replace the more expensive platinum and rhodium metals.

To assess the prevalence of a panel of serologic markers that reflect gut barrier dysfunction in a mixed cohort of pediatric and adult primary sclerosing cholangitis (PSC) patients. METHODSSera of 67 PSC patients [median age (range): 32 (5-79) years, concomitant IBD: 67% and cirrhosis: 20%] were assayed for the presence of antibodies against to F-actin (AAA IgA/IgG) and gliadin (AGA IgA/IgG)] and for serum level of intestinal fatty acid-binding protein (I-FABP) by ELISA. Markers of lipopolysaccharide (LPS) exposure [LPS binding protein (LBP)] and various anti-microbial antibodies [anti-OMP Plus IgA and endotoxin core IgA antibody (EndoCAb)] were also determined. Poor disease outcome was defined as orthotopic liver transplantation and/or liver-related death during the follow-up [median: 99 (14-106) mo]. One hundred and fifty-three healthy subjects (HCONT) and 172 ulcerative colitis (UC) patients were the controls. RESULTSA total of 28.4%, 28.0%, 9% and 20.9% of PSC patients were positive for AAA IgA, AAA IgG, AGA IgA and AGA IgG, respectively. Frequencies of AAA IgA and AAA IgG (P AAA IgA posvsneg: 365 vs 166 pg/mL, P = 0.011), but not with serum LBP level.
