Fire-needle acupuncture for upper limb spastic paralysis after stroke:Study protocol for a randomized controlled trial

Background: Fire-needle acupuncture, an important kind of acupuncture therapy, has been clinically used to treat upper limb spastic paralysis(ULSP) after stroke. Clinical experience has indicated that fire-needle acupuncture treatment takes less time, requires fewer visits, and has more rapid results and fewer side effects compared to chemical medicine alternatives. This study will evaluate the effects of fire-needle acupuncture for ULSP in the context of standardized clinical research and provide high-quality data to inform clinical procedures and future study ***/Design: A randomized controlled trial will be carried out to evaluate the effects of fire-needle acupuncture therapy in patients with ULSP from stroke. ULSP patients(n = 120) will be recruited at Changhai Hospital in Shanghai, China. Patients will be randomly divided into three groups, including fire-needle acupuncture group(FAG), filiform-needle acupuncture group(FFAG) and rehabilitation treatment group(RTG). During the 3-week treatment, the FAG will be treated every two days, while FFAG and RTG will be treated 5 d in a row and then rest for 2 d. The Simplified Fugl-Meyer Motor Function Scale and Modified Ashworth Scale will be used as the primary outcome measures. Statistical analysis will be conducted by an independent ***: Through this study, the utility of fire-needle acupuncture in treating ULSP after stroke will be tested, and some specific claims of fire-needle acupuncture therapy will be evaluated, such as relieving spasm and muscular tension, improving activities of daily living, rapidity of response and less frequency of treatment compared with other treatments.

A new method for constructing interpolating Loop subdivision surfaces is presented. The new method is an extension of the progressive interpolation technique for B-splines. Given a triangular mesh M, the idea is to iteratively upgrade the vertices of M to generate a new control mesh M such that limit surface of M would interpolate M. It can be shown that the iterative process is convergent for Loop subdivision surfaces. Hence, the method is well-defined. The new method has the advantages of both a local method and a global method, i.e., it can handle meshes of any size and any topology while generating smooth interpolating subdivision surfaces that faithfully resemble the shape of the given meshes. The meshes considered here can be open or closed.

AIM: To investigate the effect of vascular endothelial growth factor (VEGF) transfection on hepatic sinusoidal capillarization. METHODS: Enhanced green fluorescent protein (EGFP)/ VEGF transfection was confirmed by immunofluorescence microscopy and immunohistoche-mistry both in primary hepatocytes and in normal liver. Cirrhotic rats were generated by thioacetamide (TAA) administration and then divided into a treatment group, which received injections of 400 μg of plasmid DNA encoding an EGFP- VEGF fusion protein, and a blank group, which received an equal amount of normal saline through the portal vein. The portal vein pressure was measured in the normal and cirrhotic state, in treated and blank groups. The average number of fenestrae per hepatic sinusoid was determined using transmission electron microscopy (TEM), while the relative abundance of VEGF transcripts was examined by Gene array. RESULTS: Green fluorescent protein was observed in the cytoplasms of liver cells under immunofluorescence microscopy 24 h after transfection with EGFP/VEGF plasmid in vitro. Staining with polyclonal antibodies against VEGF illustrated that hepatocytes expressedimmunodetectable VEGF both in vitro and in vitro. There were significant differences in the number of fenestrae and portal vein pressures between normal and cirrhotic rats (7.40 ± 1.71 vs 2.30 ± 1.16 and 9.32 ± 0.85 cmH2O vs 17.92 ± 0.90 cmH2O, P < 0.01), between cirrhotic and treated rats (2.30 ± 1.16 cmH2O vs 4.60 ± 1.65 and 17.92 ± 0.90 cmH2O vs 15.52 ± 0.93 cmH2O, P < 0.05) and between the treatment group and the blank group (4.60 ± 1.65 cmH2O vs 2.10 ± 1.10 cmH2O and 15.52 ± 0.93 cmH2O vs 17.26 ± 1.80 cmH2O, P < 0.05). Purpose:Urosepsis in adults comprises approximately 25% of all sepsis cases, and is due to complicated urinary tract infections in most cases. However, its mechanism is not fully clarified. Urosepsis is a very complicated disease with no effective strategy for early diagnosis and treatment. This study aimed to identify possible target-related proteins involved in urosepsis using proteomics and establish possible networks using ***:Fifty patients admitted to the Urology Unit of Lanzhou General PLA (Lanzhou, China), from October 2012 to October 2015, were enrolled in this study. The patients were further divided into shock and matched-pair non-shock groups. 2-DE technique, mass spectrometry and database search were used to detect differentially expressed proteins in serum from the two ***:Six proteins were found at higher levels in the shock group compared with non-shock individuals, including serum amyloid A-1 protein (SAA1), apolipoprotein L1 (APOL1), ceruloplasmin (CP), haptoglobin (HP), antithrombin-III (SERPINC1) and prothrombin (F2), while three proteins showed lower levels, including serotransferrin (TF), transthyretin (TTR) and alpha-2-macroglobulin (A2M).Conclusion:Nine proteins were differentially expressed between uroseptic patients (non-shock groups) and severe uroseptic patients (shock groups), compared with non-shock groups, serum SAA1, APOL1,CP, HP, SERPINC1 and F2 at higher levels, while TF, TTR and A2M at lower levels in shock *** proteins were mainly involved in platelet activation, signaling and aggregation, acute phase protein pathway, lipid homeostasis, and iron ion transport, deserve further research as potential candidates for early diagnosis and treatment. 针对云存储中敏感数据的机密性保护问题,在基于属性的加密基础上提出了一种密文访问控制方法HCRE。其思想是设计一种基于秘密共享方案的算法,将访问控制策略变更导致的重加密过程转移到云端执行,从而降低权限管理的复杂度,实现高效的动态密文访问控制。实验分析表明HCRE显著降低了权限管理的时间代价,而且没有向云端泄露额外的信息,保持了数据机密性。

Effects of Pretreatment by the Flavanol Ampelopsin on Porcine Kidney Epithelial Cell Injury Induced by Hydrogen Peroxide

This study investigated the protective effects of ampelopsin on H2O2-induced oxidative injury in porcine kidney epithelial cell line, PK-15; cells were pretreated with medium containing 0, 15, 30, and 60 ug mL-1 ampelopsin, respectively, for 1 h prior to the addition of 100 umol L-1 H2O2. After 2 h, the cell viability, intracellular superoxide dismutase （SOD） and glutathione peroxidase （GSH-Px） activity, cellular malondialdehyde （MDA） content, and DNA strand breakage were determined. Results showed that ampelopsin pretreatment did not modify the SOD and GSH-Px inactivation but significantly decreased MDA production; except for the medium dose （30 ug mL-1）, all the tested concentrations of ampelopsin did not significantly inhibit cell viability reduction and gave no influence on DNA damage. The results suggested that all the tested doses of ampelopsin pretreatment before H2O2 addition can protect PK-15 cell from H2O2-induced lipid peroxidation, which might not be mediated by the SOD and GSH-Px responsees. The appropriate dose of ampelopsin pretreatment prevents PK-15 cell from DNA damage and cell viability reduction.

Six rumen-cannulated lactating Guanzhong goats were used to investigate changes in ruminal fermentation pattern and the microbiota following a subacute rtmainal acidosis （SARA） inducing procedure. Induction of SARA was performed by increasing dietary non-fiber carbohydrate （NFC） to neutral detergent fiber （NDF） ratio from 1.02 （stage 1） to 1.24 （stage 2）, 1.63 （stage 3） and 2.58 （stage 4）. A dynamic pH monitoring system, real-time fluorescent quantitative PCR and conventional anaerobic culture were used to assess changes in ruminal pH and microbiota. Results indicated that rumen fermentation patterns changed significantly with increased NFC：NDF ratio. The decline in ruminal pH was caused by increased ruminal total volatile fatty acids （TVFA）, which was mainly attributed to a significant increase in ruminal butyrate, rather than the accumulation of ruminal lactic acid. In addition, in the course of SARA, the number of rumen microoganisms altered significantly, with increases in ruminal amylolytic bacteria, Lactobacilli, Streptococcus bovis and Megasphaera elsdenii, the latter particularly dramatically indicating that it may be the main factor responsible for the increase in butyrate, and decrease in protozoa.

目的为硫酸庆大霉素碳酸铋胶囊中庆大霉素C组分及有关物质建立质控方法。方法采用HPLC-ELSD方法,色谱柱:GRACEApollo C18柱(250 mm×4.6 mm,5μm);流动相:0.2 mol/L三氟乙酸溶液-甲醇(96:4,V/V),流速0.6 mL/min;柱温30℃;进样量20μL;Waters低温分流型蒸发光散射检测器,载气流量40 psi,漂移管温度55℃。结果所建方法通过方法学验证,庆大霉素C组分与各杂质间分离完全,庆大霉素、西索米星和小诺霉素分别在0.5~6 mg/mL、25~100μg/mL和25~100μg/mL的浓度范围内,各浓度的对数值与对应色谱峰面积的对数值呈现出良好的线性。采用所建方法对市售16家生产企业43批次硫酸庆大霉素碳酸铋胶囊样品进行测定,获得该制剂产品中各组分及有关物质的数据。结论所建方法是在中国药典方法基础上优化并验证适用于硫酸庆大霉素碳酸铋胶囊C组分及有关物质的含量测定,方法简便准确,专属性强,稳定性好。由获得的实验数据可知,该复方制剂中C组分及有关物质的含量在各厂家间或同厂家不同批次间差异显著,可能存在影响用药安全的潜在隐患,建议在该剂型质量标准中增加相关质控项。

Background Dehydroepiandrosterone （DHEA） is widely known for its beneficial effect on postmenopausal osteoporosis, although the underlying mechanisms remain mainly unclear. In this study, we tried to determine the activation of mitogen-activated protein kinase signal pathways during DHEA treatment and the indirect role of osteoblasts （OBs） on osteoclasts under the DHEA treatment of postmenopausal osteoporosis. Methods Primary human OBs and osteoclast-like cells were cultured and, we pretreated OBs with or without U0126 （a highly selective inhibitor of both MEK1 and MEK2）. The OBs were treated with DHEA. We then tested the effects of DHEA on human osteoblastic viability, osteoprotegerin production and the expression of phosphor-ERK1/2 （extracellular signal-regulated kinase）. In the presence or absence of OBs, the function of osteoclastic resorption upon DHEA treatment was calculated. Results DHEA promoted the human osteoblastic proliferation and inhibited the osteoblastic apoptosis within the concentration range of 108-106 mol/L （P 〈0.05, P 〈0.01, respectively）. Within the effective concentration range, the expression of phosphor-ERK1/2 and osteoprotegerin was increased by DHEA and blocked by U0126. In the presence of OBs, DHEA could significantly decrease the number and the area of bone resorption lacuna （P 〈0.05 and P 〈0.01, respectively）. Without OBs, however, the effects of DHEA on the bone resorption lacuna were almost completely abolished. Conclusions DHEA could indirectly inhibit the human osteoclastic resorption through promoting the osteoblastic viability and osteoprotegerin production, which is mediated by mitogen-activated protein kinases signal pathway involving the phosphor-ERK1/2.
