Adherence and invasion of mouse-adapted H pylori in different epithelial cell lines

AIM： To assess the adhesion and invasion abilities of different mouse adapted H py/or/strains in different cell lines in vitro and investigate their effects on the virulence factors cagA and vacA. METHODS： The adherence and invasion abilities of different N pylori strains in different epithelial cell lines were examined by the gentamycin protection assay. The null mutants of cagA and vacA were processed by direct PCR mutation method. The morphologic changes of different cell lines after N pylori attachment were examined by microscopy. RESULTS： The densities of adherence to and invasion into cells in vitro were different from those in the mouse infection experiments. 88-3887 strain could invade and adhere to cells stronger than SS1 and X47. All tested strains had better adhering and invasive abilities in SCG-7901 cell. CagA and vacA minus mutants had the same invasion and adherent abilities as their wild types. In all strains and cell lines tested, only AGS cell had the significant hummingbird phenotype after inoculation with the 88-3887 wild-type. CONCLUSION： Both the host cells and the bacteria play important parts in the invasion and adhesion abilities of Hpylori. CagA and VacA are not related to the ability of invasion and adhesion of Hpylori in different cell lines in vitro.

AIM： To investigate the relationship between urinary peptide changes and Helicobacter pylori （H. pylorl） infection using urinary peptidome profiling. METHODS： The study was performed in volunteers （n = 137） who gave informed consent. Urinary peptides were enriched by magnetic beads based weak cation exchange chromatography＇and spectrums acquired by matrix-assisted laser desorption/ionization time-of-flight （MALDI-TOF） mass spectrometry （MS）. ClinProTools bioinformatics software was used for statistical analysis and the recognition of peptide patterns. The marker peptides were identified by LTQ Obitrap XL tandem MS. RESULTS： Approximately 50 proteins or peptides which loaded onto the magnetic beads were detected by MAL-DI-TOF MS. By optimizing the parameters of the model, the Genetic Algorithm model had good recognition capability （97%） and positive predictive value （94%）. Based on the model, 2 markers with molecular masses of 6788 and 1912 Da were found that differentiated between H. pylori positive and negative volunteers. The m/z 1912 sequence was parsed as SKQFTSSTSYN- RGDSTF. The peptide was identified as isoform 1 of the fibrinogen a chain precursor, whose concentration in urine was markedly higher in H. pylori infected volunteers than in H. pylori non-infected ones. CONCLUSION： The appearance of urinary fibrinogen degradation products is caused by an active H. pyloriinduced process.

Combined en bloc liver/pancreas transplantation （CLPT） was used primarily in the treal^nent of otherwise non- resectable upper abdominal malignancy. In fact, a more appropriate indication is in patients with liver disease and insulin-dependent diabetes mellitus （IDDM）. Here, we report on two successful cases of CLPT at our hospital. One was a patient with non-resectable advanced liver cancer. The recipient survived for 23 mo and finally died of recurrent tumor. The other was a patient with severe biliary complication after orthotopic liver transplantation and preoperative IDDM. We performed CLPT with a modified surgical technique of preserving the native pancreas. He is currently liver-disease- and insulin-free more than 27 mo post-transplant. Based on our experience in two cases of abdominal cluster transplantation, we describe the technical details of CLPT and a modification of the surgical procedure.

The mechanical behavior of cemented gangue backfill materials(CGBMs)is closely related to particle size distribution(PSD)of aggregates and properties of cementitious ***,the true triaxial compression tests,CT scanning,SEM,and EDS tests were conducted on cemented gangue backfill samples(CGBSs)with various carbon nanotube concentrations(P_(CNT))that satisfied fractal theory for the PSD of *** mechanical properties,energy dissipations,and failure mechanisms of the CGBSs under true triaxial compression were systematically *** results indicate that appropriate carbon nanotubes(CNTs)effectively enhance the mechanical properties and energy dissipations of CGBSs through micropore filling and microcrack bridging,and the optimal effect appears at P_(CNT)of 0.08wt%.Taking PSD fractal dimension(D)of 2.500 as an example,compared to that of CGBS without CNT,the peak strength(σ_(p)),axial peak strain(ε_(1,p)),elastic strain energy(Ue),and dissipated energy(U_(d))increased by 12.76%,29.60%,19.05%,and90.39%,***,excessive CNTs can reduce the mechanical properties of CGBSs due to CNT agglomeration,manifesting a decrease inρ_(p),ε_(1,p),and the volumetric strain increment(Δε_(v))when P_(CNT)increases from 0.08wt%to 0.12wt%.Moreover,the addition of CNTs improved the integrity of CGBS after macroscopic failure,and crack extension in CGBSs appeared in two modes:detour and pass through the ***σ_(p)and U_(d)firstly increase and then decrease with increasing D,and porosity shows the opposite ***ε_(1,p)andΔε_(v)are negatively correlated with D,and CGBS with D=2.150 has the maximum deformation parameters(ε_(1,p)=0.05079,Δε_(v)=0.01990)due to the frictional slip effect caused by coarse *** increasing D,the failure modes of CGBSs are sequentially manifested as oblique shear failure,"Y-shaped"shear failure,and conjugate shear failure.

The microstructure evolution and mechanical properties of a Fe-0.12C-0.2Si-1.6Mn-0.3Cr-0.0025B(wt.%)steel with different initial microstructures,i.e.,hot rolled(HR)and cold rolled-annealed(CRA),were studied through optical microscopy,scanning electron microscopy,electron channeling contrast imaging,microhardness and room temperature uniaxial tensile *** water quenching from 930℃ to room temperature,a fully martensitic microstructure was obtained in both as-quenched HR and CRA specimens,which shows a microhardness of 480±5 HV,and no significant difference in microstructure and microhardness was *** test results show that the product of tensile strength and total elongation(UTS×TE)of the as-quenched HR specimen,i.e.,24.1 GPa%,is higher than that of the as-quenched CRA specimen,i.e.,18.9 GPa%.While,after being tempered at 300℃,the martensitic microstructures and mechanical properties of the two as-quenched specimens change significantly due to the synergy role of the matrix phase softening and the precipitation *** the maximum UTS×TE,it is 18.9 GPa%obtained in the as-quenched CRA one,while that is 24.4 GPa%obtained in the HR specimen after tempered at 300℃ for 5 min.

After the global pandemic,DaaS(desktop as a service)has become the first choice of many companies’remote working *** the desktops are usually deployed in the public cloud when using DaaS,customers are more cost-sensitive which boosts the requirement of proactive power *** researches in this area focus on virtual desktop infrastructure(VDI)session logon behavior modeling,but for the remote desktop service host(RDSH)-shared desktop pools,logoff optimization is also *** systems place sessions by round-robin or in a pre-defined order without considering their logoff ***,these approaches usually suffer from the situation that few left sessions prevent RDSH servers from being powered-off which introduces cost *** this paper,we propose session placement via adaptive user logoff prediction(SODA),an innovative compound model towards proactive RDSH session ***,an ensemble machine learning model that can predict session logoff time is combined with a statistical session placement bucket model to place RDSH sessions with similar logoff time in a more centralized manner on RDSH ***,the infrastructure cost-saving can be improved by reducing the resource waste introduced by those RDSH hosts with very few hanging sessions left for a long *** on real RDSH pool data demonstrate the effectiveness of the proposed proactive session placement approach against existing static placement techniques.

Dear Editor,This letter proposes a deep synchronization control(DSC) method to synchronize grid-forming converters with power grids. The method involves constructing a novel controller for grid-forming converters based on the stable deep dynamics model. To enhance the performance of the controller, the dynamics model is optimized within the deep reinforcement learning(DRL) framework. Simulation results verify that the proposed method can reduce frequency deviation and improve active power responses.

杂交是物种进化动力的源泉之一,它有助于快速积累遗传变异,增强群体的遗传多样性,促进新物种的产生,因此在不同物种类群中识别古老和近期的杂交现象具有重要意义。杂交事件的识别方法可以采用多种特征,包括传统的遗传表型、代谢产物、染色体数目,以及群体遗传学和系统发育基因组学提供的基因水平的分子表型。本研究通过选取28个十字花科物种及两个外类群物种作为研究材料,采用比较基因组学和系统发育基因组学研究手段,在叶绿体和核基因两个方向上联合分子表型信息鉴定十字花科内的杂交事件。结果表明:1、以单基因树映射物种树的结果为依据,十字花科物种主要分为两类:其一,姐妹类群分布相对集中、以拟南芥(Arabidopsis thaliana)为代表;其二,姐妹类群分布相对分散暗示基因存在多种来源,以诚实花(Lunaria annua)为代表。2、进一步通过叶绿体和核基因的系统发育关系对比法分析,我们认为诚实花可能是一个新的杂交物种,其39.1%的基因与父本来源的C类群基因关系最近;38.5%的基因与C类群以外类群的基因最近,特别是来自B、C结合类群共同祖先的10号节点(占比3%)支持其作为诚实花的母本来源。3、通过对父母关键进化节点的功能富集分析,发现父本主要富集在生物降解、次级代谢产物、RNA调控等途径上,表明其可能通过RNA调控生物降解、次级代谢过程以适应胁迫环境;母本主要富集在代谢途径上,尤其是碳水化合物、核酸代谢等方面,表明其可能通过增强自身代谢获取相对竞争优势。
