Obesity,which can arise from genetic or environmental factors,has been shown to cause serious damages to the reproductive *** ovary,as one of the primary regulators of female fertility,is a complex organ comprised of heterogeneous cell types that work together to maintain a normal ovarian microenvironment(OME).Despite its importance,the effect of obesity on the entire ovary remains poorly *** this study,we performed ovary single-cell and nanoscale spatial RNA sequencing to investigate how the OME changed under different kinds of obesity,including high-fat diet(HFD)induced obesity and Leptin ablation induced obesity(OB).Our results demonstrate that OB,but not HFD,dramatically altered the proportion of ovarian granulosa cells,theca-interstitial cells,luteal cells,and endothelial ***,based on the spatial dynamics of follicular development,we defined four subpopulations of granulosa cell and found that obesity drastically disrupted the differentiation of mural granulosa cells from small to large antral ***,HFD enhanced follicle-stimulating hormone(FSH)sensitivity and hormone conversion,while OB caused decreased sensitivity,inadequate steroid hormone conversion,and impaired follicular *** differences can be explained by the differential expression pattern of the transcription factor ***,our study provides a powerful and high-resolution resource for profiling obesity-induced OME and offers insights into the diverse effects of obesity on female reproductive disorders.

Reducing the short-circuit rate and increasing the effective discharge rate are important targets for improving the servo control effect of micro-electrical discharge machining(micro-EDM),as these two indicators are closely related to the machining efficiency and *** this study,a feed-pulse collaborative control(FPCC)method is proposed for micro-EDM based on two dimensions(space and time).In the spatial dimension,a feed control strategy with a discharge holding process is ***,in the time dimension,a forward-looking pulse control strategy is adopted,in which the pulse interval is adjusted based on a sequence analysis of feed commands and discharge *** experiments are carried out to determine the key parameters used in this method,including the discharge holding threshold and pulse interval adjustment value(T_(off_(adj))).The feed smoothness and discharge sufficiency analyses of the experimental results show that compared to the traditional double threshold average voltage method,the FPCC method reduces the number of long-distance retreats by 64%and improves the effective discharge time by 40%.

The phase behavior of gas condensate in reservoir formations differs from that in pressure-volume-temperature(PVT)cells because it is influenced by porous media in the reservoir *** was used as a sample to investigate the influence of porous media on the phase behavior of the gas *** pore structure was first analyzed using computed tomography(CT)scanning,digital core technology,and a pore network *** sandstone core sample was then saturated with gas condensate for the pressure depletion *** each pressure-depletion state was stable,realtime CT scanning was performed on the *** scanning results of the sample were reconstructed into three-dimensional grayscale images,and the gas condensate and condensate liquid were segmented based on gray value discrepancy to dynamically characterize the phase behavior of the gas condensate in porous *** network models of the condensate liquid ganglia under different pressures were built to calculate the characteristic parameters,including the average radius,coordination number,and tortuosity,and to analyze the changing mechanism caused by the phase behavior change of the gas *** types of condensate liquid(clustered,branched,membranous,and droplet ganglia)were then classified by shape factor and Euler number to investigate their morphological changes dynamically and *** results show that the dew point pressure of the gas condensate in porous media is 12.7 MPa,which is 0.7 MPa higher than 12.0 MPa in PVT *** average radius,volume,and coordination number of the condensate liquid ganglia increased when the system pressure was between the dew point pressure(12.7 MPa)and the pressure for the maximum liquid dropout,Pmax(10.0 MPa),and decreased when it was below *** volume proportion of clustered ganglia was the highest,followed by branched,membranous,and droplet *** study provides crucial experimental evidence for the phase behavior chan

Quasi-solid-state lithium metal batteries(QSSLMBs)assembled with polyvinylidene fluoride(PVDF)are a promising class of next-generation rechargeable batteries due to their safety,high energy density,and superior interfacial ***,PVDF has a series of inherent drawbacks such as low ionic conductivity,ease of crystallization,and hydrophobic character that leading to poor cell *** tackle these issues,a lignin-reinforced PVDF electrolyte is proposed in this work to solve these drawbacks of PVDF and enhance the comprehensive performance of *** lithophilic polar groups of lignin can promote uniform deposition of Li on the *** with the improved mechanical properties can efficiently prevent Li dendrites penetration through the *** addition,more active sites provided by lignin can also enhance Li^(+)transport and lead to a faster electrochemical reaction *** from the ingenious design,Li symmetric cells with 5%lignin-PVDF quasi-solid-state electrolyte can operate for 900 h at a high current density/capacity of 5 mA·cm^(-2)/5 mAh·cm^(-2),while shortcircuiting occurs after 56 h for the counterpart(pure PVDF).Moreover,a full cell of Li/5%lignin-PVDF/LFP cell demonstrates a high capacity of 96.2 mAh·g^(-1)after 2000 cycles at 10 *** work is expected to open up promising opportunities to develop other high-energy/power-density QSSLMBs.

Effectiveness of Acupoint Application in Patients with Pharyngeal Pain: Evidence from CHUNBO, A Prospective Real-World Study

Objective: To assess the outcomes after acupoint application in patients with pharyngeal pain in a real-world settings, and analyze the characteristics of effective population and prescription characteristics of acupoint application. Methods: Based on CHUNBO platform, patients with pharyngeal pain who were candidates for acupoint application on the basis of physician-evaluation, were enrolled in a nationwide, prospective, 69-week multicenter observational study from August 2020 to February 2022. Propensity score matching(PSM) was used to match the confounding factors and the association rules were used to analyze the characteristics of effective population and prescription characteristics of acupoint application. Outcome assessments included the disappearance rate of pharyngeal pain(within 3, 7, and 14 days), disappearance time of pharyngeal pain, as well as adverse events. Results: Of 7,699 enrolled participants, 6,693(86.9%) received acupoint application and 1,450(21.7%) with non-acupoint application. After PSM, there were 1,004 patients each in the application group(AG) and non-application group(NAG). The disappearance rate of pharyngeal pain in the AG at 3, 7, and 14 days were all higher than those in the NAG(P<0.05). The disappearance time of pharyngeal pain in the AG were shorter than that in the NAG(logrank P<0.001, hazard ratio=1.51, 95% confidence interval: 1.41–1.63). The median age of effective cases was 4 years, mainly 3–6 years old(40.21%). The disappearance rate of pharyngeal pain in the application group with tonsil diseases was 2.19 times higher than that in the NAG(P<0.05). The commonly used acupoints for the effective cases were Tiantu(RN 22), Shenque(RN 8) and Dazhui(DU 14). The commonly used herbs for the effective cases were Natrii sulfas, Radix et Rhizoma Rhei, and Herba Ephedrae. Among them, Natrii sulfas was applied to RN 8 most frequently(support 84.39%). Three-dimensional(3D)cell cultures have contributed to a variety of biological research fields by filling the gap between monolayers and animal *** modern optical sectioning microscopic methods make it possible to probe the complexity of 3D cell cultures but are limited by the inherent *** tissue optical clearing methods have emerged as powerful tools for investigating whole-mount tissues in 3D,they often have limitations,such as being too harsh for fragile 3D cell cultures,requiring complex handling protocols,or inducing tissue deformation with shrinkage or *** address this issue,we proposed a modified optical clearing method for 3D cell cultures,called MACS-W,which is simple,highly efficient,and *** our evaluation of MACS-W,we found that it exhibits excellent clearing capability in just 10 min,with minimal deformation,and helps drug evaluation on tumor *** summary,MACS-W is a fast,minimally-deformative and fluorescence compatible clearing method that has the potential to be widely used in the studies of 3D cell cultures.

Portable fluorescence instrument for detecting membrane integrity in membrane bioreactor (MBR)

This study proposed the design, fabrication, and assembly of membrane integrity detection instruments in membrane bioreactors (MBR) based on fluorescence spectroscopy. Based on the PARAFAC model, we found that the peak at 280/335 nm strengthened after membrane breakage. The peak at 340/430 nm reflected the sludge concentration in the MBR and reduced the influence of internal filtration effects on detection. Therefore, we determined that the dual-LED light source excitation detection system can detect tryptophan-like substances at 280 nm (T-peak) and humic acid at 340 nm (C-peak). T-peak was identified as the core index indicating membrane integrity. Moreover, the C-peak is the reference indicator factor for a sensitive response to changes in the sludge concentration. The portable fluorescence instrument exhibited high sensitivity and good feedback accuracy compared to particle counting and turbidity detection, where the log reduction value was greater than 3.5. This overcomes the disadvantage of false alarms in particle counters and is not affected by the position of the pump system. This portable instrument provides a flexible and highly sensitive method for the assessment of industrial membrane integrity.

OBJECTIVE: To explore the four Qi of Pfaffia glomerata(PG) and endow this foreign folk herb with the properties of Chinese medicine, make it Chinese medicinal and localized, and could be used as a Chinese medicine. METHODS: The normal group, six cold herb groups, six hot herb groups, six cool herb groups, PG prescription group(PGPG), and Dangshen(Radix Codonopsis) prescription group(CPPG) were prepared with corresponding concentrations of water extracts, these herb extracts were administered by gavage to SpragueDawley rats, and the 12 h urine at night on the 29th day of the SD rats in each group were collected, Liquid Chromatograph Mass Spectrometer system was used to analyze them, the best discriminant models for the medicinal properties of cold-hot and cold-cool were set up, so as to the medicinal properties of PGPG, CPPG and PG were predicted. Based on the Progenesis QI, Human Metabolome Database, Kyoto Encyclopedia of Genes and Genomes, MetaboAnalyst 5.0 database, we enriched metabolic pathway and classification mechanism of medicinal properties of cold-cool Chinese herbs and the molecular mechanism of PG prescription. RESULTS: We established a best model of cold-hot herbal discrimination in the positive ion mode, then the probability that PGPG was predicted cold property was 88.9%. Furthermore, a model of cold-cool herbal discrimination was established, then the probability of PGPG containing the cool property was 77.8%. In addition, typical cold and cool herbs mainly affected nine biomarkers such as tyrosine-proline,(R)-3',7-Dihydroxy-2',4'-dimethoxyisoflavane in rats. The regulation trend of PGPG on markers was basically as same as the cool herbs and mainly involved in regulating the two pathways of cytochrome P450 and purine metabolism. CONCLUSION: The results showed PGPG had a cool medicinal property as same as CPPG, and the regulation trend of PGPG on markers was consistent with cool herbs.
