Monotropein Induced Apoptosis and Suppressed Cell Cycle Progression in Colorectal Cancer Cells

Objective: To determine whether monotropein has an anticancer effect and explore its potential mechanisms against colorectal cancer(CRC) through network pharmacology and molecular docking combined with experimental verification. Methods: Network pharmacology and molecular docking were used to predict potential targets of monotropein against CRC. Cell counting kit assay, plate monoclonal assay and microscopic observation were used to investigate the antiproliferative effects of monotropein on CRC cells HCT116, HT29 and LoVo. Flow cytometry and scratch assay were used to analyze apoptosis and cell cycle, as well as cell migration, respectively in HCT116, HT29, and LoVo cells. Western blotting was used to detect the expression of proteins related to apoptosis, cell cycle, and cell migration, and the expression of proteins key to the Akt pathway. Results: The Gene Ontology and Reactome enrichment analyses indicated that the anticancer potential of monotropein against CRC might be involved in multiple cancer-related signaling pathways. Among these pathways, RAC-beta serine/threonine-protein kinase(Akt1, Akt2), cyclin-dependent kinase 6(CDK6), matrix metalloproteinase-9(MMP9), epidermal growth factor receptor(EGFR), cell division control protein 42 homolog(CDC42) were shown as the potential anticancer targets of monotropein against CRC. Molecular docking suggested that monotropein may interact with the 6 targets(Akt1, Akt2, CDK6, MMP9, EGFR, CDC42). Subsequently, cell activity of HCT116, HT29 and LoVo cell lines were significantly suppressed by monotropein(P<0.05). Furthermore, our research revealed that monotropein induced cell apoptosis by inhibiting Bcl-2 and increasing Bax, induced G_1–S cycle arrest in colorectal cancer by decreasing the expressions of CyclinD1, CDK4 and CDK6, inhibited cell migration by suppressing the expressions of CDC42 and MMP9(P<0.05), and might play an anticancer role through Akt signaling pathway. Bright InP quantum dots by Ga-doping for red emitters

Environment-friendly indium phosphide(InP)-based quantum dots(QDs)with efficient red-emitting properties are sufficiently needed to satisfy the requirement of burgeoning display and lighting ***,the syntheses of InP QDs using tris(trimethylsilyl)phosphine as the precursor are highly toxic and ***,we successfully introduced gallium(Ga)ions into tris(dimethylamino)phosphine-based red InP cores through thermally-promoted cation exchange,and the obtained Ga-doped InP cores exhibited significantly increased photoluminescence quantum yields(PLQY)of up to 26%.The existence of Ga was directly confirmed by energy-dispersive X-ray spectroscopy and X-ray photoelectron spectroscopy,and the functions of Ga were systematically *** subsequent coating of Ga-doped InP cores with ZnSeS and ZnS shells,the resulting Ga-InP/ZnSeS/ZnS QDs achieved a high PLQY of 62%with an emission maximum at 640 *** contrast,without Ga-doping,the PLQY only attained 36%using the same synthetic *** indicated an approximate 1.7-fold increase in *** enhancement of photoluminescence was related to the Ga^(3+),as it not only passivated surface defects of InP cores but also reduced core–shell interface *** Ga-InP/ZnSeS/ZnS QDs exhibited good stability towards heat treatment and ultraviolet(UV)***,the red light-emitting diode(LED)based on Ga-InP/ZnSeS/ZnS QDs performed well in a wide injected current range of 2 to 200 mA,with a maximum power efficiency of 0.68 lm/*** work showcases Ga-doping through cation exchange as a promising strategy for enhancing the efficiency of InP-based red emitters.

Design and fabrication of compound varifocal lens driven by polydimethylsiloxane film elastic deformation

A compound varifocal lens based on electromagnetic drive technology is designed and fabricated, where the polydimethylsiloxane(PDMS) film acts as a driving component, while the PDMS biconvex lens and the plane-concave lens form a coaxial compound lens system. The plane-concave lens equipped with driving coils is installed directly above the PDMS lens surrounded by the annular magnet. When different currents are applied, the annular magnet moves up and down, driving the PDMS film to undergo elastic deformation, and then resulting in longitudinal movement of the PDMS lens. The position change of the PDMS lens changes the focal length of the compound lens system. To verify the feasibility and practicability of this design, a prototype of our compound lens system is fabricated in experiment. Our proposed compound lens shows that its zoom ability reaches 9.28 mm when the current ranges from -0.20 A to 0.21 A.

Field test of high-power microwave-assisted mechanical excavation for deep hard iron ore

Quasi-in-situ Observation and SKPFM Studies on Phosphate Protective Film and Surface Micro-Galvanic Corrosion in Biological Mg-3Zn-xNd Alloys

The phosphate protective film and micro-galvanic corrosion of biological Mg-3Zn-xNd (x = 0, 0.6, 1.2) alloys were investigated by scanning and transmission electron microscopy, quasi-in-situ observation, scanning Kelvin probe force microscopy (SKPFM) and electrochemical tests. The results revealed the Mg-Zn-Nd phases formed in Mg-3Zn alloy contained with Nd. Adding Nd resulted in a significant decline in the cracks of the phosphate protective film and micro-galvanic corrosion of alloys, which were recorded by quasi-in-situ observation. In addition, the Volta potential difference of Mg-Zn-Nd/α-Mg (~ 188 mV) was lower than MgZn/α-Mg (~ 419 mV) and Zn-rich/α-Mg (~ 260 mV), and the corrosion rates of alloys markedly decreased after the addition of 0.6 wt% Nd. The improvement in corrosion resistance of Nd-containing alloys was mainly attributed to the following: (i) the addition of Nd reduced the Volta potential difference (second phases/α-Mg);(ii) the phosphate protective film containing Nd_(2)O_(3) deposited on the surface of the alloys, effectively preventing the penetration of harmful anions.

p38 MAPK inhibitor SB202190 suppresses ferroptosis in the glutamate-induced retinal excitotoxicity glaucoma model

Glutamate excitotoxicity has been shown to play an important role in glaucoma, and glutamate can induce ferroptosis. The p38 mitogenactivated protein kinase(MAPK) pathway inhibitor SB202190 has a potential ability to suppress ferroptosis, and its downstream targets, such as p53, have been shown to be associated with ferroptosis. However, whether ferroptosis also occurs in retinal ganglion cells in response to glutamate excitotoxicity and whether inhibition of ferroptosis reduces the loss of retinal ganglion cells induced by glutamate excitotoxicity remain unclear. This study investigated ferroptosis in a glutamate-induced glaucoma rat model and explored the effects and molecular mechanisms of SB202190 on retinal ganglion cells. A glutamate-induced excitotoxicity model in R28 cells and an N-methyl-D-aspartate-induced glaucoma model in rats were used. In vitro experiments showed that glutamate induced the accumulation of iron and lipid peroxide and morphological changes of mitochondria in R28 cells, and SB202190 inhibited these changes. Glutamate induced the levels of p-p38 MAPK/p38 MAPK and SAT1 and decreased the expression levels of ferritin light chain, SLC7A11, and GPX4. SB202190 inhibited the expression of iron death-related proteins induced by glutamate. In vivo experiments showed that SB202190 attenuated N-methyl-D-aspartate-induced damage to rat retinal ganglion cells and improved visual function. These results suggest that SB202190 can inhibit ferroptosis and protect retinal ganglion cells by regulating ferritin light chain, SAT1, and SLC7A11/Gpx4 pathways and may represent a potential retina protectant.

The Ailaoshan Orogen in the southeastern Tibet Plateau,situated between the Yangtze and Simao blocks,underwent a complex structural,magmatic,and metamorphic evolution resulting in different tectonic subzones with varying structural lineaments and elemental *** elements can conceal or reduce anomalies due to the mutual effect between different anomaly *** the whole zone into subzones based on tectonic settings,ore cluster areas,or sample catchment basins(Scb),geochemical and structural anomalies associated with gold(Au)mineralization have been identified utilizing mean plus twice standard deviations(Mean+2STD),factor analysis(FA),concentration-area(CA)modeling of stream sediment geochemical data,and lineament density in both the Ailaoshan Orogen and the individual *** FA in the divided 98 Scbs with 6 Scbs containing Au deposits can roughly ascertain unknown rock types,identify specific element associations of known rocks and discern the porphyry or skarn-type Au *** with methods of Mean+2STD and C-A model of data in the whole orogen,which mistake the anomalies as background or act the background as anomalies,the combined methods of FA and C-A in the separate subzones or Scbs works well in regional metallogenic potential *** of lineament densities with a 10-km circle diameter is not suitable to locate Au deposits because of the delineated large areas of medium-high lineament *** contrast,the use of circle diameters of 1.3 km or 1.7 km in the ore cluster scale delineates areas with a higher concentration of lineament density,consistent with the locations of known Au *** analyzing the map of faults and Au anomalies,two potential prospecting targets,Scbs 1 and 63 with a sandstone as a potential host rock for Au,have been identified in the Ailaoshan *** use of combined methods in the divided subzones proved to be more effective in improving geological understanding and identifying
