Gene-modified leucoconcentrate for personalized ex vivo gene therapy in a mini pig model of moderate spinal cord injury

We previously demonstrated that gene-modified umbilical cord blood mononuclear cells overexpressing a combination of recombinant neurotrophic factors are a promising therapeutic approach for cell-mediated gene therapy for neurodegenerative diseases,neurotrauma,and *** this study,using a mini pig model of spinal cord injury,we proposed for the first time the use of gene-modified leucoconcentrate prepared from peripheral blood in the plastic blood bag for personalized ex vivo gene *** obtained from mini pig peripheral blood was transduced with a chimeric adenoviral vector(Ad5/35 F)that carried an enhanced green fluorescent protein(EGFP)reporter gene in the plastic blood *** day after blood donation,the mini pigs were subjected to moderate SCI and four hours post-surgery they were intravenously autoinfused with gene-modified leucoconcentrate.A week after gene-modified leucoconcentrate therapy,fluorescent microscopy revealed EGFP-expressing leucocytes in spinal cord at the site of contusion *** the spleen the groups of EGFP-positive cells located in the lymphoid follicles were *** vitro flow cytometry and fluorescent microscopy studies of the gene-modified leucoconcentrate samples also confirmed the production of EGFP by ***,the efficacy of leucocytes transduction in the plastic blood bag and their migratory potential suggest their use for temporary production of recombinant biologically active molecules to correct certain pathological *** paper presents a proof-of-concept of simple,safe and effective approach for personalized ex vivo gene therapy based on gene-modified leucoconcentrate *** animal protocols were approved by the Kazan State Medical University Animal Care and Use Committee(approval No.5)on May 27,2014.

Combination of epidural electrical stimulation with ex vivo triple gene therapy for spinal cord injury:a proof of principle study

Despite emerging contemporary biotechnological methods such as gene-and stem cell-based therapy,there are no clinically established therapeutic strategies for neural regeneration after spinal cord *** previous studies have demonstrated that transplantation of genetically engineered human umbilical cord blood mononuclear cells producing three recombinant therapeutic molecules,including vascular endothelial growth factor(vEGF),glial cell-line derived neurotrophic factor(GDNF),and neural cell adhesion molecule(NCAM)can improve morpho-functional recovery of injured spinal cord in rats and *** investigate the efficacy of human umbilical cord blood mononuclear cells-mediated triple-gene therapy combined with epidural electrical stimulation in the treatment of spinal cord injury,in this study,rats with moderate spinal cord contusion injury were intrathecally infused with human umbilical cord blood mononuclear cells expressing recombinant genes vEGF165,GDNF,NCAM1 at 4 hours after spinal cord *** days after injury,epidural stimulations were given simultaneously above the lesion site at C5(to stimulate the cervical network related to forelimb functions)and below the lesion site at L2(to activate the central pattern generators)every other day for 4 *** subjected to the combined treatment showed a limited functional improvement of the knee joint,high preservation of muscle fiber area in tibialis anterior muscle and increased H/M ratio in gastrocnemius muscle 30 days after spinal cord ***,beneficial cellular outcomes such as reduced apoptosis and increased sparing of the gray and white matters,and enhanced expression of heat shock and synaptic proteins were found in rats with spinal cord injury subjected to the combined epidural electrical stimulation with gene *** study presents the first proof of principle study of combination of the multisite epidural electrical stimulation with ex vivo triple gene therapy(vEGF,GDNF and NCAM

Handheld diffuse optical breast scanner probe for cross-sectional imaging of breast tissue

Diffuse optical spectroscopy is a relatively new,noninvasive and nonionizing technique for breast cancer *** the present study,we have introduced a novel handheld diffuse optical breast scan(DOB-Scan)probe to measure optical properties of the breast in vivo and create functional and compositional images of the *** addition,the probe gives more information about breast tissue's constituents,which helps distinguish a healthy and cancerous *** symmetrical light sources,each including four different wavelengths,are used to illuminate the breast tissue.A high-resolution linear array detector measures the intensity of the back-scattered photons at different radial destinations from the illumination sources on the surface of the breast tissue,and a unique image reconstruction algorithm is used to create four cross-sectional images for four different *** fromfiber optic-based illumination techniques,the proposed method in this paper integrates multi-wavelength light-emitting diodes to act as pencil beam sources into a scattering medium like breast *** unique design and its compact structure reduce the complexity,size and cost of a potential *** the introduced technique miniaturizes the probe,this study points to the reliability of this technique in the phantom study and clinical breast *** have received ethical approval to test the DOB-Scan probe on patients and we are currently testing the DOB-Scan probe on subjects who are diagnosed with breast cancer.

Development of a handheld diffuse optical breast cancer assessment probe

Diffuse Optical Spectroscopy(DOS)is a promising non-invasive and non-ionizing technique for breast anomaly *** this study,we have developed a new handheld DOS probe to measure optical properties of breast *** the proposed probe,the breast tissue is illuminated with four near infrared(NIR)wavelengths light emitting diodes(LED),which are encapsulated in a package(eLEDs),and two PIN photodiodes measure the intensity of the scattered photons at two di®erent *** proposed technique of using eLEDs is introduced,in order to have a multi-wavelength pointed-beam illumination source instead of using the laser-coupledflberoptic technique,which increases the complexity,size,and cost of the *** the fact that the proposed technique miniaturizes the probe and reduces the complexity of the DOS,the study proves that it is accurate and reliable in measuring optical properties of the *** measurements are performed at the rate of 10 Hz which is suitable for dynamic measurement of biological activity,*** multi-spectral evaluation algorithm is used to reconstruct four main absorber concentrations in the breast including oxy-hemoglobin(cHb),deoxy-hemoglobin(cHbO_(2)),water(cH2O),fat(cFat),and average scattering coeficient of the medium,as well as concentration changes in Hb(cHb)and HbO_(2)(cHbO_(2)).Although the probe is designed for breast cancer diagnosis,it can be used in a wide range of applications for both static and dynamic measurements such as functional brain imaging.A series of phantoms,comprised of Delrinr,Intralipidr,PierceTM and Black ink,are used to verify performance of the *** probe will be tested on human subjects,in-vivo,in the next phase.

The aim of this work is to model and analyze the behavior of a new smart nano force *** do so,the carbon nanotube has been used as a suspended gate of a metal-oxide-semiconductor field-effect transistor(MOSFET).The variation of the applied force on the carbon nanotube(CNT)generates a variation of the capacity of the transistor oxide-gate and therefore the variation of the threshold voltage,which allows the MOSFET to become a capacitive nano force *** sensitivity of the nano force sensor can reach 0.12431v/*** sensitivity is greater than results in the *** have found through this study that the response of the sensor depends strongly on the geometric and physical parameters of the *** the results obtained in this study,it can be seen that the increase in the applied force increases the value of the MOSFET threshold voltage *** this paper,we first used artificial neural networks to faithfully reproduce the response of the nano force sensor *** neural model is called direct ***,secondly,we designed an inverse model called an intelligent sensor which allows linearization of the response of our developed force sensor.

An entropy-stabilized multicomponent ultrahigh-temperature ceramic(UHTC)coating,(Ti_(0.25)v_(0.25)Zr_(0.25)Hf_(0.25))B_(2),on a graphite substrate was in-situ sintered by spark plasma sintering(SPs)from constituent transition metal diboride ***(Ti_(0.25)v_(0.25)Zr_(0.25)Hf_(0.25))B_(2) coating had a hardness of 31.2±2.1 GPa and resisted 36.9 GPa of stress before delamination,as observed at the *** temperature-dependent thermal properties of the multicomponent diboride(Ti_(0.25)v_(0.25)Zr_(0.25)Hf_(0.25))B_(2) were obtained by molecular dynamics(MD)simulations driven by a machine learning force field(MLFF)trained on density functional theory(DFT)*** thermal conductivity,density,heat capacity,and coefficient of thermal expansion obtained by the MD simulations were used in time-dependent thermal stress finite element model(FEM)*** low thermal conductivity(visible cracking or *** ablation mechanisms were mechanical denudation and evaporation of B_(2)O_(5) and light v-Ti oxides,which caused a decrease in the mass and thickness of the coating and resulted in mass and linear ablation rates of-0.51 mg·s^(-1) and -1.38μm·s^(-1),respectively,after 60 *** findings demonstrated the thermal and mechanical stability of multicomponent entropy-stabilized diborides as coatings for carbon materials in engineering components under extreme environments.

Influence of growth conditions on the electrochemical synthesis of SnS thin films and their optical properties

Tin sulfide （SnS） thin films were prepared by electrodeposition onto fluorine-doped tin oxide （FTO） glass substrates using an aqueous solution containing SnCl2 and Na2S2O3 at various deposition potentials （L） and bath concentrations. The pH value and temperature of the solution were kept constant. The deposited films were characterized using X-ray diffraction （XRD）, field-emission scanning electron microscopy （FESEM）, photoluminescence （PL）, and ultraviolet-visible （Uv-vis） spectroscopy. The FESEM images demonstrated that changes in the deposition potential （E） and solution concentration led to marked changes in the morphology of the deposited SnS films. Energy-dispersive X-ray analysis （EDXA） results showed that the SrdS atomic ratio strongly depended on both the solution concentration and the deposition potential. To obtain an SrdS atomic ratio approximately equal to l, the optimal Sn2＋/S2O2- 3 molar ratio and E parameter were 1/8 and -1.0 v, respectively. The XRD patterns showed that the synthesized SnS was obviously polycrystalline, with an orthorhombic structure. The effects of the variations of bath concentration and deposition potential on the band-gap energy （Eg） were studied using PL and Uv-vis experiments. The PL spectra of all the SnS films contained two peaks in the visible region and one peak in the infrared （IR） region. The Uv-vis spectra showed that the optical band-gap energy varies from 1.21 to 1.44 ev.

The present study reports the titanium dioxide(TiO_(2))films synthesized from TiCl3 precursor on ITO glass substrates using two electrochemical techniques,namely direct electro-deposition(DE)and pulse electro-deposition(PE).The synthesis potential during the time-on(Ton)period was fixed at-1.5 ***,the open-circuit potential was applied during the time-off(Toff)*** effect of the technique of electro-deposition and Toff duration on the properties of TiO_(2)films and their photoelectron-catalytic activity were *** obtained films were investigated by X-ray diffraction(XRD),scanning electron microscopy(SEM),Uv-vIS spectrometer,and photocurrent measurement *** is found that the use of the PE technique at different Toff improves the properties of TiO_(2)films compared to the DE *** XRD patterns show the anatase phase with a marked preferential orientation along the(101)direction for all *** the SEM analysis,it is seen a significant change from big multigrain agglomerates at DE to a dense film structure and small multigrain agglomerates at different *** the Toff decreases from 3 s to 1 s,the photocurrent response rises and reaches a high value of about 12 m A/cm^(2).Compared with DE,and under Uv light the photocatalytic property of TiO_(2)film synthesis via PE has been improved in the degradation of methyl orange(MO).Finally,the films deposited at low Toff(Toff=1 s)show a faster degradation of MO.
