Differential expression of genes involved in the epigenetic regulation of cell identity in normal human mammary cell commitment and differentiation

The establishment and maintenance of mammary epithelial cell identity depends on the activity of a group of proteins, collectively called maintenance proteins, that act as epigenetic regulators of gene transcription through DNA methylation, histone modification, and chromatin remodeling. Increasing evidence indicates that dysregulation of these crucial proteins may disrupt epithelial cell integrity and trigger breast tumor initiation. Therefore, we explored in silico the expression pattern of a panel of 369 genes known to be involved in the establishment and maintenance of epithelial cell identity and mammary gland remodeling in cell subpopulations isolated from normal human mammary tissue and selectively enriched in their content of bipotent progenitors, committed luminal progenitors, and differentiated myoepithelial or differentiated luminal cells. The results indicated that, compared to bipotent cells, differentiated myoepithelial and luminal subpopulations were both characterized by the differential expression of 4 genes involved in cell identity maintenance: CBX6 and PCGF2, encoding proteins belonging to the Polycomb group, and SMARCD3 and SMARCE1, encoding proteins belonging to the Trithorax group. In addition to these common genes, the myoepithelial phenotype was associated with the differential expression of HDAC1, which encodes histone deacetylase 1, whereas the luminal phenotype was associated with the differential expression of SMARCA4 and HAT1, which encode a Trithorax protein and histone acetylase 1, respectively. The luminal compartment was further characterized by the overexpression of ALDH1A3 and GATA3, and the down-regulation of NOTCH4 and CCNB1, with the latter suggesting a block in cell cycle progression at the G2 phase. In contrast, myoepithelial differentiation was associated with the overexpression of MYC and the down-regulation of CCNE1, with the latter suggesting a block in cell cycle progression at the G1 phase.

Epithelial cell identity in hyperplastic precursors of breast cancer

Introduction:In the adult human breast,hyperplastic enlarged lobular unit(HELU) and atypical ductal hyperplasia(ADH) are two common abnormalities that frequently coexist with ductal carcinoma in situ(DCIS).For this reason,they have been proposed as the early steps in a biological continuum toward breast ***:We investigated in silico the expression of 369 genes experimentally recognized as involved in establishing and maintaining epithelial cell identity and mammary gland remodeling,in HELUs or ADHs with respect to the corresponding patient-matched normal ***:Despite the common luminal origin,HELUs and ADHs proved to be characterized by distinct gene profiles that overlap for 5 genes *** HELUs were associated with the overexpression of progesterone receptor(PGR),ADHs were characterized by the overexpression of estrogen receptor 1(ESR1) coupled with the overexpression of some proliferation-associated ***:This unexpected finding contradicts the notion that in differentiated luminal cells the expression of estrogen receptor(ER) is dissociated from cell proliferation and suggests that the establishing of an ER-dependent signaling is able to sustain cell proliferation in an autocrine manner as an early event in tumor *** clinical evidence indicates that only a fraction of HELUs and ADHs evolve to invasive cancer,present findings warn that exposure to synthetic progestins,frequently administered as hormone-replacement therapy,and estrogens,when abnormally produced by adipose cells and persistently present in the stroma surrounding the mammary gland,may cause these hyperplastic lesions.

When decisions are based on empirical observations,a trade-off arises between flexibility of the decision and ability to generalize to new *** this paper,we focus on decisions that are obtained by the empirical minimization of the Conditional Value-at-Risk(CVa R)and argue that in CVa R the trade-off between flexibility and generalization can be understood on the ground of theoretical results under very general assumptions on the system that generates the *** results have implications on topics related to order and structure selection in various applications where the CVa R risk-measure is used.A study on a portfolio optimization problem with real data demonstrates our results.

Background: Sympatric species adapt to, and temporally or geographically segregate access to similar limiting ***: We compared nesting habitat and diet affinities of six raptor species in central India between the years ***: A large composition of reptiles in the diet was characteristic for Circaetus gallicus, Spilornis cheela and Elanus caeruleus, while Aquila fasciata, Falco chicquera and Nisaetus cirrhatus show a higher proportion of birds. Species with greatest similarity of diet were C. gallicus and S. cheela. Considering the environmental characterization of areas where raptors built the nest, some species were ecologically closer than others. N. cirrhatus and S. cheela were related to the presence of water bodies, dry deciduous forest and evergreen forests, while E. caeruleus and C. gallicus preferred more open habitats. A. fasciata bred either on cliffs or trees, F. chicquera on trees or mobile towers, and the other four species built their nests exclusively in ***: We conclude that although there was overlap in diet and nesting ecology for a number of species, the geographical separation likely limits competition for resources.

Impact of Frequency Anti-windup Limiter on Synchronization Stability of Grid Feeding Converter

Loss of synchronization is one of the main issues for a grid-feeding converter in a weak grid after being subjected to a large *** synchronous transient is highly nonlinear due to phase movement and frequency ***,none of the previous research has considered the anti-windup PI in the phase-locked loop,which is commonly implemented in reality and introduced as an additional nonlinear *** work provides a taxonomy to evaluate and compare the effect of different anti-windup PI limiters on synchronization stability,including clamping,back-calculation and combined *** anti-windup PI limiters allocate zeros and poles differently and have different impacts on damping and stability enhancement.A case study implemented in Matlab/Simulink serves to compare the trajectory of the converter phase and frequency using different anti-windup PI in the scenario of both with and without equilibrium points during the *** results show that anti-windup PI limiters increase damping during the fault and thus improve the synchronization stability margin.

A resistant method for landmark-based analysis of individual asymmetry in two dimensions

Symmetry of biological structures can be thought as the repetition of their parts in different positions and orientations. Asymmetry analyses, therefore, focuses on identifying and measuring the location and extent of symmetry departures in such structures. In the context of geometric morphometrics, a key step when studying morphological variation is the estimation of the symmetric shape. The standard procedure uses the least-squares Procrustes superimposition, which by averaging shape differences often underestimates the symmetry departures thus leading to an inaccurate description of the asymmetry pattern. Moreover, the corresponding asymmetry values are neither geometrically intuitive nor visually perceivable. Methods： In this work, a resistant method for landmark-based asymmetry analysis of individual bilateral symmetric structures in 2D is introduced. A geometrical derivation of this new approach is offered, while its advantages in comparison with the standard method are examined and discussed through a few illustrative examples. Results： Experimental tests on both artificial and real data show that asymmetry is more effectively measured by using the resistant method because the underlying symmetric shape is better estimated. Therefore, the most asymmetric （respectively symmetric） landmarks are better determined through their large （respectively small） residuals. The percentage of asymmetry that is accounted for by each landmark is an additional revealing measure the new method offers which agrees with the displayed results while helping in their biological interpretation. Conclusions： The resistant method is a useful exploratory tool for analyzing shape asymmetry in 2D, and it might be the preferable method whenever a non homogeneous deformation of bilateral symmetric structures is possible. A new hybrid AI optimal management method for renewable energycommunities

In this study, we propose a hybrid AI optimal method to improve the efficiency of energy managementin a smart grid such as Renewable Energy Community. This method adopts a Time Delay Neural Networkto forecast the future values of the energy features in the community. Then, these forecasts are used by astochastic Model Predictive Control to optimize the community operations with a proper control strategy ofBattery Energy Storage System. The results of the predictions performed on a public dataset with a predictionhorizon of 24 h return a Mean Absolute Error of 1.60 kW, 2.15 kW, and 0.30 kW for photovoltaic generation,total energy consumption, and common services, respectively. The model predictive control fed with suchpredictions generates maximum income compared to the competitors. The total income is increased by 18.72%compared to utilizing the same management system without exploiting predictions from a forecasting method.
