<i>In Silico</i>and <i>in Vitro</i>Approach for the Understanding of the Xanthine Oxidase Inhibitory Activity of Uruguayan Tannat Grape Pomace and Propolis Poliphenols

The use of food additives with xanthine oxidase (XO) inhibitory activity offers an alternative approach to hyperuricemic and gout disease treatment, and provides an example of antioxidant nutraceutics. The in vitro and in silico XO inhibitory activity of polyphenols from Uruguayan Tannat grape pomaces and propolis extracts was evaluated as well as the scavenging capacity of said compounds. When comparing propolis and grape pomace samples, the in vitro studies demonstrated that polyphenols extracted from propolis are more active as free radical scavengers than those from Tannat grape pomace. Both natural products effectively inhibited XO but the capacity of phenols present in GP is higher than the one present in P. The high content of anthocyanins in GP, absent in P, could account for this observation. In silico assays allowed us to determine relevant ligand-receptor interactions between polyphenols, from a database built with previously reported polyphenols from both natural products, and the active site of XO. The in silico results showed that compound (E)-isoprenylcaffeate from propolis was the best potential XO inhibitor displaying hydrophobic aromatic interaction between the conjugated ring of the caffeate moiety and polar interactions between hydroxyl groups from caffeate with the active site polar residues. Among grape pomaces, the Cyanidin-3-O-(6-(E)-p-coumaroyl)-glucoside was the best XO inhibitor;its moiety oxychromenyl being relevant to the docking stabilization. All these results lead us to propose Uruguayan propolis and Tannat grape pomace extracts as food additives as well as phytopharmaceuticals to decrease the uric acid levels in gout disease and to act against oxidative stress.

Background:Platinum chemotherapy (CT) remains the backbone of systemic therapy for patients with small-cell lung cancer (SCLC). The nucleotide excision repair (NER) pathway plays a central role in the repair of the DNA damage exerted by platinum agents. Alteration in this repair mechanism may affect patients' survival.Materials and Methods:We conducted a retrospective analysis of data from 38 patients with extensive disease (ED)-SCLC who underwent platinum-CT at the Clinical Oncology Unit, Careggi University Hospital, Florence (Italy), from 2015 to 2020. mRNA expression analysis and single nucleotide polymorphism (SNP) characterization of three NER pathway genes-namely , , and -were performed on patient tumor samples.Results:Overall, elevated expression of genes was observed in SCLC patients compared to healthy controls. Patients with low and expression levels exhibited a better median progression-free survival (mPFS = 7.1 . 4.9 months, = 0.39 for and mPFS = 6.9 . 4.8 months, = 0.093 for ) and overall survival (mOS = 8.7 . 6.0 months, = 0.4 for and mOS = 7.2 . 6.2 months, = 0.13 for ). Genotyping analysis of five SNPs of genes showed a longer survival in patients harboring the wild-type genotype or the heterozygous variant of the rs11615 SNP ( = 0.24 for PFS and = 0.14 for OS) and of the rs13181 and rs1799793 SNPs ( = 0.43 and = 0.26 for PFS and = 0.21 and = 0.16 for OS, respectively) compared to patients with homozygous mutant genotypes.Conclusions:The comprehensive analysis of gene expression and SNP variants appears to identify patients who derive greater survival benefits from platinum-CT.

Touristic urbanization and greening of coastal dune fields:A long-term assessment of a temperate sandy barrier of Argentina

Dune barrier systems represent highly sought-after coastal landscapes for tourism and urban development around the world. However, a century ago, they were considered hazardous environments due to their great dynamic nature. As a result, stabilization practices were considered necessary. The systematic introduction of fast-growing exotic trees helped stabilize the sand, making it easier for tourism urbanization to take place, but also leading to erosion processes. This paper aims to assess long-term changes in vegetation cover over a large temperate barrier in Argentina. This complex region includes urban resorts, afforestation zones, and protected areas. A GIS-based geospatial analysis was conducted using a large satellite database(> 350 images), and the future evolution of the vegetation was modeled. The results revealed two primary spatiotemporal patterns associated with a gradual expansion of vegetation cover, accompanied by a concurrent reduction in sandy areas. In 1986, the dune area comprised 75% more surface than vegetation, whereas in 2021, it represented 60% less than vegetation. Furthermore, the 2050 scenario suggests a potential 40% reduction of dunes in certain areas. It is necessary to enhance management actions aimed at maintaining dune mobility and ensuring local and regional sediment balance. Long-term management strategies must focus on restoring native plant communities and controlling invasive species, and avoiding new dune fixation initiatives based on the introduction of exotic species.

γγ interactions with KLOE

Studies of γγ interactions are performed with the KLOE detector, without tagging of the outgoing e^＋e^-. The data sample is from an integrated luminosity of 240 pb^-1. It was collected at the φ-factory DAONE with e^＋e^- beams colliding at √s ≌1 GeV, below the φ resonance peak. We present preliminary results concerning the observation of the γγ→η process and the evidence for γγ→π^0π^0 production at low π^0π^0 invariant masses.

Colored bicycle lanes and intersection treatments:International overview and best practices

The use of colored pavements for bicycle facilities represents an interesting strategy for supporting the implementation of a high-quality cycling network. The distinctive coloration of dedicated lanes, tracks or shared paths provides a clearly delineated route, making the infrastructure more attractive as well as legible and intuitive, especially through intersections and junctions. The additional emphasis resulting from the coloring enhances a change in the road users' behavior, but its safety effectiveness seems to be particularly related to several site-specific contextual features. Although some of these benefits are well established and multiple applications of colored pavements are widespread around the world, most of cycling design standards and guides, which are usually written only in the language of the country that drafted them and are often difficult to find, completely lack in technical recommendations and concepts regarding the use of color for cycling ***, based on an extensive worldwide literature search from more than 80 official design guidelines, manuals and standards and 50 scientific articles, the authors have structured a guided overview of the state of the art/practice concerning the use of color for bike facilities at the international level, including the latest innovative and experimental schemes. The review is not intended to be a summary of manuals, but is conceived as a decision-makers' and designers' tool for a rational planning, construction, maintenance and management of colored cycling infrastructures, able to favor a cultural and technical interchange. After a first section in which the main locations where to use colored surfaces were highlighted(corridor or spot treatments), the commonly used colors(red, green and blue) have been identified, explaining the hierarchical and/or combined reasons(regulatory and legal limitations, visual-perceptual aspects and application/maintenance requirements) behind this choice. Despite governing heat management in any realistic device,the microscopic mechanisms of heat transport in all-solid-state electrolytes are poorly known:existing calculations,all based on simplistic semi-empirical models,are unreliable for superionic conductors and largely overestimate their thermal *** this work,we deploy a combination of state-of-the-art methods to calculate the thermal conductivity of a prototypical Li-ion conductor,the Li_(3)ClO *** leveraging ab initio,machine learning,and force-field descriptions of interatomic forces,we are able to reveal the massive role of anharmonic interactions and diffusive defects on the thermal conductivity and its temperature dependence,and to eventually embed their effects into a simple rationale which is likely applicable to a wide class of ionic conductors.

Forecasting Zonda Wind Occurrence with Vertical Sounding Data

Zonda wind is a typical downslope windstorm over the eastern slopes of the Central Andes in Argentina,which produces extremely warm and dry conditions and creates substantial socioeconomic *** aim of this work is to obtain an index for predicting the probability of Zonda wind *** Principal Component Analysis(PCA)is applied to the vertical sounding data on both sides of the *** the use of a binary logistic regression,the PCA is applied to discriminate those soundings associated with Zonda wind events from those that are not,and a probabilistic forecasting tool for Zonda occurrence is *** index is able to discriminate between Zonda and non-Zonda events with an effectiveness close to 91%.The best model consists of four variables from each side of the *** an eventbased statistical perspective,the probability of detection of the mixed model is above 97%with a probability of false detection lower than 7%and a missing ratio below 1%.From an alarm-based perspective,models exhibit false alarm rate below 7%,a missing alarm ratio lower than 1.5%and higher than 93%for the correct alarm *** zonal component of the wind on both sides of the Andes and the windward temperature are the key variables in class *** vertical structure of Zonda wind includes two wind maximums and an unstable lapse rate at midlevels on the lee side and a wind maximum at 700 h Pa accompanied by a relatively stable layer near the mountain top.
