Glycerophospholipids pathways and chromosomal instability in gastric cancer: Global lipidomics analysis

BACKGROUND Based on the breakthrough of genomics analysis, The Cancer Genome Atlas Research Group recently proposed an integrative genomic analysis, dividing gastric cancer(GC) into four subtypes, characterized by the chromosomal instability(CIN) status. However, the CIN status of GC is still vaguely characterized and lacking the valuable easy-to-use CIN markers to diagnosis in molecular and histological *** To explore the associations of CIN with downstream lipidomics *** We collected cancerous and noncancerous tissue samples from 18 patients with GC; the samples were divided into CIN and non-CIN types based on the system of The Cancer Genome Atlas Research Group and 409 sequenced oncogenes and tumor suppressor genes. We identified the lipidomics profiles of the GC samples and samples of their adjacent noncancerous tissues by using liquid chromatography–mass spectrometry. Furthermore, we selected leading metabolites based on variable importance in projection scores of > 1.0 and P <*** Twelve men and six women participated in this study; the participants had a median age of 67.5 years(range, 52–87 years) and were divided into CIN(n = 9)and non-CIN(n = 9) groups. The GC samples exhibited distinct profiles of lysophosphocholine, phosphocholine, phosphatidylethanolamine,phosphatidylinositol, phosphoserine, sphingomyelin, ceramide, and triglycerides compared with their adjacent noncancerous tissues. The glycerophospholipid levels(phosphocholine, phosphatidylethanolamine, and phosphatidylinositol)were 1.4-to 2.3-times higher in the CIN group compared with the non-CIN group(P < 0.05). Alterations in the glycerolipid and glycerophospholipid pathways indicated progression of GC toward *** The lipidomics profiles of GC samples were distinct from those of their adjacent noncancerous tissues. Microbial diversity of soil bacteria in agricultural field contaminated with heavy metals

In this study we evaluated the bacterial diversity in a soil sample from a site next to a chemical industrial factory previously contaminated with heavy metals. Analysis of 16S rDNA sequences amplified from DNA directly extracted from the soil revealed 17 different bacterial types （genera and/or species）. They included Polyangium spp., Sphingomonas spp., Variovorax spp., Hafina spp., Clostridia, Acidobacteria, the enterics and some uncultured strains. Microbes able to tolerate high concentrations of cadmium （500μmol/L and above） were also isolated from the soil. These isolates included strains of Acinetobacter （strain CD06）, Enterobacter sp. （strains CD01, CD03, CD04 and CD08）（similar strains also identified in culture-independent approach） and a strain of Stenotrophomonas sp. The results indicated that the species identified from direct analysis of 16S rDNA of the soil can be quite different from those strains obtained from enrichment cultures and the microbial activities for heavy metal resistance might be more appropriately addressed by the actual isolates.

Biomimetic flexible plate actuators are faster and more efficient with a passive attachment

Using three-dimensional computer simulations, we probe biomimetic free swimming of an internally actuated flexible plate in the regime near the first natural frequency. The plate is driven by an oscillating internal moment approximating the actuation mechanism of a piezoelectric macro fiber composite (MFC) bimorph. We show in our simulations that the addition of a passive attachment increases both swimming velocity and efficiency. Specifically, if the active and passive sections are of similar size, the overall performance is the best. We determine that this optimum is a result of two competing factors. If the passive section is too large, then the actuated portion is unable to generate substantial deflection to create sufficient thrust. On the other hand, a large actuated section leads to a bending pattern that is inefficient at generating thrust especially at higher frequencies.

Porous Cube-like In_2O_3 Nanoparticles and Their Sensing Characteristics toward Ethanol

Porous cube-like crystalline In2O3 nanoparticles with an average diagonal length of 34.8 nm were fabricated by a laser ablation-reflux process to form In（OH）3, followed by a calcination treatment to yield porous In2O3. HRTEM （high-resolution transmission electronic microscopy）, XRD （X-ray diffraction）, BET （Brunauer-Emmett-Teller）, and XPS （X-ray photoelectron spectroscopy） analysis were used to characterize their crystalline structures, grain sizes, surface areas, and surface compositions. The as-prepared porous in-dium oxides were tested for their sensing properties toward ethanol. Non-porous In2O3 nanopowder （about 40 nm） was also examined in order to compare the results with the as-prepared porous In2O3 nanomaterials. The porous In2O3 exhibited much better performance than that of non-porosus In2O3, and showed enhanced sensitivity with a lower detection limit than other reported In2O3-based materials when exposed to ethanol. Good gas sensitivity and linear behavior as a function of ethanol concentration were observed in the porous In2O3 nanoparticles.

Prognostic analysis of surgical treatment of peripheral cholangiocarcinoma: Two decades of experience at Chang Gung Memorial Hospital

AIM: To analyze the prognostic factors influencing the overall survival of peripheral cholangiocarcinoma (PCC) patients undergoing surgical treatment during 25 years at a single institution. METHODS: This study retrospectively reviewed prospectively collecting data about 373 patients with historically proven PCC who underwent surgical treatment between 1977 and 2001. RESULTS: Three hundred and seventy-three PCC patients (159 men and 214 women) underwent surgical treatment from 1977 to 2001. Among them, 187 PCC patients underwent hepatectomy and 135 had curative resection (curative resectability rate: 36.2%). The follow-up duration ranged from 1.05 to 167.6 mo (mean/median = 14.1/7.2 mo). Overall cumulative survival rates at 1, 3, and 5 years were 32.5%, 9.2%, and 4.1%, respectively. Univariate log-rank analysis identified the following as adverse influences on overall survival: presence of symptoms, absence of mucobilia, elevated CEA and CA 19-9 levels, non-papillary tumor type, receiving non-hepatectomy, advanced tumor staging, lack of post-operative chemotherapy, and radiotherapy. Meanwhile, multivariate Cox's proportional hazard analysis demonstrated that absence of mucobilia, non-papillary tumor type, advanced tumor staging, non-hepatectomy, and lack of post-operative chemotherapy were the five independent prognostic factors that adversely affected overall survival. CONCLUSION: Favorable overall survival of PCC patients undergoing surgical treatment depends on early tumor stage, presence of mucobilia, papillary tumor type, hepatic resection, and post-operative chemotherapy.

Comparison of murine cirrhosis models induced by hepatotoxin administration and common bile duct ligation

AIM： To build up the research models of hepatic fibrosis in ***： Inbred wild-type FVB/N mice were either treated with alpha-naphthyl-isothiocyanate （ANIT）, allyl alcohol （AA）,carbon tetrachloride （CCl4）, 3,5-diethoxycarbonyl-l,4-dihydrocollidine （DDC）, and silica, or subjected to common bile duct ligation （CBDL） to induce hepatic injury. Liver biopsies were performed every 4 wk to evaluate hepatic fibrosis over a period of 6 mo. Cumulative cirrhosis and survival curves were constructed by life table method and compared with Wilcoxon ***： Under the dosages used, there was neither mortality nor cirrhosis in AA and silica-treated groups. DDC and ANIT caused cirrhosis within 4-12 and 12-24 wk, *** showed significantly faster cirrhosis induction at high dosages without significant alteration of survival. The duration for cirrhosis induction by CCl4 ranged from 4 to 20 wk, mainly dependent upon the dosage. However, the increase in CCl4 dosage significantly worsened survival. Intraperitoneal CCl4 administration resulted in better survival in comparison with garage administration at high dosage, but not at medium and low dosages. After CBDL, all the mice developed liver cirrhosis within 4-8 wk and then died by the end of 26 ***： CBDL and administrations of ANIT, CCl4, and DDC ensured liver cirrhosis. CBDL required the least amount of time in cirrhosis induction, but caused shortened lives of mice. It was followed by DDC and ANIT administration with favorable survival. As for CCl4, the speed of cirrhosis induction and the mouse survival depended upon the dosages and the administration route.

Sunitinib for Taiwan Residents patients with gastrointestinal stromal tumor after imatinib treatment failure or intolerance

AIM:To report preliminary results of the efficacy and safety of sunitinib in the management of Taiwan Residents gastrointestinal stromal tumors (GIST) patients facing imatinib mesylate (IM) intolerance or ***:Between 2001 and May 2010,199 Taiwan Residents patients with metastatic GIST were treated at Chang Gung Memorial *** them,23 (11.6%) patients receiving sunitinib were ***:Sixteen male and 7 female patients with a median age of 59 years (range:24-83 years) received ***-two GIST patients changed to sunitinib because of IM failure and 1 because of *** median duration of sunitinib administration was 6.0 mo (range:2-29 mo).The clinical benefit was 65.2% [2 complete response (CR),4 partial response (PR),and 9 stationary disease (SD);15/23].In 12 patients harboring mutations of the kit gene at exon 11,the clinical benefit rate (CR,PR,and SD) was 75.0% and 6 patients with tumors containing kit exon 9 mutations had a clinical benefit of 50.0% (not significant,P=0.344).The progression free survival (PFS) and overall survival (OS) did not differ between patients whose GISTs had wild type,KIT exon 9,or KIT exon 11 ***-foot syndrome was the most common cause of grade Ⅲ adverse effect (26.1%),followed by anemia (17.4%),and neutropenia (13.0%).During the median 7.5-mo follow-up after sunitinib use,the median PFS and OS of these 23 GIST patients after sunitinib treatment were 8.4 and 14.1 mo,***:Sunitinib appears to be an effective treatment for Taiwan Residents with IM-resistant/intolerant GISTs and induced a sustained clinical benefit in more than 50% of Taiwan Residents advanced GIST patients.

APPLYING PROCESS MINING APPROACH TO SUPPORT THE VERIFICATION OF A MULTI-AGENT SYSTEM

Using agent development tools to construct an agent-based system is a well applied approach. However, the development tools usually do not have the function to check the feasibility about the workflow of the agent system during it implementation stage. Therefore, to develop an evaluation approach to analyze the feasibility of a developing agent system such that the improper workflow of an agent system can be found in the early design stage is a necessary task to reduce the risk of implementation. In this research, a Petri Net （PN） based three-stage evaluation approach was developed. In the conceptual stage, the pitfall of the current agent system developing process was examined and an improvement analysis process was specified. Then, in the system design stage, an evaluation approach which extracted the process log file from a developing agent system into a PN model in terms of a process mining approach-or algorithm was proposed. This model was simulated in a PN simulation package. The agent system performance was evaluated in terms of analyzing the deadlock phenomena of the PN model. Finally, in the implementation stage, the proposed concept was implemented by using an agent developing tool JADE and a PN simulation tool CPN. An agent-based robotic assembly system was used to examine the possible deadlock of the agent system.

Uniqueness, scale, and resolution issues in groundwater model parameter identification

This paper first visits uniqueness, scale, and resolution issues in groundwater flow forward modeling problems. It then makes the point that non-unique solutions to groundwater flow inverse problems arise from a lack of information necessary to make the problems well defined. Subsequently, it presents the necessary conditions for a well-defined inverse problem. They are full specifications of （1） flux boundaries and sources/sinks, and （2） heads everywhere in the domain at at least three times （one of which is t = 0）, with head change everywhere at those times being nonzero for transient flow. Numerical experiments are presented to corroborate the fact that, once the necessary conditions are met, the inverse problem has a unique solution. We also demonstrate that measurement noise, instability, and sensitivity are issues related to solution techniques rather than the inverse problems themselves. In addition, we show that a mathematically well-defined inverse problem, based on an equivalent homogeneous or a layered conceptual model, may yield physically incorrect and scenario-dependent parameter values. These issues are attributed to inconsistency between the scale of the head observed and that implied by these models. Such issues can be reduced only if a sufficiently large number of observation wells are used in the equivalent homogeneous domain or each layer. With a large number of wells, we then show that increase in parameterization can lead to a higher-resolution depiction of heterogeneity if an appropriate inverse methodology is used. Furthermore, we illustrate that, using the same number of wells, a highly parameterized model in conjunction with hydraulic tomography can yield better characterization of the aquifer and minimize the scale and scenario-dependent problems.
