Visualizing Wallerian degeneration in the corticospinal tract after sensorimotor cortex ischemia in mice

Stroke can cause Wallerian degeneration in regions outside of the brain,particularly in the corticospinal tract.To investigate the fate of major glial cells and axons within affected areas of the corticospinal tract following stroke,we induced photochemical infarction of the sensorimotor cortex leading to Wallerian degeneration along the full extent of the corticospinal tract.We first used a routine,sensitive marker of axonal injury,amyloid precursor protein,to examine Wallerian degeneration of the corticospinal tract.An antibody to amyloid precursor protein mapped exclusively to proximal axonal segments within the ischemic cortex,with no positive signal in distal parts of the corticospinal tract,at all time points.To improve visualization of Wallerian degeneration,we next utilized an orthograde virus that expresses green fluorescent protein to label the corticospinal tract and then quantitatively evaluated green fluorescent protein-expressing axons.Using this approach,we found that axonal degeneration began on day 3 post-stroke and was almost complete by 7 days after stroke.In addition,microglia mobilized and activated early,from day 7 after stroke,but did not maintain a phagocytic state over time.Meanwhile,astrocytes showed relatively delayed mobilization and a moderate response to Wallerian degeneration.Moreover,no anterograde degeneration of spinal anterior horn cells was observed in response to Wallerian degeneration of the corticospinal tract.In conclusion,our data provide evidence for dynamic,pathogenic spatiotemporal changes in major cellular components of the corticospinal tract during Wallerian degeneration.

Attempts have been made to use cell transplantation and biomaterials to promote cell proliferation,differentiation,migration,and survival,as well as angiogenesis,in the context of brain injury.However,whether bioactive materials can repair the damage caused by ischemic stroke by activating endogenous neurogenesis and angiogenesis is still unknown.In this study,we applied chitosan gel loaded with basic fibroblast growth factor to the stroke cavity 7 days after ischemic stroke in rats.The gel slowly released basic fibroblast growth factor,which improved the local microenvironment,activated endogenous neural stem/progenitor cells,and recruited these cells to migrate toward the penumbra and stroke cavity and subsequently differentiate into neurons,while enhancing angiogenesis in the penumbra and stroke cavity and ultimately leading to partial functional recovery.This study revealed the mechanism by which bioactive materials repair ischemic strokes,thus providing a new strategy for the clinical application of bioactive materials in the treatment of ischemic stroke.

CFD-DEM study of impacts of the porous distributor medium on fluidization characteristics of a 2D-fluidized bed

Gas-solid bubbling fluidized beds are widely used in chemical,energy,construction,and other industrial fields due to their excellent fluidization performance.2D-fluidized beds with planar rectangular columns of finite thickness are widely used in fluidized bed studies,and it is essential for understanding the hydrodynamics of gas-particle systems.Moreover,the distributor(porous medium)at the bottom of the bed has a crucial influence on the fluidization performance of the 2D-fluidized bed.In this work,the fluidization mechanism and gas-solid dynamic characteristics of the fluidized bed under three porous media are studied by CFD-DEM coupled with the porous medium model,and the accuracy of numerical simulation is verified by a high-speed photography experiment.Results show that with the increase in the flow resistance of the porous medium,the average and standard deviation of the bubble diameter decrease,and the time and position of particles in the dead zone to participate in the core-annular flow advance.At a low fluidization velocity,the dead zone in the bed can be considerably reduced by increasing the flow resistance.

Cellular metabolism is an intricate network satisfying bioenergetic and biosynthesis requirements of cells.Relevant studies have been constantly making inroads in our understanding of pathophysiology,and inspiring development of therapeutics.As a crucial component of epigenetics at post-transcription level,RNA modification significantly determines RNA fates,further affecting various biological processes and cellular phenotypes.To be noted,immunometabolism defines the metabolic alterations occur on immune cells in different stages and immunological contexts.In this review,we characterize the distribution features,modifying mechanisms and biological functions of 8 RNA modifications,including N6-methyladenosine(m6A),N6,2'-O-dimethyladenosine(m6Am),N1-methyladenosine(m1A),5-methylcytosine(m5C),N4-acetylcytosine(ac4C),N7-methylguanosine(m7G),Pseudouridine(V),adenosine-to-inosine(A-to-l)editing,which are relatively the most studied types.Then regulatory roles of these RNA modification on metabolism in diverse health and disease contexts are comprehensively described,categorized as glucose,lipid,amino acid,and mitochondrial metabolism.And we highlight the regulation of RNA modifications on immunometabolism,further influencing immune responses.Above all,we provide a thorough discussion about clinical implications of RNA modification in metabolism-targeted therapy and immunotherapy,progression of RNA modification-targeted agents,and its potential in RNAtargeted therapeutics.Eventually,we give legitimate perspectives for future researches in this field from methodological requirements,mechanistic insights,to therapeutic applications.

Distribution and bioavailability of mercury in size-fractioned atmospheric particles around an ultra-low emission power plant in Southwest China

Ultra-low emission(ULE)technology retrofits significantly impact the particulate-bound mercury(Hg)emissions from coal-fired power plants(CFPPs);however,the distribution and bioavailability of Hg in size-fractioned particulate matter(PM)around the ULE-retrofitted CF-PPs are less understood.Here,total Hg and its chemical speciation in TSP(total suspended particles),PM_(10)(aerodynamic particle diameter≤10μm)and PM_(2.5)(aerodynamic particle diameter≤2.5μm)around a ULE-retrofitted CFPP in Guizhou Province were quantified.Atmospheric PM_(2.5)concentration was higher around this ULE-retrofitted CFPP than that in the intra-regional urban cities,and it had higher mass Hg concentration than other sizefractioned PM.Total Hg concentrations in PM had multifarious sources including CFPP,vehicle exhaust and biomass combustion,while they were significantly higher in autumn and winter than those in other seasons(P40%).Mass concentrations of exchangeable,HCl-soluble,elemental,and residual Hg in PM_(2.5)were higher than those in other size-fractioned PM,and were markedly elevated in autumn and winter(P<0.05).In PM_(2.5),HCl-soluble Hg presented a significantly positive relationship with elemental Hg(P<0.05),while residual Hg showed the significantly positive relationships with HCl-soluble Hg and elemental Hg(P<0.01).Overall,these results suggested that atmospheric PM-bound Hg around the ULE-retrofitted CFPP tends to accumulate in finer PM,and has higher bioavailable fractions,while has potential transformation between chemical speciation.

Dynamic ocean inverse modeling based on differentiable rendering

Learning and inferring underlying motion patterns of captured 2D scenes and then re-creating dynamic evolution consistent with the real-world natural phenomena have high appeal for graphics and animation.To bridge the technical gap between virtual and real environments,we focus on the inverse modeling and reconstruction of visually consistent and property-verifiable oceans,taking advantage of deep learning and differentiable physics to learn geometry and constitute waves in a self-supervised manner.First,we infer hierarchical geometry using two networks,which are optimized via the differentiable renderer.We extract wave components from the sequence of inferred geometry through a network equipped with a differentiable ocean model.Then,ocean dynamics can be evolved using the reconstructed wave components.Through extensive experiments,we verify that our new method yields satisfactory results for both geometry reconstruction and wave estimation.Moreover,the new framework has the inverse modeling potential to facilitate a host of graphics applications,such as the rapid production of physically accurate scene animation and editing guided by real ocean scenes.

Prospects in the application of ultrasensitive chromosomal aneuploidy detection in precancerous lesions of gastric cancer

Gastric cancer(GC)is a prevalent malignant tumor within the digestive system,with over 40%of new cases and deaths related to GC globally occurring in China.Despite advancements in treatment modalities,such as surgery supplemented by adjuvant radiotherapy or chemotherapeutic agents,the prognosis for GC remains poor.New targeted therapies and immunotherapies are currently under invest-igation,but no significant breakthroughs have been achieved.Studies have indicated that GC is a heterogeneous disease,encompassing multiple subtypes with distinct biological characteristics and roles.Consequently,personalized treatment based on clinical features,pathologic typing,and molecular typing is crucial for the diagnosis and management of precancerous lesions of gastric cancer(PLGC).Current research has categorized GC into four subtypes:Epstein-Barr virus-positive,microsatellite instability,genome stability,and chromosome instability(CIN).Technologies such as multi-omics analysis and gene sequencing are being employed to identify more suitable novel testing methods in these areas.Among these,ultrasensitive chromosomal aneuploidy detection(UCAD)can detect CIN at a genome-wide level in subjects using low-depth whole genome sequencing technology,in conjunction with bioinformatics analysis,to achieve qualitative and quantitative detection of chromosomal stability.This editorial reviews recent research advancements in UCAD technology for the diagnosis and management of PLGC.

BACKGROUND Laparoscopic rectal cancer radical surgery is a complex procedure affected by various factors.However,the existing literature lacks standardized parameters for the pelvic region and soft tissues,which hampers the establishment of consistent conclusions.AIM To comprehensively assess 16 pelvic and 7 soft tissue parameters through computerized tomography(CT)-based three-dimensional(3D)reconstruction,providing a strong theoretical basis to address challenges in laparoscopic rectal cancer radical surgery.METHODS We analyzed data from 218 patients who underwent radical laparoscopic surgery for rectal cancer,and utilized CT data for 3D pelvic reconstruction.Specific anatomical points were carefully marked and measured using advanced 3D modeling software.To analyze the pelvic and soft tissue parameters,we emp-loyed statistical methods including paired sample t-tests,Wilcoxon rank-sum tests,and correlation analysis.RESULTS The investigation highlighted significant sex disparities in 14 pelvic bone parameters and 3 soft tissue parameters.Males demonstrated larger measurements in pelvic depth and overall curvature,smaller measurements in pelvic width,a larger mesorectal fat area,and a larger anterior-posterior abdominal diameter.By contrast,females exhibited wider pelvises,shallower depth,smaller overall curvature,and an increased amount of subcutaneous fat tissue.However,there were no significant sex differences observed in certain parameters such as sacral curvature height,superior pubococcygeal diameter,rectal area,visceral fat area,waist circumference,and transverse abdominal diameter.CONCLUSION The reconstruction of 3D CT data enabled accurate pelvic measurements,revealing significant sex differences in both pelvic and soft tissue parameters.This study design offer potential in predicting surgical difficulties and creating personalized surgical plans for male rectal cancer patients with a potentially“difficult pelvis”,ultimately improving surgical outcomes.Further research and utilization of these parameters

G0 Pythagorean-Hodograph Curves Closest to Prescribed Planar Bézier Curves

The task of identifying the quintic PH curve G0“closest” to a given planar Bézier curve with or without prescribed arc length is discussed here using Gauss-Legendre polygon and Gauss-Lobatto polygon respectively. By expressing the sum of squared differences between the vertices of Gauss-Legendre or Gauss-Lobatto polygon of a given Bézier and those of a PH curve, it is shown that this problem can be formulated as a constrained polynomial optimization problem in certain real variables, subject to two or three quadratic constraints, which can be efficiently solved by Lagrange multiplier method and Newton-Raphson iteration. Several computed examples are used to illustrate implementations of the optimization methodology. The results demonstrate that compared with Bézier control polygon, the method with Gauss-Legendre and Gauss-Lobatto polygon can produce the G0PH curve closer to the given Bézier curve with close arc length. Moreover, good approximations with prescribed arc length can also be achieved.

This editorial comments on an article published in a recent issue of World Journal of Gastroenterology,entitled“Association of low muscle strength with metabolic dysfunction-associated fatty liver disease:A nationwide study”.We focused on the association between muscle strength and the incidence of non-alcoholic fatty liver disease(NAFLD)and metabolic-associated fatty liver disease(MAFLD),as well as the mechanisms underlying the correlation and related clinical applications.NAFLD,which is now redefined as MAFLD,is one of the most common chronic liver diseases globally with an increasing prevalence and is characterized by malnutrition,which may contribute to decreased muscle strength.Reduction of muscle strength reportedly has a pathogenesis similar to that of NAFLD/MAFLD,including insulin resistance,inflammation,sedentary behavior,as well as insufficient vitamin D.Multiple studies have focused on the relationship between sarcopenia or muscle strength and NAFLD.However,studies investigating the relationship between muscle strength and MAFLD are limited.Owing to the shortage of specific medications for NAFLD/MAFLD treatment,early detection is essential.Furthermore,the relationship between muscle strength and NAFLD/MAFLD suggests that improvements in muscle strength may have an impact on disease prevention and may provide novel insights into treatments including dietary therapy,as well as tailored physical activity.
