Disulfiram ameliorates STING/MITA-dependent inflammation and autoimmunity by targeting RNF115

STING(also known as MITA)is an adaptor protein that mediates cytoplasmic DNA-triggered signaling,and aberrant activation of STING/MITA by cytosolic self-DNA or gain-of-function mutations causes severe ***,we show that STING-mediated inflammation and autoimmunity are promoted by RNF115 and alleviated by the RNF115 inhibitor disulfiram(DSF).Knockout of RNF115 or treatment with DSF significantly inhibit systemic inflammation and autoimmune lethality and restore immune cell development in Trex1^(–/–)mice and STING^(N153S/WT) bone marrow chimeric *** addition,knockdown or pharmacological inhibition of RNF115 substantially downregulate the expression of IFN-α,IFN-γand proinflammatory cytokines in PBMCs from patients with systemic lupus erythematosus(SLE)who exhibit high concentrations of dsDNA in peripheral ***,knockout or inhibition of RNF115 impair the oligomerization and Golgi localization of STING in various types of cells transfected with cGAMP and in organs and cells from Trex1^(–/–)***,knockout of RNF115 inhibits the activation and Golgi localization of STINGN153S as well as the expression of proinflammatory cytokines in myeloid cells but not in endothelial cells or *** together,these findings highlight the RNF115-mediated cell type-specific regulation of STING and STINGN153S and provide potential targeted intervention strategies for STING-related autoimmune diseases.

Various SARS-CoV-2-related coronaviruses have been increasingly identified in pangolins,showing a potential threat to *** we report the infectivity and pathogenicity of the SARS-CoV-2-related virus,PCoV-GX/P2V,which was isolated from a Malayan pangolin(Manis javanica).PCoV-GX/P2V could grow in human hepatoma,colorectal adenocarcinoma cells,and human primary nasal epithelial *** replicated more efficiently in cells expressing human angiotensin-converting enzyme 2(hACE2)as SARS-CoV-2 *** intranasal inoculation to the hACE2-transgenic mice,PCoV-GX/P2V not only replicated in nasal turbinate and lungs,but also caused interstitial pneumonia,characterized by infiltration of mixed inflammatory cells and multifocal alveolar *** population immunity established by SARS-CoV-2 infection and vaccination may not protect people from PCoV-GX/P2V *** findings further verify the hACE2 utility of PCoV-GX/P2V by in vivo experiments using authentic viruses and highlight the importance for intensive surveillance to prevent possible cross-species transmission.

Component-based software reuse and development is considered to be the best way to improve the efficiency and quality of *** meet the demands of aircraft instrument exhibition in an aviation virtual library,we present a modeling and simulation tool for cockpit instruments on component methods,achieving display functionality and providing a visual interaction platform in the aviation virtual *** the rapid model construction and reuse of virtual library resources,we classify entities in the cockpit into several categories by their behavior,and design a template for each category to describe its *** efficiently reduces the modeling cycle,and thus largely helps users to take advantage of the library resource.

Newborns' sleep-wake cycle development on amplitude integrated electroencephalography

Background:To observe the development of neonatal sleep among healthy infants of different conceptional age(CA)by analyzing the amplitude-integrated electroencephalography(aEEG)of their sleep-wake cycles(SWC).Methods:Bedside aEEG monitoring was carried out for healthy newborns from 32 to 46 weeks CA between September 1,2011 and August 30,*** each aEEG tracing,mean duration of every complete SWC,number of SWC repetition within 12 hours,mean duration of each narrow and broadband of SWC,mean voltage of the upper edge and lower edge of SWC,mean bandwidth of SWC were counted and *** of the correlations between voltages or bandwidth of SWC and CA was performed to assess the developmental changes of central nervous system of newborns with different ***:The SWC of different CA on aEEG showed clearly identifiable trend after 32 weeks of *** occurrence of SWC gradually increases from preterm to post-term infants;term infants had longer SWC *** voltage of upper edge of the broadband decreased at 39 weeks,while the lower edge voltage increases and the bandwidth of broadband declined along with the growing *** upper edge of the narrowband dropped while the lower edge rised gradually,especially in preterm *** width of the narrowband narrowed down while CA ***:The SWC on aEEG of 32-46 weeks infants showed a continuous,dynamic and developmental *** appearance of SWC and the narrowing bandwidth of narrowband is the main indicator to identify the CA-dependent SWC from the preterm to the late preterm *** lower edge of the broadband identifi es the term to post-term period.

A novel collector 1-(2-hydroxyphenyl)hex-2-en-1-one oxime(HPHO)was synthesized from 2-hydroxy acetophenone and *** flotation performance and adsorption mechanism to malachite were investigated by flotation test,zeta potential,Fourier transform infrared(FTIR)and X-ray photoelectron spectroscopy(XPS)analysis *** with benzohydroxamic acid(BA),1-(2-hydroxyphenyl)ethan-1-one oxime(HPEO)and sodium isobutyl xanthate(SIBX),HPHO exhibited excellent collecting power to malachite without additional reagents,such as Na2S regulator and methyl isobutyl carbinol(MIBC)*** of zeta potential indicated that HPHO was coated on malachite surfaces through a chemisorption *** and XPS data gave clear evidence for the formation of Cu−oxime complex on malachite surfaces after HPHO adsorption through the linkage between C=C,—OH,N—OH group and Cu species.

Clinical and pathological portraits of axillary presentation breast cancer and effects of preoperative systemic therapy

There is a lack of investigation into the biological characteristics and preoperative systemic therapy （PST） for occult breast cancer （OBC）. For this study, departmental records in Breast Disease Center of Peking University First Hospital from January 2008 to December 2015 were retrospectively reviewed to identify cases of OBC. Eleven cases were included, and all patients were female, with a median age of 56 （range： 29-75） years. The sensitivity of magnetic resonance imaging （MRD was I00%, and the false positive rate was 33.3%. Based on histologic analysis of the axillary node, 9 （81.8%） cases were grade 3, and 2 （18.2%） cases were grade 2; 4 （36.4%） cases were 〉10% estrogen receptor （ER） positive and 6 （54.5%） human epidermal growth receptor 2 （HER2） positive. Nine cases （81.8%） exhibited over 30% Ki67 expression. PST was performed in 5 of the 11 cases. The lymph node response rate was 100% （5/5）, but no complete remission was achieved. In conclusion, aggressive subtypes were predominant among the included cases, and PST should be considered for OBC treatment options.

Ocean bottom node(OBN)data acquisition is the main development direction of marine seismic exploration;it is widely promoted,especially in shallow sea ***,the OBN receivers may move several times because they are easily affected by tides,currents,and other factors in the shallow sea environment during long-term *** uncorrected,then the imaging quality of subsequent processing will be *** conventional secondary positioning does not consider the case of multiple movements of the receivers,and the accuracy of secondary positioning is *** first arrival wave of OBN seismic data in shallow ocean mainly comprises refracted *** this study,a nonlinear model is established in accordance with the propagation mechanism of a refracted wave and its relationship with the time interval curve to realize the accurate location of multiple receiver *** addition,the Levenberg-Marquart algorithm is used to reduce the influence of the first arrival pickup error and to automatically detect the receiver movements,identifying the accurate dynamic relocation of the *** simulation and field data show that the proposed method can realize the dynamic location of multiple receiver movements,thereby improving the accuracy of seismic imaging and achieving high practical value.

In-orbit performance of HE onboard Insight-HXMT in the first 5 years

Purpose The high-energy X-ray telescope(HE),one of the three main payloads of the Insight-HXMT mission,is composed of eighteen NaI(Tl)/CsI(Na)phoswich detectors,where NaI(Tl)serves as the primary detector covering 20–250 keV,and CsI(Na)is used as an active shield detector to suppress the background of NaI(Tl)and also serves as an all-sky gammaray burst monitor covering 0.2–*** this paper,we review the in-orbit performance of HE in the first 5 years since Insight-HXMT was launched on June 15,*** The major performances we concern include the gain and energy resolution of NaI(Tl)and CsI(Na)detectors,the performance of pulse shape discriminator(PSD)and system dead *** this work,we investigate these performances mainly using the data of blank-sky observations and the data when the telescope is in earth *** The overall performance of HE/NaI(Tl)is very stable in the first 5 years,whereas the gain of HE/CsI(Na)shows a continuously increasing trend and should be calibrated *** In general,HE is still in good health and well-calibrated status after five-year’s *** in-orbit performance of HE has no significant deviation from *** is expected to be in operation healthily for another several years of extended mission life.
