Background Culex pipiens pallens andCulex pipiens quinquefasciatus are the dominant species ofCulex mosquitoes in China and important disease vectors. Long-term use of insecticides can cause mutations in the voltage-gated sodium channel (vgsc) gene of mosquitoes, but little is known about the current status and evolutionary origins ofvgsc gene in different geographic populations. Therefore, this study aimed to determine the current status ofvgsc genes inCx. p. pallens andCx. p. quinquefasciatus in China and to investigate the evolutionary inheritance of neighboring downstream introns of thevgsc gene to determine the impact of insecticides on long-term evolution.Methods Sampling was conducted from July to September 2021 in representative habitats of 22 provincial-level administrative divisions in China. Genomic DNA was extracted from 1308 mosquitoes, the IIS6 fragment of thevgsc gene on the nerve cell membrane was amplified using polymerase chain reaction, and the sequence was used to evaluate allele frequency and knockdown resistance (kdr) frequency. MEGA 11 was used to construct neighborjoining (NJ) tree. PopART was used to build a TCS network.Results There were 6 alleles and 6 genotypes at the L1014 locus, which included the wild-type alleles TTA/L and CTA/L and the mutant alleles TTT/F, TTC/F, TCT/S and TCA/S. The geographic populations with akdr frequency less than 20.00% were mainly concentrated in the regions north of 38° N, and the geographic populations with akdr frequency greater than 80.00% were concentrated in the regions south of 30° N.kdr frequency increased with decreasing latitude. And within the same latitude, the frequency ofkdr in large cities is relatively high. Mutations were correlated with the number of introns. The mutant allele TCA/S has only one intron, the mutant allele TTT/F has three introns, and the wild-type allele TTA/L has 17 introns.Conclusions Cx. p. pallens andCx. p. quinquefasciatus have developed resistance to insecticides in most regions of China. Background The current prevention and control strategy forAedes albopictus heavily relies on comprehensive management, such as environmental management and chemical control. However, the wide application of pyrethroids has facilitated the development of insecticide resistance, primarily via mutations in the voltage-gated sodium channel (VGSC) gene. This study aims to develop a novel strategy for detecting mutations in the VGSC gene inAe. albopictus using multiplex PCR-mass spectrometry (MPCR-MS) minisequencing technology.Methods We established a new strategy for detecting mutations in the VGSC gene inAe. albopictus using MPCR-MS minisequencing technology. MPCR amplification and mass probe extension (MPE) were first used, followed by single nucleotide polymorphism (SNP) typing mass spectrometry, which allows the simultaneous detection of multiple mutation sites of the VGSC gene in 96 samples ofAe. albopictus. A total of 70 wild-collectedAe. albopictus were used to evaluate the performance of the method by comparing it with other methods.Results Three target sites (1016, 1532, 1534) in the VGSC gene can be detected simultaneously by double PCR amplification combined with matrix-assisted laser desorption ionization-time-of-flight mass spectrometry, achieving a detection limit of 20 fg/μl. We applied this method to 70 wild-collectedAe. albopictus, and the obtained genotypes were consistent with the routine sequencing results, suggesting the accuracy of our method.Conclusions MPCR-MS minisequencing technology provides a sensitive and high-throughput approach toAe. albopictus VGSC gene mutation screening. Compared with conventional sequencing, this method is economical and time-saving. It is of great value for insecticide resistance surveillance in areas with a high risk of vector-borne disease.

Introduction:Mosquito-borne flavivirus can lead to serious infectious diseases worldwide and cause high mortality and disability.In order to strengthen epidemiological investigation of flavivirus and meet the needs of early warning of diseases,a simple,rapid,and sensitive detection method needs to be established to prevent and control mosquito-borne diseases.Methods:Using the NS5 gene of flavivirus in GenBank,3 universal primers targeting the conserved regions were designed.The complementary DNAs(cDNAs)of Japanese encephalitis virus(JEV),dengue virus(DENV),West Nile virus(WNV),and yellow fever virus(YFV)were used as the template to optimize the reaction conditions.A heminested reversetranscriptase polymerase chain reaction(hnRT-PCR)was established to verify the sensitivity and specificity of this method and to detect field-caught mosquitoes.Results:Our results showed that this method exhibited better specificity,higher sensitivity,and the ability to detect multiple viruses simultaneously.The lowest detection limit of JEV,DENV-2,YFV,and WNV was 3×10^(4),3×10^(6),3×10^(5),and 3×10^(4)copies/μL,respectively.Mosquito-borne flavivirus was successfully detected in the field-caught mosquito samples using the method established in this study.Discussion:The hnRT-PCR method established in this study can be employed for the rapid detection of flavivirus and provide technical support for early and rapid diagnosis of mosquito-borne flavivirus.

Epidemiological characteristics and temporal-spatial analysis of overseas imported dengue fever cases in outbreak provinces of China, 2005–2019

Background Overseas imported dengue fever is an important factor in local outbreaks of this disease in the mainland of China.To better prevent and control such local outbreaks,the epidemiological characteristics and temporal-spatial distribution of overseas imported dengue fever cases in provincial-level administrative divisions(PLADs)where dengue fever is outbreak in the mainland of China were explored.Methods Using the Chinese National Notifiable Infectious Disease Reporting Information System(CNNDS),we identified overseas imported dengue fever cases in dengue fever outbreak areas in the mainland of China from 2005 to 2019 to draw the epidemic curve and population characteristic distribution of overseas imported cases in each PLAD.Based on spatial autocorrelation analysis of ArcGIS 10.5 and temporal-spatial scanning analysis of SaTScan 9.5,we analyzed the temporal-spatial distribution of overseas imported dengue fever in dengue fever outbreak areas in the mainland of China.Results A total of 11,407 imported cases,mainly from Southeast Asia,were recorded from 2005 to 2019 in these 13 PLADs.Of which 62.1%were imported into Yunnan and Guangdong Provinces.Among the imported cases,there were more males than females,mainly from the 21–50 age group.The hot spots were concentrated in parts of Yunnan,Guangdong and Fujian Provinces.We found the cluster of infected areas were expanding northward.Conclusions Based on the analysis of overseas imported dengue cases in 13 PLADs of the mainland of China from 2005 to 2019,we obtained the epidemiological characteristics and spatial distribution of imported dengue cases.Border controls need to pay attention to key population sectors,such as 21–50 years old men and education of key populations on dengue prevention.There is a need to improve the awareness of the prevention and control of imported cases in border areas.At the same time,northern regions cannot relax their vigilance.

Vermicompost:In situ retardant of antibiotic resistome accumulation in cropland soils

The dissemination of antibiotic resistance genes(ARGs)in soil has become a global environmental issue.Vermicomposting is gaining prominence in agricultural practices as a soil amendment to improve soil quality.However,its impact on soil ARGs remains unclear when it occurs in farmland.We comprehensively explored the evolution and fate of ARGs and their hosts in the field soil profiles under vermicompost application for more than 3years.Vermicompost application increased several ARG loads in soil environment but decreased the high-risk bla-ARGs(bla_(ampC),bla_(NDM),and bla_(GES-1))by log(0.04-0.43).ARGs in soil amended with vermicompost primarily occurred in topsoil(approximately 1.04-fold of unfertilized soil),but it is worth noting that their levels in the 40-60 cm soil layer were the same or even less than in the unfertilized soil.The microbial community structure changed in soil profiles after vermicompost application.Vermicompost application altered the microbial community structure in soil profiles,showing that the dominant bacteria(i.e.,Proteobacteria,Actinobacteriota,Firmicutes)were decreased 2.62%-5.48%with the increase of soil depth.A network analysis further revealed that most of ARG dominant host bacteria did not migrate from surface soil to deep soil.In particular,those host bacteria harboring high-risk bla-ARGs were primarily concentrated in the surface soil.This study highlights a lower risk of the propagation of ARGs caused by vermicompost application and provides a novel approach to reduce and relieve the dissemination of ARGs derived from animals in agricultural production.

Risk factors for postpartum posttraumatic stress disorder after emergency admission

BACKGROUND:Postpartum posttraumatic stress disorder(PTSD)can occur in women who give birth after emergency admission.The identification of risk factors for this condition is crucial for developing effective preventive measures.This retrospective study aimed to explore the incidence and risk factors for postpartum PTSD in women who give birth after emergency admission.METHODS:Medical records of women who gave birth after emergency admission were collected between March 2021 and April 2023.The patients’general conditions and perinatal clinical indicators were recorded.The puerperae were divided into PTSD group and control group based on symptom occurrence at six weeks postpartum.Multivariate logistic regression analysis was performed to identify risk factors.RESULTS:A total of 276 puerperae were included,with a PTSD incidence of 20.3% at six weeks postpartum.Multivariate logistic regression analysis identified emergency cesarean section(odds ratio[OR]=2.102;95%confidence interval[CI]:1.114-3.966,P=0.022),admission to the emergency department after midnight(12:00 AM)(OR=2.245;95%CI:1.170-4.305,P<0.001),and cervical dilation(OR=3.203;95%CI:1.670–6.141,P=0.039)as independent risk factors for postpartum PTSD.Analgesia pump use(OR=0.500;95%CI:0.259–0.966,P=0.015)was found to be a protective factor against postpartum PTSD.CONCLUSION:Emergency cesarean section,admission to the emergency department after midnight,and cervical dilation were identified as independent risk factors for postpartum PTSD,while analgesic pump use was a protective factor.These findings provide insights for developing more effective preventive measures for women who give birth after emergency admission.

Osteosarcoma is the most common malignant bone tumor affecting children and adolescents.Currently,the most common treatment is surgery combined with neoadjuvant chemotherapy.Although the survival rate of patients with osteosarcoma has improved in recent years,it remains poor when the tumor(s)progress and distant metastases develop.Therefore,better animal models that more accurately replicate the natural progression of the disease are needed to develop improved prognostic and diagnostic markers,as well as targeted therapies for both primary and metastatic osteosarcoma.The present review described animal models currently being used in research investigating osteosarcoma,and their characteristics,advantages,and disadvantages.These models may help elucidate the pathogenic mechanism(s)of osteosarcoma and provide evidence to support and develop clinical treatment strategies.

Sequential neoadjuvant chemotherapy using pegylated liposomal doxorubicin and cyclophosphamide followed by taxanes with complete trastuzumab and pertuzumab treatment for HER2-positive breast cancer: A phase Ⅱ single-arm study

Objective: Despite cardiotoxicity overlap, the trastuzumab/pertuzumab and anthracycline combination remains crucial due to significant benefits. Pegylated liposomal doxorubicin(PLD), a less cardiotoxic anthracycline, was evaluated for efficacy and cardiac safety when combined with cyclophosphamide and followed by taxanes with trastuzumab/pertuzumab in human epidermal growth factor receptor-2(HER2)-positive early breast cancer(BC).Methods: In this multicenter, phase II study, patients with confirmed HER2-positive early BC received four cycles of PLD(30-35 mg/m^(2)) and cyclophosphamide(600 mg/m^(2)), followed by four cycles of taxanes(docetaxel,90-100 mg/m^(2) or nab-paclitaxel, 260 mg/m^(2)), concomitant with eight cycles of trastuzumab(8 mg/kg loading dose,then 6 mg/kg) and pertuzumab(840 mg loading dose, then 420 mg) every 3 weeks. The primary endpoint was total pathological complete response(tp CR, yp T0/is yp N0). Secondary endpoints included breast p CR(bp CR),objective response rate(ORR), disease control rate, rate of breast-conserving surgery(BCS), and safety(with a focus on cardiotoxicity).Results: Between May 27, 2020 and May 11, 2022, 78 patients were treated with surgery, 42(53.8%) of whom had BCS. After neoadjuvant therapy, 47 [60.3%, 95% confidence interval(95% CI), 48.5%-71.2%] patients achieved tp CR, and 49(62.8%) achieved bp CR. ORRs were 76.9%(95% CI, 66.0%-85.7%) and 93.6%(95% CI,85.7%-97.9%) after 4-cycle and 8-cycle neoadjuvant therapy, respectively. Nine(11.5%) patients experienced asymptomatic left ventricular ejection fraction(LVEF) reductions of ≥10% from baseline, all with a minimum value of >55%. No treatment-related abnormal cardiac function changes were observed in mean N-terminal pro-BNP(NT-pro BNP), troponin I, or high-sensitivity troponin.Conclusions: This dual HER2-blockade with sequential polychemotherapy showed promising activity with rapid tumor regression in HER2-positive BC. Importantly, this regimen showed an acceptable safety profile,especially a low risk of cardiac events,
