Culture Optimization for Mass Production of <i>Rhizobium</i>Using Bioreactor Made of Readily Available Materials and Agitated by Air Flow

Large quantities of Rhizobium leguminosarum by conventional technique are a time-consuming and labor-intensive method of production. The selection of different types of nutrient media for the production of large amounts of rhizobium at low cost, determination of environmental regulators of culture medium and the effect of different levels of air flow into nutrition media have been investigated. Rhizobium cultured in modified air lift bioreactor as improved conventional system using Yeast Extract Mannitol Broth by assessing aeration and agitation in their submerged growth. For selection of suitable nutrient media for Rhizobium before mass production, YMA (Yeast Mannitol Agar), YSA (Yeast Sucrose Agar), YMA + bromthymol blue (BTB), YMA + Congo red (CR) medium was tested. To optimize the growing conditions of different temperatures, pH levels and salt strengths were investigated. Three air flow speeds at the rate of 0.01, 0.05 and 0.1 VVM for oxygen supply were ensured. A thick off-white bacterial colony was observed on the YMA plate among different culture media. Room temperature (25˚C - 30˚C), neutral pH (7.0), low salinity (1%) was observed to be suitable among the environmental conditions for Rhizobium mass production. The maximum density of Rhizobium was observed using air supply of 0.1 VVM level in bioreactor system. The result seemed that the air supply and agitation was an important growth factor in submerged cultivation. This study provides useful information on large scale production of Rhizobium of interest employing low cost effective modified air lift bioreactor within less time.

In this paper,we propose a novel Local Macroscopic Conservative(LoMaC)low rank tensor method with discontinuous Galerkin(DG)discretization for the physical and phase spaces for simulating the Vlasov-Poisson(VP)*** LoMaC property refers to the exact local conservation of macroscopic mass,momentum,and energy at the discrete *** recently developed LoMaC low rank tensor algorithm(arXiv:2207.00518)simultaneously evolves the macroscopic conservation laws of mass,momentum,and energy using the kinetic flux vector splitting;then the LoMaC property is realized by projecting the low rank kinetic solution onto a subspace that shares the same macroscopic *** paper is a generalization of our previous work,but with DG discretization to take advantage of its compactness and flexibility in handling boundary conditions and its superior accuracy in the long *** algorithm is developed in a similar fashion as that for a finite difference scheme,by observing that the DG method can be viewed equivalently in a nodal *** the nodal DG method,assuming a tensorized computational grid,one will be able to(i)derive differentiation matrices for different nodal points based on a DG upwind discretization of transport terms,and(ii)define a weighted inner product space based on the nodal DG grid *** algorithm can be extended to the high dimensional problems by hierarchical Tucker(HT)decomposition of solution tensors and a corresponding conservative projection *** a similar spirit,the algorithm can be extended to DG methods on nodal points of an unstructured mesh,or to other types of discretization,e.g.,the spectral method in velocity *** numerical results are performed to showcase the efficacy of the method.

Synergistic enhancement of photocatalytic and antimicrobial efficacy of nitrogen and erbium co-doped ZnO nanoparticles

Using the chemical co-precipitation approach,a series of nitrogen(N) and erbium(Er) co-doped ZnO nanoparticles(NPs) was effectively synthesized to enhance the photocatalytic and antibacterial *** characterization techniques,including X-ray diffraction(XRD),X-ray photoelectron spectro scopy(XPS),scanning electron microscopy(SEM),UV-vis,and photoluminescence(PL) spectroscopy,were carried out to validate the evaluated photocatalytic and antibacterial *** analysis confirms the pure wurtzite ZnO phase without the presence of any secondary *** analysis confirms the succe ssful incorporation of nitrogen and erbium into the ZnO *** optical bandgap of ZnO calculated from UV-vis spectroscopy shows a redshift after Er-N co-doping,with the lowest bandgap of 3.215 eV calculated for Zn_(0.97)Er_(0.03)N_(0.01)O_(0.99) *** images demonstrate the formation of nanorods after N-Er co-doping,followed by gradually increase d rod diameter and length after N-Er ***,the photocatalytic activities of ZnO samples we re measured by their ability to facilitate the photodegradation of Rhodamine B under UV *** with 1 mol% N doping exhibits 88% photodegradation of RhB under UV light within 360 min,and the photodegradation and antibacterial activity are greatly improved with Er *** fact,3 mol% Er-1 mol% N doped ZnO NPs demonstrate the highest photocatalytic activity,with approximately 96% degradation after 360 min,as well as superior antibacterial activity against Staphylococcus aureus(Gram-positive bacteria) and Pseudomonas aeruginosa(Gram-negative bacteria) with the highest zone of inhibition(ZOI) of 16 nm,due to nanorod formation,increased reactive oxygen species(ROS),and decreased electron-hole recombination,as validated by SEM,XPS,and PL spectroscopy.

Multi-strain probiotics provide the most dependable approaches to improve health,immune response,and disease resistance in farmed *** the present study,we examined the effects of multi-species probiotics on the survival,growth,immune response,and disease resistance of rohu(Labeo rohita)*** hatched larvae from the day of first feeding(average weight of 0.003 g)were reared with multi-species probiotics having a combination of Bacillus subtilis(109 colony forming units(cfu)/mL)and Lactobacillus spp.(Lactobacillus plantarum,Lactobacillus buchneri-1011 cfu/mL)in water containing doses of 0(control-C),0.5 mL/L(treatment 1-T1),and 1.0 mL/L(treatment 2-T2)in triplicates for 90 *** the experiment,a challenge test was performed to assess the fish's resistance to pathogenic Aeromonas *** higher survival was recorded in larvae of treated groups(87%in T2 and 79%in T1)compared to the control(62%).Significantly higher growth performance(weight gain and specific growth rate—SGR)was shown by the probiotic-treated larval groups compared to the *** supplementation resulted in significantly higher counts of total viable colony(TVC)and lactic acid bacteria(LAB)in the *** immunological parameters(mucosal fold fattening,goblet cell abundance,expansion of lamina propria and enterocytes)of the gut were significantly better in probiotic-treated *** liver of treated fish showed irregular shape nuclei turning into regular shape and reducing spaces between the hepatic ***-treated fish had the highest post-challenge survival rate(90%)against *** *** erythrocytes of challenged fish treated with probiotics had significantly lower frequencies of various nuclear and cellular *** findings suggest that multi-species probiotic supplements could improve the survival,growth,health status,and immune response of rohu in the early stages of its development.

Macroplastics on Soil-Plant System: Inhibiting Effects of Macroplastics on the Growth of Green Amaranth (<i>Amaranthus viridis</i>)

In recent time Bangladesh faces a serious problem of soil pollution due to plastic contamination. However, the degree of the extent to which the effects of plastics on plant growth occur is not properly identified. An experiment was conducted to measure the effects of mixed plastic (polyethylene and disposable plastic glass) on the growth of Amaranthus viridis. Different doses of mixed plastics (T0, T1, T2, and T3) were applied with a fixed amount of soil for each of the treatments e.g., T0 (control), T1 (10 gm mixed plastics/3kg soil), T2 (15 gm mixed plastics/3kg soil) and T3 (20 gm mixed plastic/3kg soil), and the growth response of Amaranthus viridis against plastic was observed for six consecutive weeks. The growth was measured in terms of plant height and girth diameter. The results showed that the presence of mixed plastic had a significant effect on the growth of Amaranthus viridis and particularly in treatment T3 (3 kg soil/20gm mixed plastic), the plants showed a slower growth response compared to control and the rest of the treatments applied in case of both plant height as well as girth diameter. The statistical analysis (one-way Analysis of Variance) also proved the significance of the treatments (p-values < 0.05) for six consecutive weeks. The experiment was successfully able to set an index on which plastics had their effects on the growth of green amaranth. In addition, the obtained data will be helpful in future research of the study in determining the possible effects of plastic on plant growth viz. green amaranth.

Geochemistry of Tertiary sandstones from southwest Sarawak,Malaysia: implications for provenance and tectonic setting

Tertiary sandstones collected from southwest Sarawak, Malaysia, were analyzed to decipher their provenance, weathering, and tectonic setting. The studied sandstones have a sublitharenite composition and are dominantly composed quartz with little mica and feldspar, and a small amount of volcanic fragments. These sand- stones were generally derived from quartz-rich recycled orogenic sources. They have relatively high SiO2 content with low Na20, CaO, MnO, and MgO contents. Values of Chemical Index of Alteration （CIA） of these rock samples vary from 71 to 93, with an average of 81, implying intense chemical alteration during weathering. A felsic igneous source is suggested by a low concentration of TiO2 com- pared to CIA, enrichment of Light Rare Earth Elements, depletion of Heavy Rare Earth Elements, and negative Eu anomalies. A felsic origin is further supported by a Eu/Eu＊ range of 0.65-0.85 and high Th/Sc, La/Sc, La/Co, and Th/ Co ratios. This work presents the first reported geochemical data of Tertiary sandstones of the Sarawak Basin. These data led us to conclude that the sandstones were dislodged from recycled orogenic sources and deposited in a slowly subsiding rifted basin in a passive continental tectonic setting.

A Comparative Study of Pollutant Removal Stability in Hybrid Wetland Columns

This study investigated the effects of two alternative substrates(wood mulch and zeolite) on the performance of three laboratory-scale hybrid wetland systems that had identical system components and *** system consisted of a vertical flow(VF) wetland column,followed by a horizontal flow(HF) column and a vertical flow *** substrates employed were wood mulch,gravel and zeolite,and Phragmites australis were planted in each *** systems received synthetic wastewater,with pollutant loadings in the range of 8.5-38.0 g/(m2·d) total nitrogen(TN) and 4.0-46.4 g/(m2·d) biological oxygen demand(BOD5).Wood mulch and zeolite substrates showed higher efficiencies in terms of removing nitrogenous compounds and biodegradable *** supply of organic carbon from the organic mulch substrates enhanced denitrification,while adsorption of influent ammoniacal nitrogen(NH4-N) in zeolite played a major role in the removal of nitrogenous species in the wetland ***,the average percentage removals of TN and BOD5 reached >66% and >96% respectively,indicating stable performances by the hybrid wetland systems under the experimental loading *** models were developed,based on the combination of Monod kinetics and continuously-stirred tank reactor(CSTR) flow patterns to describe the degradation of nitrogenous *** by the models closely matched the experimental data,indicating the validity and potential application of Monod kinetics in the modelling and design of treatment wetlands.
