Transcriptome profile and its partly verification of human hepatocellular carcinoma cells exposed to Yuzhizi(Fructus Akebiae Quinatae) seed extract

OBJECTIVE: To investigate the efficacy of Yuzhizi(Fructus Akebiae Quinatae) seed extract(FAQSE) on inhibiting the proliferation of hepatocellular carcinoma(HCC) cells in vitro and to explore the anti-HCC action mechanism of FAQSE. METHODS: Human HCC Hep G2 and Huh7 cells were used to investigate the anti-HCC effect of FAQSE in vitro. 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) method was used to measure cell viability. Affymetrix microarray was adopted to detect the expression of transcriptome. The differentially expressed genes(DEGs) of each cell line were identified. For coDEGs of both cell lines, the gene ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway were enriched using the Database for Annotation, Visualization and Integrated Discovery(DAVID), and the network analysis of protein-protein interaction(PPI) was mapped using the Retrieval of Interacting Genes/Proteins(STRING) and Cytoscape software. Some important genes in the PPI network of coDEGs were selected to verify by quantitative real-time reverse transcription-polymerase chain reaction, Western blot and enzyme-linked immunosorbent assay. RESULTS: FAQSE decreased the viability of Hep G2 and Huh7 cells. There were 211 co-upregulated and 86 codownregualted genes in both cell lines after FAQSE treatment. The enriched GO terms of co-upregulated DEGs were primarily involved cell-cell adhesion, viral process, transcription initiation from RNA polymerase II promoter, positive regulation of transcription from RNA polymerase II promoter and actin cytoskeleton organization. The GO terms of co-downregulated DEGs were mainly enriched in the processes of SRP-dependent cotranslational protein targeting to membrane, viral transcription, nuclear-transcribed m RNA catabolic process, nonsense-mediated decay, translational initiation and r RNA processing. Both the cluster chemistry of tin and lanthanides have attracted extensive research interest,showing wide applications in catalysis,magnetism,luminescence,and lithography.However,their fusion into heterometallic Sn-Ln oxo clusters is still to be explored.In this study,through the stabilization of alkenyl-type cis-5-norbornene-endo-2,3-dicarboxylic acid(H_(2)NE)ligands,a series of atomically precise Sn-Ln oxo nanoclusters have been successfully constructed from the assembly of heterometallic tetranuclear Sn_(x)Ln((4-x))building blocks.Thereinto,Sn_(12)Eu_(8) and Sn_(13)Er_(6) with the highest nuclearities are built from multiple assembly of 8{Sn_(2)Eu_(2)}units and 6{Sn_(3)Er}and{Sn_(2)Er_(2)}units,respectively.ESI-MS analysis indicates that Sn_(13)Er_(6) has high solution stability,allowing their packing into thin films for lithography applications.As a result of electron beam lithography(EBL)studies,the condensation of Sn_(13)Er_(6) can be triggered by low energy radiation of 10μC/cm^(2),and 50 nm lines have been fabricated at expose energy of 50μC/cm^(2),confirming the satisfying sensitivity and resolution of Sn_(13)Er_(6).Hence,the success of this study develops the chemistry of heterometallic tin-lanthanide clusters that can be applied as novel negative photoresist materials.

Decoding six basic emotions from brain functional connectivity patterns

Although distinctive neural and physiological states are suggested to underlie the six basic emotions,basic emotions are often indistinguishable from functional magnetic resonance imaging(f MRI)voxelwise activation(VA)patterns.Here,we hypothesize that functional connectivity(FC)patterns across brain regions may contain emotion-representation information beyond VA patterns.We collected whole-brain f MRI data while human participants viewed pictures of faces expressing one of the six basic emotions(i.e.,anger,disgust,fear,happiness,sadness,and surprise)or showing neutral expressions.We obtained FC patterns for each emotion across brain regions over the whole brain and applied multivariate pattern decoding to decode emotions in the FC pattern representation space.Our results showed that the whole-brain FC patterns successfully classified not only the six basic emotions from neutral expressions but also each basic emotion from other emotions.An emotion-representation network for each basic emotion that spanned beyond the classical brain regions for emotion processing was identified.Finally,we demonstrated that within the same brain regions,FC-based decoding consistently performed better than VA-based decoding.Taken together,our findings revealed that FC patterns contained emotional information and advocated for paying further attention to the contribution of FCs to emotion processing.

Nonlinear free vibration of piezoelectric semiconductor doubly-curved shells based on nonlinear drift-diffusion model

In this paper, the nonlinear free vibration behaviors of the piezoelectric semiconductor(PS) doubly-curved shell resting on the Pasternak foundation are studied within the framework of the nonlinear drift-diffusion(NLDD) model and the first-order shear deformation theory. The nonlinear constitutive relations are presented, and the strain energy, kinetic energy, and virtual work of the PS doubly-curved shell are derived.Based on Hamilton's principle as well as the condition of charge continuity, the nonlinear governing equations are achieved, and then these equations are solved by means of an efficient iteration method. Several numerical examples are given to show the effect of the nonlinear drift current, elastic foundation parameters as well as geometric parameters on the nonlinear vibration frequency, and the damping characteristic of the PS doublycurved shell. The main innovations of the manuscript are that the difference between the linearized drift-diffusion(LDD) model and the NLDD model is revealed, and an effective method is proposed to select a proper initial electron concentration for the LDD model.

Current research on circular RNAs and their potential clinical implications in breast cancer

Breast cancer(BC)is one of the most common cancers and the leading causes of death among women worldwide,and its morbidity rate is growing.Discovery of novel biomarkers is necessary for early BC detection,treatment,and prognostication.Circular RNAs(circ RNAs),a novel type of endogenous non-coding RNAs with covalently closed continuous loops,have been found to have a crucial role in tumorigenesis.Studies have demonstrated that circ RNAs are aberrantly expressed in the tumor tissues and plasma of patients with BC,and they modulate gene expression affecting the proliferation,metastasis,and chemoresistance of BC by specifically binding and regulating the expression of micro RNAs(mi RNAs).Therefore,circ RNAs can be used as novel potential diagnostic and prognostic markers,and therapeutic targets for BC.This article summarizes the properties,functions,and regulatory mechanisms of circ RNAs,particularly current research on their association with BC proliferation,metastasis,and chemoresistance.

Modeling the arc and ring structures in the HD 143006 disk

Rings and asymmetries in protoplanetary disks are considered as signposts of ongoing planet formation.In this work,we con-duct three-dimensional radiative transfer simulations to model the intriguing disk around HD143006 that has three dust rings and a bright arc.A complex geometric configuration,with a misaligned inner disk,is assumed to account for the asymmetric structures.The two-dimensional surface density is constructed by iteratively fitting the ALMA data.We find that the dust tem-perature displays a notable discontinuity at the boundary of the misalignment.The ring masses range from 0.6 to 16 M⊕that are systematically lower than those inferred in the younger HL Tau disk.The arc occupies nearly 20%of the total dust mass.Such a high mass fraction of dust grains concentrated in a local region is consistent with the mechanism of dust trapping into vortices.Assuming a gas-to-dust mass ratio of 30 that is constant throughout the disk,the dense and cold arc is close to the threshold of being gravitationally unstable,with the Toomre parameter Q∼1.3.Nevertheless,our estimate of Q relies on the assumption for the unknown gas-to-dust mass ratio.Adopting a lower gas-to-dust mass ratio would increase the inferred Q value.Follow-up high resolution observations of dust and gas lines are needed to clarify the origin of the substructures.

Equilibria and Stability Analysis of Cohen-Grossberg BAM Neural Networks on Time Scale

This paper considers the Cohen-Grossberg BAM neural networks(CG-BAMNNs) on time scale, which can unify and generalize the continuous and discrete systems. First, the criteria for the existence and uniqueness of the equilibrium of CG-BAMNNs are derived on time scale. Then based on that, the authors give the criteria for the stability and estimation of equilibrium of the CG-BAMNNs on time scale. The method proposed in this paper unifies and generalizes the continuous and discrete CGBAMNNs systems, and is applicable to some other neural network systems on time scale with practical meaning. The effectiveness of the proposed criteria for delayed CG-BAMNNs is demonstrated by numerical simulation.

An air-rail inter-modal strategy for aircraft recovery

It is common that airlines encounter a disruption of a flight schedule,which is mainly caused by resource shortages.In case of a disruption,subject to scarce resources,most of airlines lose flexibility of performing aircraft recovery on the basis of business interest priorities and need to delay,swap,and cancel flights.This paper proposes an air-rail inter-modal strategy to incorporate a High-Speed Rail(HSR)transport mode into an aviation network for aircraft recovery.The air-rail inter-modal strategy focuses on occasionally operational integration of existing airside and HSR infrastructure capacities.It is different from air-rail cooperation implemented in Europe which emphasizes a long-term strategy.In addition to modelling the air-rail inter-modal strategy,an inter-modal time-band network is presented.Modelling is applied to a pure aviation network and the inter-modal network.Comparison results show that the inter-modal air-rail strategy helps to reduce the number of cancelled flights and the total disruption cost.

Secure control for discrete-time hidden Markov jump systems subject to replay attacks via output feedback

This paper investigates the static output feedback secure control problem for discrete-time hidden Markov jump systems against replay attacks. The main purpose is to realise that closed-loopsystems are stochastically stable with or without replay attacks. Firstly, the tampered sensorsunder replay attacks can be identified via the proposed detection method. Then, an asynchronousstatic output feedback controller is designed, which can eliminate the negative impactcaused by replay attacks in view of the detection results. Based on the linear matrix inequalitytechnique, some sufficient conditions which ensure the closed-loop systems are stochasticallystable and meet a given H∞ performance are established. Finally, a numerical example and apractical example are given to verify the effectiveness and superiority of the proposed method.

Modeling and Stability Issues of Voltage-source Converter-dominated Power Systems:A Review

With the substantive increase in the proportion of voltage-source converter(VSC)-based equipment,traditional power systems that primarily constituted of synchronous generators(SGs)gradually evolved into VSC-dominated ones.At the same time,there is an urgent need for modeling and stability assessment of such systems,since low inertia and weak damping features impair the ability of the systems to resist random disturbances.Existing works model the system dynamic processes from various domains(i.e.,time,frequency and energy),and analyze/determine the system stability under small or large disturbances.Among them,small-signal stability assessments mainly adopt the time-domain analysis based on the state-space model while frequency-domain methods include the impedance model,phase-amplitude dynamics model,and static synchronous generator model.Large-signal stability assessments mainly exploit the time-domain simulation with detailed models(i.e.,continuous/discrete-time mixed model with differentialdifference-algebraic equations),and the energy-domain analysis is based on energy function models.This paper presents a comprehensive review of existing modeling and stability analysis methods for VSC-dominated power systems,including their basic principles,key features,application scenarios and development tendencies.Key technical issues related to modeling and stability analysis are also summarized.
