Oncogenes are critical factors in tumorigenesis of diverse cancer types and play essential roles in tumor immune escape.Mutations in Kirsten rat sarcoma viral oncogene homolog(KRAS)and epidermal growth factor receptor(EGFR)are among the most frequent gain-of-function alterations[1].After many years of in-depth research,inhibitors targeting EGFR or KRAS mutations have been successfully developed,however,their clinical benefit is relatively limited,and they will inevitably encounter the challenge of drug resistance.The emergence of resistance is attributed to secondary mutations in driver genes and other complicated factors.

Hot-swaging yields a high ultimate tensile strength of 712 MPa but a limited tensile ductility with the total elongation of3.6%at a testing temperature of 200℃in a representative W-0.5wt.%ZrC alloy.In this work,the evolution of Vickers microhardness with annealing temperatures is investigated in detail,which contributes to a rough index chart to guide the search for an optimized post-annealing temperature.Through the post-annealing around 1300℃,an outstanding tensile ductility at200℃,including a uniform elongation of 14%and a total elongation of~25%,has been achieved without the sacrifice of its strength.The evolution of dislocations and grain structures with the annealing temperatures accessed through backscattered scanning electron microscope and transmission electron microscope analysis reveals that the improved low-temperature tensile ductility has resulted from the reduction of residual dislocations and dislocation tanglement via the static recovery,which provides more room to accommodate dislocations,and hence stronger strain hardening ability and tensile ductility.

The cermet fuels have been considered as a potential key component for the nuclear thermal propulsion,and the homogeneity of the fuel particles in the metal matrix plays a crucial role in stabilizing the structure at extremely high temperatures.In this work,liquid paraffin was used as additive to improve the distribution of yttria-stabilized zirconia(YSZ,an appropriate surrogate for UO_(2) fuel)microspheres in the tungsten(W)matrix,and the W-YSZ cermet wafers(volume ratio 1:1)with a relative density of 97.6%were fabricated by spark plasma sintering with a specifically designed program.The effects of the paraffin dosage(0-5 wt.%)on the homogeneity,microstructure,mechanical properties,and the thermal conductivity of W-YSZ cermet were investigated.The W-YSZ sample with 2 wt.%paraffin shows the highest homogeneity and exhibits the best comprehensive properties,including the ultimate tensile strength of 132.2 MPa at 600℃,the bending strength of455 MPa and thermal conductivity of 50 W·m^(-1)·K^(-1)at room temperature.Moreover,the cermet could keep structurally sound after thermal shocked at a heat load of 20 MW·m^(-2).These results would be helpful for the design and optimization of the cermet fuels in the nuclear thermal propulsion.

BACKGROUND The treatment of acute respiratory distress syndrome(ARDS)complicated by sepsis syndrome(SS)remains challenging.AIM To investigate whether combined adipose-derived mesenchymal-stem-cells(ADMSCs)-derived exosome(EXAD)and exogenous mitochondria(mitoEx)protect the lung from ARDS complicated by SS.METHODS In vitro study,including L2 cells treated with lipopolysaccharide(LPS)and in vivo study including male-adult-SD rats categorized into groups 1(sham-operated-control),2(ARDS-SS),3(ARDS-SS+EXAD),4(ARDS-SS+mitoEx),and 5(ARDS-SS+EXAD+mitoEx),were included in the present study.RESULTS In vitro study showed an abundance of mitoEx found in recipient-L2 cells,resulting in significantly higher mitochondrial-cytochrome-C,adenosine triphosphate and relative mitochondrial DNA levels(P<0.001).The protein levels of inflammation[interleukin(IL)-1β/tumor necrosis factor(TNF)-α/nuclear factor-κB/toll-like receptor(TLR)-4/matrix-metalloproteinase(MMP)-9/oxidative-stress(NOX-1/NOX-2)/apoptosis(cleaved-caspase3/cleaved-poly(ADP-ribose)polymerase)]were significantly attenuated in lipopolysaccharide(LPS)-treated L2 cells with EXAD treatment than without EXAD treatment,whereas the protein expressions of cellular junctions[occluding/β-catenin/zonula occludens(ZO)-1/E-cadherin]exhibited an opposite pattern of inflam-mation(all P<0.001).Animals were euthanized by 72 h post-48 h-ARDS induction,and lung tissues were harvested.By 72 h,flow cytometric analysis of bronchoalveolar lavage fluid demonstrated that the levels of inflam-matory cells(Ly6G+/CD14+/CD68+/CD11b/c+/myeloperoxidase+)and albumin were lowest in group 1,highest in group 2,and significantly higher in groups 3 and 4 than in group 5(all P<0.0001),whereas arterial oxygen-saturation(SaO2%)displayed an opposite pattern of albumin among the groups.Histopathological findings of lung injury/fibrosis area and inflammatory/DNA-damaged markers(CD68+/γ-H2AX)displayed an identical pattern of SaO2%among the groups(all P<0.0001).The protein expressions of inflammatory(TLR-4/MMP-9/IL-

Objective: Optimal luteinizing hormone (LH) levels forin vitro fertilization/intracytoplasmic sperm injection (IVF/ICSI) patients following the gonadotropin-releasing hormone (GnRH) antagonist protocol remain controversial. This study aimed to estimate the association between serum LH levels on the triggering day (LHLOT) and clinical pregnancy outcomes in IVF/ICSI patients following the GnRH antagonist protocol.Methods: We enrolled 4743 IVF/ICSI cycles using flexible or fixed GnRH antagonist protocols between January 2017 and June 2021. The patients were stratified into four groups according to the LHLOT quartiles (Q1–Q4). Each group was further divided into two subgroups according to the median basal LH/LHLOT. The main outcomes were clinical pregnancy, birth, and miscarriage rates. Secondary outcomes were oocyte yield, number of MII, fertilized oocytes (2PN), and high-quality embryos.Results: Serum LHLOT levels were negatively correlated with oocyte yield and the numbers of MII, 2PN, and high-quality embryos. Reproductive outcomes were compared among the different quartiles and subgroups. The absolute difference in the clinical pregnancy rates between the Q4 and Q1 groups was 6.4% (47.2%vs. 40.8%,P 3.04 and ≤3.04 were 32.7% and 48.6% (P 2.14 and ≤2.14 were 50.2% and 41.9%, respectively (P <0.05). The greater the decrease in LH levels, the lower the oocyte maturation rate across all quartiles.Conclusions: According to the LHLOT quartiles, the Q1 of serum LHLOT <1.43 IU/L may result in a higher number of oocytes but reduced oocyte maturation and lower clinical pregnancy rates. A decrease in LH levels by approximately 2.14 to 3.04 times compared to basal LH levels may lead to higher clinical pregnancy rates in IVF/ICSI patients following the GnRH antagonist protocol.

为满足航空发动机及机载产品研制过程贴近使用环境的振动考核试验需求,需根据发动机实测振动数据给出振动考核试验所需的输入谱图。依据GJB/Z 126-99中给出的环境测量数据归纳方法,建立了发动机实测振动环境谱统计归纳方法并通过程序实现。利用发动机多架次实测试飞振动数据统计归纳得到发动机测点位置的振动实测谱。基于能量等效及信号频域特征分布一致原则,将归纳得到的实测谱转化为可用于振动台输入的振动环境谱,并在振动台上进行了振动信号的复现试验。结果表明:振动台输出信号与发动机实测振动信号频域分布特征一致,在统计频率带宽范围内振动总量最大相差5.7%,证明了转化方法是合理的,为航空发动机机载设备贴近使用环境的振动考核试验方法提供了真实的输入谱图。

针对纺机钣金车间生产过程实时监控差、准确性不足及可视化程度低等问题,提出一种基于数字孪生的纺机钣金车间可视化监控系统构建方法。首先,基于纺机钣金车间的生产要素构建其孪生几何模型,并对孪生几何模型进行轻量化处理以满足实时响应要求;然后,基于赋时Petri网设计其行为逻辑模型,并通过碰撞检测算法实现在孪生车间中的触发响应。在此基础上,结合OPC UA的信息建模、事件驱动等方法实现多种工艺路线的产品加工、在制品流转等过程的忠实映射。最后,以某纺机钣金车间为应用案例,设计开发了原型系统,验证了所提方法的可行性和有效性。

甲状腺癌是内分泌系统最常见的恶性肿瘤,其中分化型甲状腺癌(differentiated thyroid carcinoma,DTC)占90%以上。多数DTC患者经过系统治疗后预后良好,但少数患者肿瘤原发灶或转移灶出现失分化现象,进展为放射性碘难治性DTC(radioiodine-refractory DTC,RAIR-DTC),预后明显变差,是甲状腺癌致死的主要原因。钠碘转运体(sodium iodide symporter,NIS)的表达和功能异常,是导致甲状腺癌碘-131治疗抵抗的主要原因,其发生受遗传学改变、表观遗传学改变、肿瘤微环境作用、自噬作用等多因素影响。遗传学改变如BRAF基因的V600E位点突变、RET/PTC基因重排等导致致癌信号通路的激活,直接或间接地影响NIS的表达及其在细胞膜上的正常定位。表观遗传学调控特定基因的表达模式,调节NIS的表达水平,进而影响甲状腺细胞的碘摄取功能。肿瘤微环境中的免疫细胞、细胞因子和细胞外基质等成分也可能通过降低NIS的表达水平和/或干扰其在细胞膜上的正常功能导致细胞碘摄取障碍。此外,自噬作为一种细胞内部的代谢调节机制,也可以调节NIS的表达及其在细胞内的分布,从而影响碘的摄取和碘-131治疗的敏感性。通过综述以上因素在甲状腺癌失分化中的作用机制,可以更全面地理解RAIR-DTC的发生和发展过程,有助于探寻新的治疗靶点,改善预后,并为患者提供更有效的个体化治疗策略。

当前,我国正在建设以金融信用为内核、公共信用为外延的农村信用体系,这对建设农业农村现代化、全面推进乡村振兴具有重要的战略意义。论文通过系统梳理2019-2023年安徽省4次联席会文件、16个地市的专项汇报材料,并于2023年9、10月深入7个首批选点区县进行田野调查,研究安徽省农村信用体系建设问题,运用扎根理论进行三级编码、构建理论模型、阐释理论机理。研究发现:安徽省农村信用体系具有显著的“嵌入与互动”特征,以多维嵌入促进多元协同的互动机制,推动金融循环、信用循环、产业循环、数据循环等产生齿轮效应,形成系统联动、循环继起的效能闭环。这就要求把握嵌入深度,构建个体循环畅通、外部联结有力的制度体系;把握协同程度,完善重点推进为牵引、系统联动为促进的运行机制;把握推进力度,发挥外源推动、内生驱动相得益彰的制度效能。

Levosimendan does not reduce the mortality of critically ill adult patients with sepsis and septic shock: a meta-analysis

Background: Previous studies on whether or not levosimendan improved the prognosis of patients with sepsis and septic shock been inconsistent. We aimed to provide an updated analysis of the therapeutic value of levosimendan in adult patients with sepsis and septic shock, in order to provide evidence-based medical evidence for its use. Methods: PubMed, Embase, Cochrane Library, Wanfang Data, and CNKI were searched until August 2018 without language restriction. Randomized controlled studies of levosimendan with either inotropic drugs or placebo for the treatment of sepsis or septic shock were enrolled. The primary outcome was mortality, and cardiac index and serum lactate levels were the secondary outcomes. Results: A total of 20 randomized controlled studies were included in this meta-analysis, including 1467 patients, with 738 patients in the experimental group (levosimendan group) and 729 patients in the control group (other inotropic drugs or placebo). There were no significant differences in mortality between the levosimendan and control groups (fixed-effect relative risk [RR]= 0.90, 95% confidence interval [CI][0.79, 1.03], P = 0.13). Levosimendan increased the cardiac index (VMD [weighted mean difference]= 0.51, 95% CI [0.06, 0.95], P = 0.02);and serum lactate levels were lower (VMD =—1.04, 95% CI [—1.47,—0.60], P< 0.00001). Conclusions: Based on current clinical evidence, levosimendan does not reduce mortality in adult critically ill patients with sepsis and septic shock. Physicians should use levosimendan with caution in patients with sepsis and septic shock.
