Pharmacological targeting cGAS/STING/NF-κB axis by tryptanthrin induces microglia polarization toward M2 phenotype and promotes functional recovery in a mouse model of spinal cord injury

The M1/M2 phenotypic shift of microglia after spinal cord injury plays an important role in the regulation of neuroinflammation during the secondary injury phase of spinal cord *** of shifting microglia polarization from M1(neurotoxic and proinflammatory type)to M2(neuroprotective and anti-inflammatory type)after spinal cord injury appears to be *** possesses an anti-inflammatory biological ***,its roles and the underlying molecular mechanisms in spinal cord injury remain *** this study,we found that tryptanthrin inhibited microglia-derived inflammation by promoting polarization to the M2 phenotype in *** promoted M2 polarization through inactivating the cGAS/STING/NF-κB ***,we found that targeting the cGAS/STING/NF-κB pathway with tryptanthrin shifted microglia from the M1 to M2 phenotype after spinal cord injury,inhibited neuronal loss,and promoted tissue repair and functional recovery in a mouse model of spinal cord ***,using a conditional co-culture system,we found that microglia treated with tryptanthrin suppressed endoplasmic reticulum stress-related neuronal *** together,these results suggest that by targeting the cGAS/STING/NF-κB axis,tryptanthrin attenuates microglia-derived neuroinflammation and promotes functional recovery after spinal cord injury through shifting microglia polarization to the M2 phenotype.

Experimental Study on Geometry Characteristics of Turbulent Premixed Flames for Natural Gas/Air Mixtures

In this study,focusing on the geometry characteristics of spherical expanding flame,the turbulent premixed flames of natural gas/air mixtures were investigated in a fan-stirred turbulent *** effects of initial temperature（T=300-400 K）,initial pressure（P=0.1-0.3 MPa）,turbulence intensity（u'=1.0-2.7 m/s）,oxygen volumetric percentage(φ（O2）=15%-21%) and carbon dioxide volumetric percentage(φ（CO2）=0-20%)were delved *** flame profile under the Cartesian coordinate system was derived from the schlieren images taken by the high-speed ***,from both macroscopic and microscopic perspectives,the influence of experimental conditions on the flame geometry characteristics was explored through flame front extraction,wavelet decomposition and network *** results demonstrate that for significant flame wrinkling,changes in species concentrations and turbulence intensity have more pronounced effects on the flame wrinkling *** wrinkling of the flame front maintains a certain degree of similarity,as evidenced by the locally concentrated distribution of the angles of the maximum fluctuation *** disturbance energy under large-scale（D6-D8） disturbances exhibits relatively high values with a similar trend,exerting a significant impact on the geometry characteristics of the flame *** peaks of correlation degree are scattered either with the decomposition scale or the development of flame radius,indicating no linear correlation between different detail ***,the probability distribution of node degrees in key wrinkled regions exhibits different trends with that of large-scale wrinkling and disturbance energy,especially with changes in initial *** occurs because the number of key wrinkles varies based on the perturbation's strength or the region's ***,an increase in the fluctuation frequency of the flame's local radius may not necessarily lead to an increase in the number of key folded regions.

Qili Qiangxin, a compound herbal medicine formula, alleviates hypoxia-reoxygenation-induced apoptotic and autophagic cell death via suppression of ROS/AMPK/mTOR pathway in vitro

Objective: Qili Qiangxin(QLQX), a compound herbal medicine formula, is used effectively to treat congestive heart failure in China. However, the molecular mechanisms of the cardioprotective effect are still unclear. This study explores the cardioprotective effect and mechanism of QLQX using the hypoxiareoxygenation(H/R)-induced myocardial injury ***: The main chemical constituents of QLQX were analyzed using high-performance liquid chromatography-evaporative light-scattering detection. The model of H/R-induced myocardial injury in H9c2 cells was developed to simulate myocardial ischemia–reperfusion injury. Apoptosis, autophagy,and generation of reactive oxygen species(ROS) were measured to assess the protective effect of QLQX. Proteins related to autophagy, apoptosis and signalling pathways were detected using Western ***: Apoptosis, autophagy and the excessive production of ROS induced by H/R were significantly reduced after treating the H9c2 cells with QLQX. QLQX treatment at concentrations of 50 and 250 μg/mL caused significant reduction in the levels of LC3Ⅱ and p62 degradation(P < 0.05), and also suppressed the AMPK/mTOR signalling pathway. Furthermore, the AMPK inhibitor Compound C(at 0.5 μmol/L),and QLQX(250 μg/mL) significantly inhibited H/R-induced autophagy and apoptosis(P < 0.01), while AICAR(an AMPK activator, at 0.5 mmol/L) increased cardiomyocyte apoptosis and autophagy and abolished the anti-apoptotic effect of QLQX. Similar phenomena were also observed on the expressions of apoptotic and autophagic proteins, demonstrating that QLQX reduced the apoptosis and autophagy in the H/R-induced injury model via inhibiting the AMPK/mTOR pathway. Moreover, ROS scavenger,N-Acetyl-L-cysteine(NAC, at 2.5 mmol/L), significantly reduced H/R-triggered cell apoptosis and autophagy(P < 0.01). Path-Dependent Progressive Failure Analysis for 3D-Printed Continuous Carbon Fibre Reinforced Composites

In order to predict the damage behaviours of 3D-printed continuous carbon fibre(CCF)reinforced composites,when additional short carbon fibre(SCF)composite components are employed for continuous printing or special functionality,a novel path-dependent progressive failure(PDPF)numerical approach is ***,a progressive failure model using Hashin failure criteria with continuum damage mechanics to account for the damage initiation and evaluation of 3D-printed CCF reinforced polyamide(PA)composites is developed,based on actual fibre placement trajectories with physical measurements of 3D-printed CCF/PA ***,an elastic-plastic model is employed to predict the plastic damage behaviours of SCF/PA ***,the accuracy of the PDPF model was validated so as to study 3D-printed CCF/PA composites with either negative Poisson's ratio or high *** results demonstrate that the proposed PDPF model can achieve higher prediction accuracies in mechanical properties of these 3D-printed CCF/PA *** analyses show that the stress distribution is generally aggregated in the CCF areas along the fibre placement paths,and the shear damage and matrix tensile/compressive damage are the key damage *** study provides a new approach with valuable information for characterising complex 3D-printed continuous fibre-matrix composites with variable mechanical properties and multiple constituents.

A review of flotation and selective separation of pyrrhotite:A perspective from crystal structures

Pyrrhotite is an associated mineral that exists widely in sulfide ore. The presence of pyrrhotite will affect the recovery of platinum group minerals. Therefore, researchers have paid increasing attention to the flotation separation of pyrrhotite. Pyrrhotite superstructures owning different Fe/S ratios results in various crystal structures, corresponding to different physical, chemical and electronic properties, and consequently different flotation behavior. In the present paper, a comprehensive review is conducted to discuss the influence of crystal structures on the natural floatability, mineral-reagent interaction, surface oxidation and flotation electrochemistry of pyrrhotite. The selective flotation process of pyrrhotite from its associated minerals is also reviewed in this paper. It is hoped that this review can summarize the newly published research results combined with some representative results from the past, to provide a theoretical basis for the study of the flotation mechanism of pyrrhotite and provide a new direction for future research on pyrrhotite.

Cooper instability and superconductivity of the Penrose lattice

Bulk superconductivity(SC) has recently been observed in the Al-Zn-Mg quasicrystal(QC). To settle several fundamental issues of the SC on the QC, we use an attractive Hubbard model to perform a systematic study on the Penrose lattice. The first issue is the Cooper instability of the QC, i.e., no Fermi surface under an infinitesimal attractive interaction. Starting from the two-electron problem outside a filled Fermi sea, we analytically prove that an infinitesimal Hubbard attraction can lead to the Cooper instability as long as the density of the state is nonzero at the Fermi level. The findings provide a basis for the SC on the QC. Our numerical results show that the Cooper pairing always takes place between two time-reversal states, satisfying Anderson’s theorem. On this theorem, we perform a mean-field(MF) study at zero and finite temperatures. The MF study shows that an arbitrarily weak attraction can lead to a pairing order, with the resulting pairing state being well described by the Bardeen-Cooper-Schrieffer theory and the thermal dynamic behaviors being well consistent with the experimental results. The second issue is about the superfluid density on the QC without translational symmetry. Our findings clarify that although the normal state of the system locates at the critical point of the metal-insulator transition, the pairing state exhibits a real SC, carrying finite superfluid density that can be verified by the Meissner effect. This finding is consistent with the experiment results. This study also reveals that the properties of the SC on the Penrose lattice are universal for all QCs.

Hydrogenation of Cinnamaldehyde to Cinnamyl Alcohol over An Ultrafine Co-B Amorphous Catalyst

A novel Co-B amorphous alloy catalyst in the form of ultrafine particles was prepared by chemical reduction of CoCl2 with aqueous NaBH4, which exhibited excellent activity and selectivity during the hydrogenation of cinnamaldehyde to cinnamyl alcohol in liquid phase. The optimum yield of cinnamyl alcohol was 87.6%, much better than the yield of using Raney Ni, Raney Co and other Co-based catalysts.

Relationships of ozone formation sensitivity with precursors emissions,meteorology and land use types,in Guangdong-Hong Kong-Macao Greater Bay Area,China

Due to the influences of precursors emissions,meteorology,geography and other factors,ozone formation sensitivity(OFS)is generally spatially and temporally *** study characterized detailed spatial and temporal variations of OFS in Guangdong-Hong Kong-Macao Greater Bay Area(GBA)from 2012 to 2016 based on OMI satellite data,and analyzed the relationships of OFS with precursors emissions,meteorology and land use types(LUTs).From 2012 to 2016,the OFS tended to be NOx-limited in GBA,with the value of FNR(HCHO/NO 2)increasing from 2.04 to *** to the total annual emission statistics of precursors,NOx emissions decreased by 33.1%and VOCs emissions increased by 35.2%from 2012 to 2016,directly resulting in OFS tending to be NO *** Grey Relation Analysis results show that total column water(TCW),surface net solar radiation(SSR),air temperature at 2 m(T2)and surface pressure(SP)are the top four meteorological factors with the greatest influences on *** are significant positive correlations between FNR and T2,SSR,TCW,and significant negative correlations between FNR and *** GBA,the OFS tends to be NO x-limited regime in wet season(higher T2,SSR,TCW and lower SP)and VOCs-limited regime in dry season(lower T2,SSR,TCW and higher SP).The FNR displays obvious gradient variations on different LUTs,with the highest in“Rural areas”,second in“Suburban areas”and lowest in“Urban areas”.
