ROS-responsive hydrogels with spatiotemporally sequential delivery of antibacterial and anti-inflammatory drugs for the repair of MRSA-infected wounds

For the treatment of MRSA-infected wounds,the spatiotemporally sequential delivery of antibacterial and anti-inflammatory drugs is a promising strategy.In this study,ROS-responsive HA-PBA/PVA(HPA)hydrogel was prepared by phenylborate ester bond cross-linking between hyaluronic acid-grafted 3-amino phenylboronic acid(HA-PBA)and polyvinyl alcohol(PVA)to achieve spatiotemporally controlled release of two kinds of drug to treat MRSA-infected wound.The hydrophilic antibiotic moxifloxacin(M)was directly loaded in the hydrogel.And hydrophobic curcumin(Cur)with anti-inflammatory function was first mixed with Pluronic F127(PF)to form Cur-encapsulated PF micelles(Cur-PF),and then loaded into the HPA hydrogel.Due to the different hydrophilic and hydrophobic nature of moxifloxacin and Cur and their different existing forms in the HPA hydrogel,the final HPA/M&Cur-PF hydrogel can achieve different spatiotemporally sequential delivery of the two drugs.In addition,the swelling,degradation,self-healing,antibacterial,anti-inflammatory,antioxidant property,and biocompatibility of hydrogels were tested.Finally,in the MRSA-infected mouse skin wound,the hydrogel-treated group showed faster wound closure,less inflammation and more collagen deposition.Immunofluorescence experiments further confirmed that the hydrogel promoted better repair by reducing inflammation(TNF-a)and promoting vascular(VEGF)regeneration.In conclusion,this HPA/M&CurPF hydrogel that can spatiotemporally sequential deliver antibacterial and anti-inflammatory drugs showed great potential for the repair of MRSA-infected skin wounds.

Clathrin-mediated endocytosis has been implicated in various physiological processes,including nutrient uptake,signal transduction,synaptic vesicle recycling,maintenance of cell polarity,and antigen presentation.Despite prior knowledge of its importance as a key regulator in promoting clathrin-mediated endocytosis,the physiological function of α-and γ-adaptin binding protein(aagab)remains elusive.In this study,we investigate the biological function of aagab during zebrafish development.We establish a loss-of-function mutant of aagab in zebrafish,revealing impaired swimming and early larval mortality.Given the high expression level of aagab in the brain,we probe into its physiological role in the nervous system.aagab mutants display subdued calcium responses and local field potential in the optic tectal neurons,aligning with reduced neurotransmitter release(e.g.,norepinephrine)in the tectal neuropil of aagab mutants.Overexpressing aagab mRNA or nervous stimulant treatment in mutants restores neurotransmitter release,calcium responses,swimming ability,and survival.Furthermore,our observations show delayed release of FM 1-43 in AAGAB knockdown differentiated neuroblastoma cells,pointing towards a probable link to defective clathrin-mediated synaptic vesicle recycling.In conclusion,our study underscores the significance of Aagab in neurobiology and suggests its potential impacts on neurological disorders.

The centrifugal pump is a prevalent power equipment widely used in different engineering patterns,and the impeller blade wrap angle significantly impacts its performance.A numerical investigation was conducted to analyze the influence of the blade wrap angle on flow characteristics and energy distribution of a centrifugal pump evaluated as a low specific speed with a value of 69.This study investigates six impellermodels that possess varying blade wrap angles(95°,105°,115°,125°,135°,and 145°)that were created while maintaining the same volute and other geometrical characteristics.The investigation of energy loss was conducted to evaluate the values of total and entropy generation rates(TEG,EGR).The fluid-structure interaction was considered numerically using the software tools ANSYS Fluent and ANSYSWorkbench.The elastic structural dynamic equation was used to estimate the structural response,while the shear stress transport k–ωturbulence model was utilized for the fluid domain modeling.The findings suggest that the blade wrap angle has a significant influence on the efficiency of the pump.The impeller featuring a blade wrap angle of 145°exhibits higher efficiency,with a notable increase of 3.76%relative to the original model.Variations in the blade wrap angle impact the energy loss,shaft power,and pump head.The model with a 145°angle exhibited a maximum equivalent stress of 14.8MPa and a total deformation of 0.084 mm.The results provide valuable insights into the intricate flow mechanism of the centrifugal pump,particularly when considering various blade wrap angles.

In this article,we explore the famous Selkov–Schnakenberg(SS)system of coupled nonlinear partial differential equations(PDEs)for Lie symmetry analysis,self-adjointness,and conservation laws.Moreover,miscellaneous soliton solutions like dark,bright,periodic,rational,Jacobian elliptic function,Weierstrass elliptic function,and hyperbolic solutions of the SS system will be achieved by a well-known technique called sub-ordinary differential equations.All these results are displayed graphically by 3D,2D,and contour plots.

Sleep posture surveillance is crucial for patient comfort,yet current systems face difficulties in providing compre-hensive studies due to the obstruction caused by blankets.Precise posture assessment remains challenging because of the complex nature of the human body and variations in sleep patterns.Consequently,this study introduces an innovative method utilizing RGB and thermal cameras for comprehensive posture classification,thereby enhancing the analysis of body position and comfort.This method begins by capturing a dataset of sleep postures in the form of videos using RGB and thermal cameras,which depict six commonly adopted postures:supine,left log,right log,prone head,prone left,and prone right.The study involves 10 participants under two conditions:with and without blankets.Initially,the database is normalized into a video frame.The subsequent step entails training a fine-tuned,pretrained Visual Geometry Group(VGG16)and ResNet50 model.In the third phase,the extracted features are utilized for classification.The fourth step of the proposed approach employs a serial fusion technique based on the normal distribution to merge the vectors derived from both the RGB and thermal datasets.Finally,the fused vectors are passed to machine learning classifiers for final classification.The dataset,which includes human sleep postures used in this study’s experiments,achieved a 96.7%accuracy rate using the Quadratic Support Vector Machine(QSVM)without the blanket.Moreover,the Linear SVM,when utilized with a blanket,attained an accuracy of 96%.When normal distribution serial fusion was applied to the blanket features,it resulted in a remarkable average accuracy of 99%.

Conformance control or water shut-off is a technique used to improve oil recovery.During conformance control,polymers block high permeability water areas and redistribute water drive toward unswept oil zones.In this study,co-polymers(denoted ATP-PGV/AM-co-AMPS)were synthesized using acrylamide(AM)and 2-acrylamide-2-methylpropane sulfonic acid(AMPS)as the monomers,polyethylene glycol(PEG)-200 and methylenebisacrylamide(MBA)as the crosslinkers,attapulgite(ATP)and bentonite(PGV)as the clay types,and ammonium persulfate(APS)as the initiator,in addition to paraffin oil and surfactants.Samples were synthesized using inverse emulsion polymerization with different concentrations of monomers,crosslinkers,and clays,and they were characterized using Fourier-transform infrared(FTIR)spectroscopy and scanning electron microscopy(SEM)with energy dispersive X-ray spectroscopy(EDX).FTIR spectra of the samples confirmed the existence of sulfonate and hydroxyl groups,which are important for polymer swelling.SEM-EDX images indicated that the morphology and elemental composition were different before and after swelling,confirming the occurrence of swelling.Moreover,samples were placed in sodium chloride solution(20,000 ppm)for 7 days to evaluate swelling at both room temperature and 90°C.Thermogravimetric analysis(TGA)and differential scanning calorimetry(DSC)were used to determine the thermal characteristics of the microparticles.Finally,rheological measurements were used to assess the deformation and rheological behavior of the hydrogels.The results showed that after 1 day,good swelling without loss of mechanical strength was achieved with the composite synthesized using 10%AM,15%AMPS,6%PGV,and 10%ATP.

In this work,we have explored wormhole(WH)solutions in F(R,L_(m))gravity by assuming the Morris-Thorne WH metric and F(R,Lm)=R/2+(1+γR)L_(m),whereγis the free model parameter.We determined the WH solutions by utilizing two newly developed shape functions(SF)that satisfy all basic conditions for a WH’s physical validity.We also observe that the null energy condition(NEC)behaves negatively.Finally,for both models,we use the volume integral quantifier(VIQ)and Tolman-Oppenheimer-Volkoff(TOV)equation to determine how much exotic matter is needed near the WH throat and the stability of the WH.The extensive detailed discussions of the matter components have been done via graphical analysis.The obtained WH geometries meet the physically acceptable conditions for a stable wormhole.

Confining stresses serve as a pivotal determinant in shaping the behavior of grouted rock bolts.Nonetheless,prior investigations have oversimplified the three-dimensional stress state,primarily assuming hydrostatic stress conditions.Under these conditions,it is assumed that the intermediate principal stress(σ_(2))equals the minimum principal stress(σ_(3)).This assumption overlooks the potential variations in magnitudes of in situ stress conditions along all three directions near an underground opening where a rock bolt is installed.In this study,a series of push tests was meticulously conducted under triaxial conditions.These tests involved applying non-uniform confining stresses(σ_(2)≠σ_(3))to cubic specimens,aiming to unveil the previously overlooked influence of intermediate principal stresses on the strength properties of rock bolts.The results show that as the confining stresses increase from zero to higher levels,the pre-failure behavior changes from linear to nonlinear forms,resulting in an increase in initial stiffness from 2.08 kN/mm to 32.51 kN/mm.The load-displacement curves further illuminate distinct post-failure behavior at elevated levels of confining stresses,characterized by enhanced stiffness.Notably,the peak load capacity ranged from 27.9 kN to 46.5 kN as confining stresses advanced from σ_(2)=σ_(3)=0 to σ_(2)=20 MPa and σ_(3)=10 MPa.Additionally,the outcomes highlight an influence of confining stress on the lateral deformation of samples.Lower levels of confinement prompt overall dilation in lateral deformation,while higher confinements maintain a state of shrinkage.Furthermore,diverse failure modes have been identified,intricately tied to the arrangement of confining stresses.Lower confinements tend to induce a splitting mode of failure,whereas higher loads bring about a shift towards a pure interfacial shear-off and shear-crushed failure mechanism.

The primary varicella-zoster virus(VzV)infection that causes chickenpox(also known as varicella),spreads quickly among people and,in severe circumstances,can cause to fever and encephalitis.In this paper,the Mittag-Leffler fractional operator is used to examine the mathematical representation of the vzV.Five fractional-order differential equations are created in terms of the disease's dynamical analysis such as S:Susceptible,V:Vaccinated,E:Exposed,I:Infectious and R:Recovered.We derive the existence criterion,positive solution,Hyers-Ulam stability,and boundedness of results in order to examine the suggested fractional-order model's wellposedness.Finally,some numerical examples for the VzV model of various fractional orders are shown with the aid of the generalized Adams-Bashforth-Moulton approach to show the viability of the obtained results.
