In pediatric advanced cancer,i.e.,when cancer-targeted treatments are not effective,care provided to patients is of palliative *** aims at limiting symptoms and optimizing quality of life(QoL).To orient care,it is necessary to know and measure what is really important for young ***,tools are still scarce in this *** aim of this paper is to describe the development process of a new QoL assessment method,the Advance QoL *** is specifically designed for the clinical context of pediatric advanced *** previous studies carried out by our team identified seven important QoL domains for this *** developed a tool named Advance QoL—parent/caregiver version.A current study aims to develop self-reported versions of the Advance QoL for children(8-12 years)and adolescents(13-18 years)with advanced *** the very near future,the Advance QoL tool will be available in three versions(parents/caregivers,children,and adolescents)ready to be tested in validation *** assessment of the QoL of young people with cancer is a central process in defining targets for care and should be carried out throughout the trajectory,including when cancer is at an advanced stage,i.e.,when no standard treatment options are available.

Design and Transdermal Delivery of Indomethacin Nanosystem

Transdermal drug delivery is one of the most promising routes of drug administration. The aim of this study was to apply nanoencapsulation technologies in order to enhance bioavailability and reduce toxicity of indomethacin after transdermal administration. Methods: Nanoparticles were produced by a two-step desolvation process. Parameters such as mean particle size, zeta potential and encapsulation efficiency were studied. A model drug was selected for encapsulation-indomethacin. The permeability of free-indomethacin in a cellulose-based vehicle and that of indomethacin-loaded nanoparticles was investigated using Franz diffusion cells. The cumulative amount of indomethacin that diffused across a model membrane was measured and the steady-state fluxes of the permeant in the different systems were determined. Results: This study showed that it is possible to prepare indomethacin-loaded gelatine nanoparticles with small particle size (mean size lower than 350 nm) with polydispersivity index ranging from 0.125 to 0.199 and negative zeta potential. encapsulation efficiency was around 70%. Permeation studies showed that nanoparticles prolonged indomethacin release. In conclusion, the present data confirm the feasibility of developing indomethacin transdermal nanosystems on an industrial scale. Further studies, now in progress, will deal with the application of the presently reported findings to human skin permeation, involving both in vitro and in vivo testing.

Broad Hormonal Responses Induced by Aluminum in Roots of Dwarf Transgenics of Solanum lycopersicum L. cv “Micro-Tom”

The spatial pattern distribution of plant hormones in response to aluminum (Al) toxicity in roots remains to be shown. This study was performed to assess the root hormonal accumulation and gene expression in response to Al toxicity in five transgenic miniature dwarf tomatoes cv. Micro-Tom (MT). MT and MT transgenics to acid indole acetic, cytokinin, gibberellin, abscisic acid and ethylene were cultivated in nutrient solutions containing different Al concentrations. Root growth elongation was measured and cellular damage was visualized by staining evans’s blue. The GUS reporter gene staining technique was used to visualize hormonal changes in MT apex root tissues. Data indicated that the MT is sensitive to Al that induced significant growth inhibition and cellular damage. Al concentration of 27 μM was significantly toxic, inducing root apex darkening and inhibition of root development. The qualitative evaluation of GUS reporter gene expression showed intense crosstalk among all hormones studied, underscoring the complexity of signaling induced by Al in apex roots. Results point out to a major understanding of the hormonal signaling in response to Al toxicity, which may induce a change of root growth and architecture with growth inhibition and cell constraints modulated by all different hormones evaluated.

Sustainability of e-Trike as Alternative Mode of Public Transportation System: The Case of Cabanatuan City, Philippines

The City of Cabanatuan is dubbed as the “Tricycle Capital of the Philippines”. Tricycle is a three wheeled vehicle used to transport people. Cabanatuan has about 30,000 registered gas-fed four-stroke motorized tricycles. Tricycle operation in this City has become the source of livelihood for around 10,000 families. Its number has tremendously increased despite growing competition and declining net daily income and the adverse effects it brings to the environment. The study analyzed the different aspects in the adoption of e-trikes (electric tricycles) as a mode of public transport system replacing motorized tricycles in the city. The conceptual framework of sustainable development was used to analyze the data. Basic accounting method in determining return on investment and payback period were used to compare the cost and return from among the current mode of transportation as against the adoption of e-trike. Data gathering employed the interviews, survey questionnaires and focus group discussion. The three areas of sustainability measured in the study were economic, socio-political and environmental. The study showed that from among 412 tricycle driver-operators randomly selected, majority were amenable to the shift from engine propelled tricycle to e-trike. However, cost of e-trike deters adoption and full implementation by the City Government. Political issue is also?the?utmost consideration since work displacement is imminent among the stakeholders once adoption of this mode takes place. On the other side, e-trike was found to be sustainable in the long run both on the point of view of operators and the riding public. The paper recommended that policy regulation be laid down, adopted, and implemented by the local government unit in response to the Philippines Cleaner Air Act.

efficacy and long-term evaluation of intramyocardial injection of autologous CD34-enriched PBMSC in old myocardial infarction

Aims: We have shown that autologous transplant of CD34+-enriched peripheral-blood mononuclear cells (PBMSC) could restore depressed myocardial function, and sustain adequate myocardial function 12 months after surgery in patients with old (>one year-old) myocardial infarction. Our aim is to report the long-term morbidity and mortality efficacy of this procedure. Methods and results: Seventy patients with an old anteroseptal myocardial infarction were followed for 2 to 7 years, 35 had a revascularization procedure and received an intra-myocardial injection of autologous CD34+-enriched PBMSC (8 × 108 mononuclear cells/ml including 3 × 107 CD34+ cells/ml)(Group A). Group B patients only had the revascularization. Abnormal pre-surgical values of LVeF (33.2% ± 4.8%), LVDV (178 ± 13.7 ml), LVSV (120 ± 16 m), LVDD (58.9 ± 3.84 mm), e and A waves without contractility in infarction area in group A patients improved to approximate normal values (50% ± 3% for LVeF;90 ± 9.3 ml for LVDV;80 ± 9.9 ml for LVSV;55.3 ± 3 mm for LVDD;5.2 ± 0.5 cm/s for e wave and 4.18 ± 0.3 cm/s for A wave) 1 year after the procedure and have remained unaltered for all the follow-up period. All the patients remain alive. Only seven patients have been readmitted to the hospital for non-myocardial related events. Group B only 11 patients continued alive to 5 years after surgery and LeVF never increased more than 6%, all of them with many hospitalizations (n ≥ 10) by heart failure events. Conclusion: Intramyocardial injection of CD34+ highly enriched PBSC represent an encouraging alternative for patients with severely scarred and dysfunctional myocardium.

Agave sisalana extract induces cell death in Aedes aegypti hemocytes increasing nitric oxide production

Objective: To investigate the effects of Agave sisalana(A. sisalana) extract on Aedes aegypti(Ae. aegypti) primary cell ***: Cells of Ae. aegypti were exposed to different concentrations of A. sisalana crude extract(0.18–6.00 mg/m L) for 24 h. Then, the cells were labeled with propidium iodide and subjected to fluorescence microscopy to verify cell viability. In addition, nitric oxide production was ***: Results showed that cells exposed to 6 mg/m L of the crude extract presented a greater percentage of death when compared to control(73.8% ± 9.6% vs. 34.6% ± 9.6%).Furthermore, there was an increase in the nitric oxide production in cells exposed to 6 mg/m L of A. sisalana crude extract [(0.81 ± 0.08) mmol/L] compared to control group[(0.41 ± 0.18) mmol/L].Conclusions: The results show that A. sisalana is cytotoxic to Ae. aegypti and may be used as raw material for new eco-friendly and inexpensive insecticides, since sisal industry discards the liquid waste for the extraction of plant fiber.

Presence of c.3956del C mutation in familial adenomatous polyposis patients from Brazil

AIM: To characterize APC gene mutations and correlate them with patient phenotypes in individuals diagnosed with familial adenomatous polyposis(FAP) in northern Brazil. MeTHODS: A total of 15 individuals diagnosed with FAP from 5 different families from the north of Brazil were analyzed in this study. In addition to patients with histopathological diagnosis of FAP,family members who had not developed the disease were also tested in order to identify mutations and for possible genetic counseling. All analyzed patients or their guardians signed a consent form approved by the Research ethics Committee of the Jo?o de Barros Barreto University Hospital(Belem,Brazil). DNA extracted from the peripheral blood of a member of each of the affected families was subjected to direct sequencing. The proband of each family was sequenced to identify germline mutations using the Ion Torrent platform. To validate the detected mutations,Sanger sequencing was also performed. The samples from all patients were also tested for the identification of mutations by real-time quantitative polymerase chain reaction using the amplification refractory mutation system. ReSULTS: Through interviews with relatives and a search of medical records,it was possible to construct genograms for three of the five families included in the study. All 15 patients from the five families with FAP exhibited mutations in the APC gene,and all mutations were detected in exon 15 of the APC gene. In addition to the patients with a histological diagnosis of FAP,family members without disease symptoms showed the mutation in the APC gene. In the present study,we detected two of the three most frequent germline mutations in the literature: the mutation at codon 1309 and the mutation at codon 1061. The presence of c.3956 del C mutation was found in all families from this study,and suggests that this mutation was introduced in the population of the State of Pará through ancestor immigration(i.e.,a de novo mutation that arose
