Marek’s disease virus-encoded microRNAs: genomics, expression and function

MicroRNAs (miRNAs) are the recently discovered small non-coding RNA molecules that have post-transcriptional regulatory functions in many important biological processes. A large number of miRNAs have been found to be encoded by viral genomes, especially in herpesviruses. Previous research regarding miRNAs encoded by herpesviruses, including Marek’s disease virus (MDV), has demonstrated their involvement in lytic replication, latent infection, T-lymphocyte transformation and tumorigenesis. MDV is an oncogenic alphaherpesvirus, with the ability to induce tumors in natural hosts; however, formation of these tumors can be prevented by immunization with attenuated or nonpathogenic forms of the virus. Marek’s disease is considered to be a good biomedical model for investigating the biology, genetics, and immunology of tumorigenesis. In this paper, we review the discovery and identification of MDV-encoded miRNAs, along with their genomics, expression profiles, and currently known functions. We also discuss the prospects and techniques possibly applicable to the further investigation of the biological roles of MDV-encoded miRNAs.

目的随着人们对代谢性疾病的广泛关注,已有诸多研究表明在一些代谢性疾病中,胆汁酸以及胆汁酸信号通路已经成为潜在的治疗靶点,例如:糖尿病、肥胖以及胆结石病。糖尿病脑病是糖尿病的一种并发症,是一种复杂的代谢性疾病,伴随着中枢神经和脑功能损伤,表现为认知功能障碍,使得患老年痴呆的风险增加。对糖尿病脑病的发病机制以及临床上的治疗方法已有很多报道,主要包括对脑组织中的各种神经细胞及神经递质的研究,以及临床上采用的中枢神经刺激的治疗方法。但是,至今仍没有胆汁酸代谢在糖尿病脑病的发生和发展过程中的作用的相关报道。所以,本课题的研究目的是在糖尿病脑病这种复杂的疾病模型中探索胆汁酸代谢及其信号通路。有文献表明,糖尿病的发生会导致迷走神经的病变,而迷走神经对于胆汁酸的代谢和信号通路的影响尚没有系统的研究。作为寻找糖尿病脑病的潜在预防治疗靶点的第一步,我们试着探寻迷走神经在胆汁酸体内平衡中的调节作用。方法为了定量检测生物组织和体液中的胆汁酸浓度,进而对生物体内的胆汁酸代谢进行分析,首先建立了简单有效的液相色谱-质谱联用(LC-MS)方法。该方法经过了线性、稳定性、准确度、精密度、检测线(LOD)、定量限(LOQ)、回收率、基质效应等验证。生物体内组织和体液中胆汁酸的提取采用简便的四步提取法:1.乙腈沉淀蛋白;2.震荡提取;3.离心后上清干燥;4.50%甲醇-水(双蒸水)复溶,样品前处理完成后等待进样检测。糖尿病脑病动物模型的建立采用C57BL/6小鼠,实验组小鼠长期喂养高脂饮食,糖尿病组和糖尿病脑病组小鼠经单次注射链脲霉素(STZ)诱导。模型构建后期,用水迷宫试验(Morris water maze test)判断小鼠疾病阶段,将小鼠分成四组,分别为正常组(NC)、高脂饮食组(HFD)、糖尿病组(unDCD)及糖尿病脑病组(DCD)。模型建立成功后,将小鼠麻醉并收集小鼠肝脏、肠道及结肠。用液相色谱-质谱联用方法(LC-MS)定量测定肝脏和肠道中胆汁酸的浓度;并采用逆转录聚合酶链式反应实时荧光定量技术(RT-PCR)和蛋白免疫印迹技术(Western Blot)检测肝脏和肠道中与胆汁酸相关的基因表达情况;采用定量PCR技术检测分析结肠内容物中细菌总量的变化和各类细菌百分比含量的变化。为了探究迷走神经对胆汁酸代谢以及信号通路的调控作用,本研究继而建立了大鼠(Wistar)膈下迷走神经切断模型,模型构建成功后,对大鼠进行麻醉,收集胆汁、血液、肝脏、肠道以及结肠。采用液相色谱-质谱联用技术定量测定胆汁、肝脏以及血清中胆汁酸浓度。并采用逆转录聚合酶链式反应实时荧光定量技术和蛋白免疫印迹技术检测肝脏和肠道中胆汁酸相关的基因表达情况。结果糖尿病脑病小鼠模型建立过程中,观察到高脂饲料喂养持续五周后,四组小鼠中的体重差异显现,高脂饮食组(HFD)小鼠体重相比于正常饮食组(NC)小鼠体重显著增加了19%,然而糖尿病(unDCD)和糖尿病脑病(DCD)组小鼠体重却有所减轻,尤其糖尿病脑病组小鼠体重比正常组小鼠显著下降了12%。饲养第六周时,三组实验组小鼠的空腹血糖水平相比正常对照组升高,糖尿病组和糖尿病脑病组小鼠血糖相比于高脂喂养组小鼠血糖均显著升高了约1.3倍。血脂检测中,实验组小鼠的总胆固醇含量相比正常组显著升高;甘油三酯的含量在糖尿病脑病小鼠中显著升高,在糖尿病小鼠中升高但无统计学意义;相比正常组,低密度脂蛋白含量在糖尿病和糖尿病脑病小鼠中均显著性升高,高密度脂蛋白在糖尿病小鼠中显著升高,在脑病小鼠中增加不明显(约44%)。在用于观察小鼠学习及记忆能力的水迷宫实验中,得到如下结果:在为期五天的观察中,从第三天开始小鼠的记忆能力差异显现,糖尿病脑病组小鼠逃避潜伏时间显著长于其它三组小鼠;首次到达平台的时间显著高于正常组,相比糖尿病组小鼠慢了约3s;穿越平台的次数相比高脂饮食组和糖尿病组减少一次,相比正常组小鼠减少四次。糖尿病脑病模型研究中胆汁酸浓度定量检测的结果显示,同正常组(NC)以及高脂饮食组(HFD)相比,在糖尿病(unDCD)和糖尿病脑病(DCD)小鼠的肝脏和肠道中总胆汁酸的含量均明显升高,尤其在糖尿病脑病小鼠中总胆汁酸的升高存在统计学差异。糖尿病小鼠肝脏中总胆汁酸的升高主要是由于如下几类胆汁酸含量的增加引起的,TMCA增加了259%,TMDCA增加了153%,TCDCA增加了342%,然而几种非结合型胆汁酸显著降低,CA降低60%,βMCA降低38%,DCA降低80%,导致肝脏中非结合型胆汁酸总量降低了55%。糖尿病脑病小鼠肝脏中不仅总胆汁酸含量升高,初级胆汁酸和结合型胆汁酸的总量也分别升高,有几种胆汁酸浓度变化明显,TMCA增加了212%,TCA增加了102%,TCDCA增加了279%,CA增加了39%,CDCA增加了46%,isoCDCA增加了403

GUoblastoma multiforme （GBM） is a highly invasive brain tumor with limited therapeutic means and poor prognosis. Recent stud- ies indicate that glioma-initiating ceUs/gUoma stem ceils （GICs/GSCs） may be responsible for tumor initiation, infiltration, and recurrence. GICs could aberrantly employ molecular machinery balancing self-renewal and differentiation of embryonic neural precursors. Here, we find that paired related homeobox 1 （PRRX1）, a homeodomain transcription factor that was previously reported to control skeletal development, is expressed in cortical neural progenitors and is required for their self-renewal and proper differentiation. Further, PRRX1 is overrepresented in gUoma samples and labels GlCs. Gtioma celts and GlCs depleted with PRRX1 could not propagate in vitro or form tumors in the xenograft mouse model. The GIC self-renewal function regulated by PRRX1 is mediated by dopamine D2 receptor （DRD2）. PRRX1 directly binds to the DRD2 promoter and transactivates its expression in GICs. Blockage of the DRD2 signaling hampers GIC self-renewal, whereas its overexpression restores the propagating and tumorigenic potential of PRRXl-depleted GlCs. Finally, PRRX1 potentiates GICs via DRD2-mediated extracetlutar signal-related kinase （ERK） and AKT activation. Thus, our study suggests that therapeutic targeting the PRRX1-DRD2-ERK/AKT axis in GICs is a promising strategy for treating GBMs.

Corystosperms, an extinct group of “seed ferns” that lived from the late Permian to the Cretaceous or Eocene, are significant for seed plant phylogeny for their unique roles in partially resolving the long-standing question of angiosperm origins.A new fossil wood taxon, Fengweioxylon sinense gen. et sp. nov. is reported here based on exceptionally preserved wood material from the Jurassic in western Liaoning, China, whose peculiar wood structure enables it to be recognized as the first unequivocal fossil wood record of a corystosperm plant in Laurasia. The anatomical structures of this corystosperm wood show gymnospermous characters. The discovery of the new specimen will provide more anatomical evidence for understanding the systematic and affinity of this seed plants.

Bimetallic Pd-Ni core-shell nanoparticles catalysts for the Suzuki reaction

A simple and efficient solution-based method for the synthesis of Pd-Ni bimetallic nanoparticles （NPs） has been developed. A series of Pd-Ni bimetallic NPs were readily achieved by reduction of PdC12 and Ni（acac）2 （acac = acetyl- acetonate） in the presence of oleylamine （OAm）, oleic acid （OA） and benzyl alcohol. Furthermore, by using high-resolution transmission electron microscopy （HRTEM）, energy-dispersive spectrometry （EDS） mapping and X-ray diffraction （XRD）, we demonstrate that the as-prepared Pd-Ni bimetallic NPs have core-shell structures with a Pd-rich core and a Ni-rich shell. In addition, the as-obtained Pd-Ni bimetallic NPs with varying compositions show excellent catalytic activities in the Miyaura-Suzuki reaction. When the nickel molar percentage was 0.23 to 0.65, the conversion with the as-obtained Pd-Ni bimetallic catalysts was above 90%. It is believed that this strategy can be employed to produce a variety of other well-defined core-shell type multimetallic nanostructures.

Background : We have discovered that patients who suffered from ulcerative colitis(UC) may also accompany with anxiety and depression, which affect the quality life. According to previous researches, anxiety and depression are usually along with brain functional changes. In order to observe the relationship between brain functional changes and anxiety, depression especially in UC patients, a functional magnetic resonance imaging(f-MRI) technology was applied to conduct the study. The purpose of this study was to investigate the spontaneous brain activities and influence factors in UC patients compared with healthy people. Methods: 27 patients diagnosed as UC were recruited from jinling hospital, 17 of them and 17 matched healthy controls with similar gander, age and years of education were underwent a functional magnetic resonance imaging(f-MRI) 3.0 T scan. Regional homogeneity(ReHo) was used to measure the synchronism of regional brain activities. T test, Chi-square test, pearson correlation were used to analyse the data. Results: On one hand, a significant decreased ReHo values in Parietalnf(except supramarginal gyrus and angular convolution) and Parietalup were found in UC patients compared with healthy controls. Moreover, there were significant negative correlations between ReHo values and SAS scores(Pearson correlation, p=0.019,0.05). On the other hand, there is statistical significance in SAS(42.72±12.543) and SDS(38.75(33.75-53.13)) scores between UC patients and healthy controls. Moreover, years of education revealed statistical significance in SAS scores(c2=9.047,p=0.009<0.05) between UC patients and healthy controls. No statistical significance was found in SDS scores between several factors in UC patients and healthy people.
