OSW-1 triggers necroptosis in colorectal cancer cells through the RIPK1/RIPK3/MLKL signaling pathway facilitated by the RIPK1- p62/SQSTM1 complex

BACKGROUND Necroptosis has emerged as a novel molecular pathway that can be targeted by chemotherapy agents in the treatment of cancer.OSW-1,which is derived from the bulbs of Ornithogalum saundersiae Baker,exerts a wide range of pharmaco-logical effects.AIM To explore whether OSW-1 can induce necroptosis in colorectal cancer(CRC)cells,thereby expanding its range of clinical applications.METHODS We performed a sequence of functional experiments,including Cell Counting Kit-8 assays and flow cytometry analysis,to assess the inhibitory effect of OSW-1 on CRC cells.We utilized quantitative proteomics,employing tandem mass tag label-ing combined with liquid chromatography-tandem mass spectrometry,to analyze changes in protein expression.Subsequent bioinformatic analysis was conducted to elucidate the biological processes associated with the identified proteins.Transmission electron microscopy(TEM)and immunofluorescence studies were also performed to examine the effects of OSW-1 on necroptosis.Finally,western blotting,siRNA experiments,and immunoprecipitation were employed to evaluate protein interactions within CRC cells.RESULTS The results revealed that OSW-1 exerted a strong inhibitory effect on CRC cells,and this effect was accompanied by a necroptosis-like morphology that was observable via TEM.OSW-1 was shown to trigger necroptosis via activation of the RIPK1/RIPK3/MLKL pathway.Furthermore,the accumulation of p62/SQSTM1 was shown to mediate OSW-1-induced necroptosis through its interaction with RIPK1.CONCLUSION We propose that OSW-1 can induce necroptosis through the RIPK1/RIPK3/MLKL signaling pathway,and that this effect is mediated by the RIPK1-p62/SQSTM1 complex,in CRC cells.These results provide a theoretical foundation for the use of OSW-1 in the clinical treatment of CRC.

Metabolic-associated fatty liver disease(MAFLD)is one of the most common chronic liver diseases worldwide.In recent years,the occurrence rate of MAFLD has been on the rise,mainly due to lifestyle changes,high-calorie diets,and imbalanced dietary structures,thereby posing a threat to human health and creating heavy social and economic burdens.With the development of 16S sequencing and integrated multi-omics analysis,the role of the gut microbiota(GM)and its metabolites in MAFLD has been further recognized.The GM plays a role in digestion,energy metabolism,vitamin synthesis,the prevention of pathogenic bacteria colonisation,and immunoregulation.The gut-liver axis is one of the vital links between the GM and the liver.Toxic substances in the intestine can enter the liver through the portal vascular system when the intestinal barrier is severely damaged.The liver also influences the GM in various ways,such as bile acid circulation.The gut-liver axis is essential in maintaining the body’s normal physiological state and plays a role in the onset and prognosis of many diseases,including MAFLD.This article reviews the status of the GM and MAFLD and summarizes the GM characteristics in MAFLD.The relationship between the GM and MAFLD is discussed in terms of bile acid circulation,energy metabolism,micronutrients,and signalling pathways.Current MAFLD treat ments targeting the GM are also listed.

High-Power Raman Soliton Generation at 1.7 μm in All-Fiber Polarization-Maintaining Erbium-Doped Amplifier

An all-fiber polarization maintaining high-power laser system operating at 1.7 μm based on the Ramaninduced soliton self-frequency shifting effect is demonstrated. The entirely fiberized system is built by erbiumdoped oscillator and two-stage amplifiers with polarization maintaining commercial silica fibers and devices, which can provide robust and stable soliton generation. High-power soliton laser with the average power of 0.28 W,the repetition rate of 42.7 MHz, and pulse duration of 515 fs is generated directly from the main amplifier.Our experiment provides a feasible method for high-power all-fiber polarization maintaining femtosecond laser generation working at 1.7 μm.

Number of parous events affects the association between physical exercise and glycemic control among women with gestational diabetes mellitus:A prospective cohort study

Background:Multiparous women are at a higher risk of gestational diabetes mellitus(GDM) than primiparas.Physical activity during pregnancy has been shown to be beneficial for GDM,but there is little evidence on the association between physical activity and glycemic control among women with GDM,whether primiparas or multiparas.Thus,the objective of the present study was to examine the association between physical activity and glycemic control in women with GDM and to determine what,if any,effects result from number of parous events.Methods:A prospective cohort of 1162 women with GDM was recruited,with 604 multiparas(51.98%).The general linear model was used to calculate the risk difference and its 95% confidence interval(95%CI) to quantify the impact of parous events on glycemic control in pregnancy as well as the association between physical activity time and glycemic control.Results:Among 1162 women with GDM,the median daily activity time was 65 mm(interquartile range(IQR):45-90 min),and the abnormal plasma glucose(PG) percentage,calculated as number of abnormal PG tests divided by the total number of PG tests,was 40.00%(IQR:22.22%-66.67%).The percentage of abnormal PG was stabilized and statistically lower with daily physical activity time exceeding 60 min among primiparas(IQR:30.89%-44.43%) and exceeding 90 min among multiparas(ranged from 27.76% to 38.82%).After adjusting for potential confoumders,primiparas tended to have a lower percentage of abnormal PG than do multiparas(rate difference=-0.39,95%CI:-3.61 to2.84).The same amount of physical activity time was significantly less effective for multiparas than for primiparas(trend p-value <0.01).Conclusion:In women with GDM,being multiparous is associated with less effective glycemic control through physical activity,such that multiparas need more physical activity to achieve glycemic control at a similar level to primiparas.

M1-type microglia can induce astrocytes to deposit chondroitin sulfate proteoglycan after spinal cord injury

After spinal cord injury(SCI),astrocytes gradually migrate to and surround the lesion,depositing chondroitin sulfate proteoglycan-rich extracellular matrix and forming astrocytic scar,which limits the spread of inflammation but hinders axon regeneration.Meanwhile,microglia gradually accumulate at the lesion border to form microglial scar and can polarize to generate a pro-inflammatory M1 phenotype or an anti-inflammatory M2 phenotype.However,the effect of microglia polarization on astrocytes is unclear.Here,we found that both microglia(CX3 CR1^(+))and astrocytes(GFAP^(+))gathered at the lesion border at 14 days post-injury(dpi).The microglia accumulated along the inner border of and in direct contact with the astrocytes.M1-type microglia(i NOS^(+)CX3 CR1^(+))were primarily observed at 3 and 7 dpi,while M2-type microglia(Arg1^(+)CX3 CR1^(+))were present at larger numbers at 7 and 14 dpi.Transforming growth factor-β1(TGFβ1)was highly expressed in M1 microglia in vitro,consistent with strong expression of TGFβ1 by microglia in vivo at 3 and 7 dpi,when they primarily exhibited an M1 phenotype.Furthermore,conditioned media from M1-type microglia induced astrocytes to secrete chondroitin sulfate proteoglycan in vitro.This effect was eliminated by knocking down sex-determining region Y-box 9(SOX9)in astrocytes and could not be reversed by treatment with TGFβ1.Taken together,our results suggest that microglia undergo M1 polarization and express high levels of TGFβ1 at 3 and 7 dpi,and that M1-type microglia induce astrocytes to deposit chondroitin sulfate proteoglycan via the TGFβ1/SOX9 pathway.The study was approved by the Institutional Animal Care and Use Committee of Anhui Medical University,China(approval No.LLSC20160052)on March 1,2016.

本文提出了一种新型局域共振夹芯超结构梁,该结构由夹芯梁基体和由软材料包裹质量块组成的局域共振单元构成。分别采用传递矩阵法和有限元法计算了该结构的带隙分布和振动传输特性,搭建了局域共振夹芯超结构梁的振动传输测试实验平台,并与数值计算结果进行对比验证,最后分析了结构参数对结构带隙分布特性的调控规律。研究结果表明,该结构在低频范围内共出现两条弯曲波带隙,带隙频率分别为196.1~339.0 Hz和377.2~655.6 Hz。而完全带隙位于弯曲波带隙内,频率范围为380.0~423.6 Hz。且在这两条弯曲波带隙范围内,弯曲振动传输会发生明显的减弱现象。另外,局域共振夹芯超结构梁的结构参数会对带隙分布特性产生较大影响,通过合理选择结构参数,可以实现局域共振夹芯超结构梁低频率和高带宽的减振要求。

Two-parameter estimation with three-mode NOON state in a symmetric triple-well potential

We propose a scheme to realize two-parameter estimation via Bose–Einstein condensates confined in a symmetric triple-well potential.The three-mode NOON state is prepared adiabatically as the initial state.The two parameters to be estimated are the phase differences between the wells.The sensitivity of this estimation scheme is studied by comparing quantum and classical Fisher information matrices.As a result,we find an optimal particle number measurement method.Moreover,the precision of this estimation scheme means that the Heisenberg scaling behaves under the optimal measurement.

Surgical treatment of left-sided renal carcinoma with grade II inferior vena cava tumour thrombus: a report and review of the literature

The surgical removal of renal cancer,along with the thrombectomy of the inferior vena cava tumour thrombus,represents a remarkable milestone in urological surgery.This procedure is not only technically demanding but also requires a high level of surgical expertise.Managing renal cancer combined with a vena cava tumour thrombus poses significant challenges,especially when dealing with combined grade Ⅱ-Ⅳ inferior vena cava tumour thrombus.The complexity of these cases is further exacerbated by the delicate anatomical structures involved and the need to preserve critical vessels while effectively removing the tumour.The Upper Urethral Tumour Treatment Centre of Weifang People's Hospital successfully treated a challenging case of left renal tumour combined with grade II inferior vena cava tumour thrombus.The surgical team,led by experienced urological surgeons,meticulously planned and executed the procedure,ensuring minimal trauma to the patient and complete removal of the tumour.This achievement not only demonstrates the hospital's commitment to providing state-of-the-art surgical care but also highlights the importance of continued research and training in urological oncology.The successful outcome of this case is a testament to the expertise and dedication of the medical team and offers hope to patients facing similar complex surgical challenges.

环境中抗生素污染对生态系统安全产生负面影响.以瓯江流域为例,采用高效液相色谱-串联质谱法对浙南诸河流域水体中6类35种抗生素浓度进行检测,分析抗生素的浓度水平和空间分布,利用相关模型评估抗生素对生态和人体的健康风险,讨论抗生素的来源.结果表明,瓯江流域地表水20个采样点中4类12种抗生素均有不同程度检出,分别是磺胺类、喹诺酮类、四环素类和林可酰胺类,总浓度为ND~1 018 ng·L^(-1).其中检出率较高的是林可霉素,为90.48%,其次是磺胺吡啶,为38.10%.检出浓度平均值最大的3种抗生素分别是氧氟沙星(12.49 ng·L^(-1))、林可霉素(11.08 ng·L^(-1))和双氟沙星(7.38 ng·L^(-1)).流域内抗生素以点状污染为主,空间分异性较大,上游抗生素浓度平均值(54.39 ng·L^(-1))大于中下游浓度平均值(46.64 ng·L^(-1)),抗生素污染程度由上游至下游呈现出“上疏下密”的特点;上游污染主要来源为医疗和畜禽养殖业废水的排放,下游污染主要为人口活动密集和经济产业快速发展所导致.生态风险评估结果表明,上游采样点H6具有最大的风险商值,其中氧氟沙星和恩诺沙星具有高风险水平,林可霉素具有中风险水平.健康风险评估结果表明,瓯江流域水体抗生素对人体健康无风险.
